Abstract

In dieser Arbeit wurde die Leitstruktur des Saccharidmimetikums 3-ß-D-Galactopyranosyloxymethyl-4-sulfatomethylfuran (Galactosulfatofuran - GSF) optimiert. GSF hemmt die Bindung von humanen Melanomzellen an Proteine der Extrazellulärmatrix und unterbindet die zweidimensionale Zellmigration. Die dafür notwendigen GSF-Konzentrationen liegen allerdings im millimolaren-Bereich. Darüberhinaus sollten die optimierten GSF-Analoga, - dem Konzept der „Green Chemistry“ folgend, - gegenüber der Leitstruktur besser biologisch abbaubar sein. Das im Laufe dieser Arbeit erarbeitete Konzept enthält Optimierungen durch Austausch und Variation verschiedener Strukturelemente der Leitstruktur. Dazu zählten zum einen der Austausch der Galactose durch andere Saccharide und zum anderen die Modifikation des Core-Moleküles, Furan. Weiterhin wurden sowohl am glycosidischen Teil als auch am Furan-Core verschiedene Umfunktionalisierungen durchgeführt. Die von mir synthetisierte Substanzbibliothek wurde in verschiedenen biologischen Assays untersucht und zusammen mit Molecular Modelling gelang es Rückschlüsse auf die für den Wirkmechanismus und die biologische Abbaubarkeit der Saccharidmimetika notwendigen Strukturkriterien zu ziehen. Hierbei wurde deutlich, dass D-Galactose das optimale Saccharid ist, als Core-Molekül erwies sich 3,4-Bishydroxymethylfuran als die wirksamste Struktureinheit. Weitere Optimierungen stellten Umfunktionalisierungen der Galatose dar. Durch Anwendung gezielter Schutzgruppenstrategien konnte Galactose an Position 6 sulfatiert werden, so dass das zweifach sulfatierte Saccharidmimetikum 3-(6-Sulfato-ß-D-galactopyranosyl-oxymethyl)- 4-(sulfatomethyl)-furan (Galacto-bis-sulfatofuran - GSSF) erhalten wurde, welches im Vergleich zur Leitstruktur GSF ein deutlich potenterer Inhibitor der Zellmigration ist. Um den Einfluss der funktionellen Gruppe an der Position 3 des 3,4-Bishydroxymethylfurans beurteilen zu können, wurde die Sulfatgruppe durch andere Funktionalitäten ersetzt. Die optimale Verbesserung der Leitstruktur stellt das fluorierte Saccharidmimetikum 3-ß-DGalactopyranosyloxymethyl-4-fluoromethylfuran (Galactofluorofuran - GFF) dar, welches in 50fach geringerer Konzentration als GSF die Adhäsion von Melanomzellen an Fibronectin hemmt. Darüberhinaus war GFF im Test mit Klärschlammbakterien deutlich besser biologisch abbaubar als die Leitstruktur GSF. Das Ziel dieser Arbeit – eine Optimierung der biologischen Wirksamkeit monoglycosidischer Saccharidmimetika parallel zu einer Verbesserung ihrer biologischen Abbaubarkeit - wurde erreicht. Weiterhin können aus den erhaltenen Ergebnissen Rückschlüsse auf die Wirkmechanismen dieser Klasse von Inhibitoren gezogen werden. Chelatoren wie GSSF eignen sich als potente Inhibitoren der Zellmigration, wogegen halogenierte Saccharidmimetika wie z.B. GFF die Zelladhäsion an Extrazellulärmatrix-Proteine wie Fibronectin hemmen.

Translation of abstract (English)

The aim of this project was to optimise the lead compound 3-ß-D-Galactopyranosyloxymethyl-4-sulfatomethylfurane (galactosulfatofurane - GSF). GSF inhibits cell adhesion to proteins of the extracellular matrix as well as cell migration, but the necessary GSF concentrations are in the millimolar range. In addition the optimised analogues should be better biodegradable than the lead compound GSF in line with the concept of “green chemistry”. The developed strategy included improvements by exchanging and varying the different structural elements of the lead compound. In various biological assays and by molecular modelling the structural requirements of the saccharide mimetics for biological activity and to make them biodegradable were elucidated. D-galactose was thereby found to be the optimal saccharide and 3,4-bishydroxymethylfurane the most effective structural unit as core molecule. By applying protecting group strategies at position 6 of D-galactose this position was sulfated yielding 3-(6-sulfato-ß-D-galactopyranosyl-oxymethyl)-4-(sulfatomethyl)-furane (galacto-bis-sulfatofurane - GSSF), which in comparison to the lead compound GSF is a much more effective inhibitor of melanoma cell migration. To assess the influence of the functional groups on the furane core the sulfate was replaced by other functionalities. The best optimisation of the lead compound was achieved with the fluorinated furan 3-ß-D-galactopyranosyloxymethyl-4-fluoromethylfurane (galactofluorofurane - GFF), which inhibits the adhesion of melanoma cells to fibronectin at a 50-fold lower concentration than GSF. Moreover GFF showed a significantly improved biodegradability in one of the tests with sewage sludge bacteria than the lead compound GSF. The aims of this thesis - the optimisation of the biological activity of monoglycosylated saccharides concomitant with an improved biodegradability - have been achieved. In addition conclusions about the structural requirements necessary to inhibit cell migration and adhesion were derived from the experimental results. Chelating agents such as GSSF are suitable as potent inhibitors of cell migration, whereas halogenated saccharide mimetics e.g. GFF inhibit cell adhesion to extracellular matrix proteins such as fibronectin.
