Abstract

Reversible Proteinphosphorylierung ist ein Schlüsselprinzip der intrazellulären Signaltransduktion. Häufig wird eine Änderung des Phosphorylierungsstatus als relative Änderung zu einem Anfangsstatus angegeben. Hingegen bietet der Phosphorylierungsgrad eine wesentlich informativere Systembeschreibung und eine fundierte Grundlage für Modellrechnungen. One-source-Peptid-/Phosphopeptidstandards wurden mit dem Ziel entwickelt, Phosphorylierungsgrade von Proteinen positionsspezifisch und mit hoher Genauigkeit zu bestimmen. Kernpunkt der Methode ist die Erzeugung einer Mischung eines stabilisotopenmarkierten Peptid-/Phosphopeptid-Standardpaars, dessen stöchiometrisches Verhältnis exakt bekannt ist. Dazu wird eine Lösung des Phosphopeptidstandards in zwei Aliquots geteilt. Ein Aliquot wird mit antarktischer Phosphatase dephosphoryliert, während das andere Aliquot unbehandelt bleibt. Die Phosphatase wird anschließend durch kurzzeitiges Erhitzen irreversibel inaktiviert und aus beiden Lösungen eine volumetrisch definierte Mischung hergestellt. Die Standardpaare wurden den immunpräzipitierten und mittels SDS-Gelelektrophorese gereinigten Zielproteinen auf der Stufe des Verdaus zugegeben und die Analysen mittels nano-Ultra-Hochleistungsflüssigchromatographie-Tandemmassenspektrometrie durchgeführt. Bei Peptiden mit zwei Phosphorylierungsstellen erlaubte die Einführung mehrerer Isotopenmarkierungen eine individuelle Quantifizierung aller vier möglichen Formen. Zunächst wurde die Methode anhand der Parameter Wiederfindung, Richtigkeit und Reproduzierbarkeit erfolgreich validiert. Der relative Fehler bei der Analyse technischer Replikate betrug durchschnittlich weniger als 10 %, bei biologischen Replikaten hingegen 29 %. Der lineare Messbereich umfasste etwa zwei Größenordnungen, und die untere Bestimmungsgrenze für Phosphopeptide lag bei 10-20 fmol. Die Methode wurde auf die Phosphorylierungsgradbestimmung der Signalproteine STAT6, Akt1, ERK1 und ERK2 in verschiedenen Zellsystemen angewandt. Im Vordergrund stand die Analyse der dualen ERK1/2-Phosphorylierung durch die MAPK-Kinase MEK. Die ERK-Signalkaskade spielt eine zentrale Rolle in der Antwort von Zellen auf extrazelluläre Stimuli wie Wachstumsfaktoren und Zytokine. Dennoch sind die Mechanismen der dynamischen ERK-Aktivierung und –Deaktivierung in vivo nicht vollständig verstanden. Mit Hilfe von one-source-Standards wurden HGF- und IL6-induzierte ERK1/2-Phosphorylierungskinetiken in primären Maushepatozyten und der Tumor-Keratinozytenzelllinie HaCaT A5 analysiert. Es wurden zwei mathematische Modelle erstellt, die die quantitativen Daten basierend auf einem prozessiven oder distributiven ERK-Aktivierungsmechanismus beschreiben. Wie die experimentelle Validierung ergab, war nur das distributive Modell in der Lage, erfolgreich die Zeitspanne bis zur vollständigen Dephosphorylierung des totalen ERK1/2-Pools in Hepatozyten bei Hemmung der MEK-Aktivität vorauszusagen. Somit wurde erstmals ein zweistufiger, distributiver ERK-Phosphorylierungsmechanismus in primären Maushepatozyten nachgewiesen.

Translation of abstract (English)

Reversible protein phosphorylation is a key mechanism for intracellular signal transduction. In most studies changes in the phosphorylation status of a protein are analyzed as relative changes compared to an initial state. On the contrary, knowledge of the phosphorylation degree offers a more informative system description and a valuable basis for mathematical modeling. One-source peptide/phosphopeptide standards have been developed for highly accurate and site-specific phosphorylation degree determination of proteins. The main feature of this method is the generation of a mixture of a stable isotopically labeled peptide/phosphopeptide standard pair with exactly defined molar ratio. For preparation, a solution of the phosphopeptide standard is divided into two identical aliquots. One aliquot is quantitatively dephosphorylated by antarctic phosphatase while the other aliquot remains untreated. After incubation, antarctic phosphatase is irreversibly inactivated by gentle heat-treatment, and phosphorylated and dephosphorylated standards are combined at the desired volumetric ratio. Appropriate amounts were spiked into target protein digests generated after immunoprecipitation and SDS gel electrophoresis for purification. Analyses were performed by nano ultra performance liquid chromatography combined with tandem mass spectrometry. If the target peptide contained two phosphorylation sites, introduction of multiple isotopic labels allowed for the individual quantification of all four possible species. Firstly, recovery, accuracy and reproducibility of the method were successfully validated. For technical replicates the relative error was below 10 % on average. On the contrary, analysis of biological replicates resulted in a relative error of 29 %. Measurements were performed within a dynamic range of about two orders of magnitude and with a minimal phosphopeptide concentration of 10 to 20 fmol. The method was applied to phosphorylation degree determination of the signaling proteins STAT6, Akt1, ERK1 and ERK2 isolated from different cell systems. The main goal was to characterize the mechanism of dual ERK1/2 phosphorylation catalyzed by its upstream kinase MEK. The ERK signaling cascade plays a major role in cellular responses to extracellular stimuli like growth factors and cytokines. However, mechanisms of dynamic ERK activation and deactivation in vivo are not fully understood. Using one-source standards HGF and IL6 induced ERK1/2 phosphorylation kinetics in primary mouse hepatocytes and the tumor keratinocyte cell line HaCaT A5 were measured. Two mathematical models were established describing the experimental data on the basis of either a processive or distributive mechanism of ERK activation. Experimental validation revealed that only the distributive model was able to successfully predict the time span for complete dephosphorylation of the total ERK1/2 pool in the hepatocytes upon MEK inhibition. Thus, for the first time it was demonstrated that ERK phosphorylation in primary mouse hepatocytes occurs via a two-step, distributive mechanism.
