Abstract

Die prolinreiche Tyrosinkinase 2 (Pyk2) beeinflusst eine Vielzahl unterschiedlicher Signaltransduktionswege wie Cytoskelettreorganisation, Zellanheftung und Zellbewegung, Neurotransmission und Neuroplastizität, Apoptose oder die Aktivierung verschiedener MAPK-Kaskaden. Es scheint plausibel, dass Pyk2 mit einer großen Zahl von Proteinpartnern interagiert und dadurch verschiedene „Downstream“-Signalwege steuert. Diese Interaktionspartner können Bindungspartner oder Substrate von Pyk2 sein. Bis heute sind nur wenige direkte Pyk2 Substrate bekannt. Das Ziel der vorliegenden Arbeit bestand darin neue Pyk2 Substrate zu identifizieren, um damit einen Beitrag zu einem besseren Verständnis der Koordination einer solchen funktionellen Diversität zu leisten. Um dieses Ziel zu erreichen wurde anhand eines bioinformatorischen Verfahrens, basierend auf bekannten, direkt von Pyk2 phosphorylierten Substraten eine Konsensussequenz entwickelt, mit deren Hilfe durch Datenbankrecherchen putative Substrate identifiziert werden konnten. Diese wurden durch in vitro Kinasereaktionen und Aktivitätsmessungen sowie zelluläre Analysen charakterisiert. Zur präzisen Kartierung der von Pyk2 phosphorylierten Tyrosinreste wurde ein breites Arsenal von Methoden angewandt und mittels MALDI-TOF/TOF-MS konnte schließlich gezeigt werden, dass der bioinformatisch vorhergesagte Tyrosinrest-351 in Brsk1 tatsächlich von Pyk2 phosphoryliert wird. Diese Befunde wurden durch gerichtete Mutagenese und Cotransfektionsstudien in Hek 293T Zellen bestätigt. Während analoge massenspektrometrische Ansätze für Brsk2 versagten, konnte in diesem Fall durch eine Kombination von gerichteter Mutagenese, Coimmunpräzipitation, in vitro Kinasereaktion und 2D-Phosphopetidkartierung Tyrosin-334 als Pyk2 Phosphorylierungsstelle in Brsk2 identifiziert werden. Ein Sequenzvergleich weiterer AMPK-verwandter Kinasen führte zur Entdeckung von vier weiteren potentiellen Pyk2 Substraten, den “MAP/microtubule affinity-regulated kinase” (Mark) Proteinen. Mark1-4 werden durch Pyk2 in vitro phosphoryliert. Neben Brsk1, Brsk2 und den vier Mark Proteinen ergaben in vitro Kinasereaktionen auch, dass Nuak1 und 2 mögliche Substrate von Pyk2 darstellen könnten. Die Aufklärung der Zusammenhänge zwischen Pyk2 und den bisher nur wenig charakterisierten AMPK-verwandten Kinasen stellt eine besondere Herausforderung dar und bietet zugleich die Möglichkeit vielleicht einen ganz neuen, bisher noch nicht identifizierten Signalweg zu beschreiben, der zu einem besseren Verständnis der Kinase Pyk2 selbst sowie ihren Substraten beitragen könnte.

Translation of abstract (English)

The proline-rich tyrosine kinase Pyk2 affects a multitude of signal transduction pathways like cytoskeletal reorganization, cell attachment and cell motility, neurotransmission and neuroplasticity, apoptosis and activation of several MAPK-cascades. Pyk2 interacts with a large number of protein partners and thereby controls different downstream signaling pathways. These interaction partners may be binding partners and eventually also substrates of Pyk2. However, until today only a few direct Pyk2 substrates are known. The aim of this study was the identification of new Pyk2 substrates to make a contribution to a better understanding of the coordination of such functional diversity. To achieve this aim a consensus sequence was engineered with the help of a bioinformatic approach based on sequences of known Pyk2 substrates, which helped to identify putative substrates via data bank research. From the list of candidate substrates, the AMPK-related kinases Brsk1 and 2 were chosen for further characterization by in vitro kinase assays, activity measurements and cellular analyses. For precise mapping of the tyrosine residues phosphorylated by Pyk2 a broad arsenal of methods were applied and it could be finally shown via MALDI-TOF/TOF-MS that the predicted tyrosine residue 351 in Brsk1 is actually phophorylated by Pyk2. These findings were confirmed by directed mutagenesis and cotransfection studies in Hek 293T cells. While similar massspectrometric approaches failed for Brsk2 a combination of directed mutagenesis, coimmunpecipitation, in vitro kinase reactions and 2D phosphopeptide mapping identified tyrosine 334 as Pyk2 phosphorylation site within Brsk2. While it could be demonstrated that the Pyk2-mediated tyrosine phosphorylation of Brsk1 and 2 increases their catalytic activity towards Tau1 and Wee1, the cellular implication of this new signalling node remains to be elucidated. A sequence alignment of other AMPK-related kinases resulted in the discovery of four more potential Pyk2 substrates, the MAP/microtubule affinity-regulated kinase (Mark) proteins. Mark1-4 were phosphorylated by Pyk2 in vitro. Beside Brsk1, Brsk2 and the four Mark proteins in vitro kinase assays also revealed that Nuak1 and 2 represent putative substrates for Pyk2. The clarification of an interrelation between Pyk2 and the so far poorly characterized AMPK-related kinases poses a peculiar challenge and provides an opportunity to describe a complete new, up to now not identified signaling pathway, that may contribute to a better understanding of Pyk2 itself and its substrates.
