Abstract

Der erste Teil der Arbeit beschreibt Derivatisierungen von Dioxabicyclooctenen. Als Ausgangsverbindung diente die 1,6-Anhydro-3,4-dideoxy-ß-D-erythro-hex-3-enopyranose, die nach 2-Tosylierung mit Kaliumbromid zur 2-Brom-1,6-anhydro-2,3,4-trideoxy-ß-D-erythro-hex-3-enopyranose und deren 4-Bromanalogon umgesetzt wurde. Die Produktzusammensetzung läßt eine SN1-Reaktion über ein allylisches Carbeniumion vermuten. Diese These wurde durch Umsetzung der 4-O-Tosyl-1,6-anhydro-2,3-dideoxy-ß-D-erythro-hex-2-enopyranose mit Kaliumbromid bestätigt, da auch in diesem Fall das 2- und das 4-Bromprodukt entstanden. Des weiteren wurden auf letzterem Weg die beiden [75Br]Bromanaloga hergestellt. Als Ligand für radioaktive Technetiumkomplexe sollte ein Thiol mit Dioxabicycloocten-Gerüst synthetisiert werden. Die Umsetzung der 4-O-Tosyl-1,6-anhydro-2,3-dideoxy-ß-D-erythro-hex-2-enopyranose mit Natriumhydrogensulfid führte zum Bis(1,6-anhydro-2,3,4-trideoxy-ß-D-erythro-hex-3-enopyranos-2-yl)thioether. Weder die Variation der Reaktionsbedingungen noch alternative Synthesewege führten zum gewünschten Produkt. Die Addition von elementarem Chlor bzw. Brom an die 1,6-Anhydro-3,4-dideoxy-ß-D-erythro-hex-3-enopyranose ergaben die 3,4-Dichlor- bzw. 3,4-Dibrom-1,6-anhydro-3,4-dideoxy-ß-D-glucopyranose. Der zweite Teil beschreibt einen einfachen und effizienten Zugang zum PET-geeigneten Proliferationsmarker [18F]FLT. Ausgehend von Thymidin wurde in einer vierstufigen Synthese der Precursor 3-N-Boc-1-(3-O-nosyl-5-O-trityl-ß-D-lyxofuranosyl)thymin erhalten, in einem selbst entwickelten Synthesemodul mit [18F]Fluorid umgesetzt und anschließend hydrolysiert. Die Aufarbeitung der Reaktionslösung erfolgte mittels HPLC und lieferte [18F]FLT in guten Ausbeuten, gebrauchsfertig für die Anwendung im Tierversuch. Als alternative 5´-Schutzgruppe wurden Dimethoxytrityl, als weitere Austrittsgruppen Tosyl bzw. Mesyl getestet, wobei die Kombination DMTr/Ns zu einer Optimierung der [18F]FLT-Ausbeute führte.

Translation of abstract (English)

The first part of this thesis describes the derivation of dioxabicyclooctenes. 1,6-Anhydro-3,4-dideoxy-ß-D-erythro-hex-3-enopyranose was tosylated in 2-position and then converted to 2-bromo-1,6-anhydro-2,3,4-trideoxy-ß-D-erythro-hex-3-enopyranose and 4-bromo-1,6-anhydro-2,3,4-trideoxy-ß-D-erythro-hex-2-enopyranose using potassium bromide. This product mixture indicates a reaction pathway via a SN1 mechanism involving an allylic carbenium ion. The SN1 mechanism was proven by the reaction of 4-O-tosyl-1,6-anhydro-2,3-dideoxy-ß-D-erythro-hex-2-enopyranose with potassium bromide forming the 2-bromo and 4-bromo analogues. Additionally, the corresponding [75Br]bromo derivatives were prepared. The treatment of 4-O-tosyl-1,6-anhydro-2,3-dideoxy-ß-D-erythro-hex-2-enopyranose with sodium sulfhydride resulted in bis(1,6-anhydro-2,3,4-trideoxy-ß-D-erythro-hex-3-enopyranos-2-yl)thioether. Neither variation of reaction conditions nor alternative synthetic routes gave the desired 4-O-thiol-1,6-anhydro-2,3-dideoxy-ß-D-erythro-hex-2-enopyranose. The addition of chlorine or bromine to 1,6-anhydro-3,4-dideoxy-ß-D-erythro-hex-3-enopyranose resulted in 3,4-dichloro- or 3,4-dibromo-1,6-anhydro-3,4-dideoxy-ß-D-glucopyranose, respectively. The second part of the thesis describes an easy and efficient synthetic route to the proliferation marker [18F]FLT. Starting from thymidine the precursor 3-N-Boc-1-(3-O-nosyl-5-O-trityl-ß-D-lyxofuranosyl)thymine was obtained in a 4-step synthesis. In a newly designed module the precursor was reacted with [18F]fluoride. After hydrolysis the reaction mixture was worked up by hplc to give [18F]FLT ready for animal treatment. Dimethoxytrityl was tested as an alternative to 5´-O-trityl protection. Additionally, the 3´-O-position was derived with tosyl or mesyl. Best results of radiofluorination were obtained employing DMTr/Ns.
