Abstract

Translation of abstract (English)

L1 is a cell adhesion molecule which was originally found in the nervous system. Recent work has identified L1 additionally as a prognostic marker in several human carcinomas. L1 promotes tumor cell invasion and motility, augments tumor growth in nude mice and facilitates experimental tumor metastasis. As a tumor-associated antigen it is therefore a suitable target molecule for tumor therapy. Aim of this study was the preclinical investigation of a series of L1 antibodies against human ovarian carcinoma. Nine different murine monoclonal antibodies were produced and characterized for biochemical and functional means. The antibodies recognized different epitopes of L1 but did not react with the closely related protein CHL1. L1-38.12 specifically bound the neuronal form of L1 because it recognizes an epitope inside the exon 2 region. Several antibodies could react with the first Ig domain and were able to inhibit the L1 homophilic binding. Among these antibodies L1-9.3 and L1-OV198.5 were selected and analyzed for their therapeutic potential in a SKOV3ip xenograft mouse model. Only antibodies containing the IgG2a isotype were able to prolong survival efficiently. This was accompanied by the recruitment of macrophages into the tumor site. The antibody interaction with macrophages resulted in decreased tumor growth. Depletion of monocytes by clodronate pretreatment of tumor bearing mice abolished the therapeutic effect. Genome-wide analysis of tumor derived mRNA revealed that L1-9.3/IgG2a therapy induces altered expression of cellular genes associated with apoptosis, angiogenesis and chemotaxis. Further a humanized HuL1-9.3 and a chimerized ChiL1-9.3 antibody were generated, which showed similar binding characteristics compared to the paternal murine antibody L1-9.3. Both antibodies were able to significantly reduce tumor burden in vivo, but failed to show ADCC function in vitro. Alterations in the expression system were made to produce a modified antibody. This HuL1-9.3/F2N antibody could significantly increase antibody-dependent tumor cell lysis. The effect is most likely due to its modified glycosylation structure. These results establish that anti-L1 mAb therapy acts via immunological and non-immunological effector mechanisms to block tumor growth. The novel antibodies targeted against L1 could become helpful tools for the therapy of L1 expressing human carcinomas.
