Abstract

Innerhalb eines North-Western-Expressionsscreens zur Identifizierung zellulärer mit dem RNA-Verpackungssignal Epsilon des Hepatitis-B-Virus interagierender Proteine, wurde aus einer Humanleber-cDNA-Bank eine für ein neues Protein ohne bekanntes RNA-Bindungsmotiv codierende cDNA (NIII-cDNA) isoliert (S.G. Kreft, Diplomarbeit 1996). Im Rahmen der vorliegenden Arbeit wurde die an ihrem 3´-Ende unvollständige NIII-cDNA zunächst komplettiert. Nach Isolierung mit der NIII-cDNA überlappender cDNAs ließen sich zwei Isoformen, RBP138 und RBP35 (=RNA-Bindungsprotein von 138 bzw. 35 kDa), ableiten. Mit dem zu dieser Zeit verfügbaren Working Draft des Humangenoms konnte sowohl das rbp138 Gen auf Chromosom 3 lokalisiert als auch die genomische Sequenz des rbp138 Gens bis zu etwa zwei Dritteln des RBP138-ORF abgeleitet werden. Als für die RNA-Bindung essentielle Region wurde eine K-reiche Domäne identifiziert, die zugleich ein neues RNA-Bindungsmotiv darstellt. Die mRNAs beider Proteine waren in sämtlichen untersuchten Geweben nur sehr niedrig exprimiert. Der Nachweis von endogenem RBP138 gelang in der Hepatomazellinie Huh7 nur nach vorausgegangener Immunpräzipitation. In Lokalisationsstudien wurde für beide Isoformen eine cytoplasmatische, vorwiegend perinukleäre Verteilung beobachtet, die eine Assoziation mit intrazellulären Membranen oder Cytoskelettstrukturen nahelegte. Aufgrund der anhaltenden Unlöslichkeit der Proteine konnte eine solche Assoziation in biochemischen Untersuchungen aber bisher noch nicht bestätigt werden. Das Auftreten einer potenziellen Protein-Protein-Interaktionsdomäne (RING-H2-Domäne) innerhalb von RBP138 läßt eine Interaktion mit weiteren zellulären Proteinen als wahrscheinlich erscheinen. Die Ergebnisse zusammengenommen, stellt RBP138 ein neues humanes RNA-Bindungsprotein mit einer neuen RNA-Bindungsdomäne dar, welches möglicherweise an cytoplasmatischen mRNA-Transport- oder Lokalisationsprozessen beteiligt ist.

Translation of abstract (English)

Previously, a cDNA (NIII-cDNA) encoding a novel protein without a known RNA-binding motif has been isolated from a human liver cDNA library in a North-Western expression screen carried out in search for cellular proteins interacting with the RNA encapsidation signal epsilon of the human hepatitis B virus (S.G. Kreft, diploma thesis 1996). In this work, first the 3´-terminally incomplete NIII-cDNA has been completed. After isolation of additional cDNAs overlapping with the NIII-cDNA it was possible to deduce two isoforms of this novel protein, RBP138 and RBP35 (RNA-binding protein of 138 and 35 kDa, respectively). Also, the rbp138 gene was mapped to chromosome 3 and rbp138´s genomic sequence was assembled up to about two thirds of the RBP138 open reading frame from the at that time publicly available working draft of the human genome. As region responsible for RNA-binding a K-rich domain representing a novel RNA-binding motif was identified. Expression studies on the mRNA level indicated only a low abundance of the transcripts of both isoforms in all tissues studied. This observation was further supported by the fact that endogenous RBP138 was detectable in Huh7 hepatoma cells only after immunoprecipitation. Both isoforms are localized exclusively in the cytoplasm, mainly perinuclearly and are probably associated with intracellular membranes or the cytoskeleton as shown by fluorescence microscopy. Thus far, it has not been possible to verify such an association of the proteins in biochemical studies because of their insolubility. Lastly, the existence of a potential protein-protein interaction domain (RING-H2 domain) within RBP138 makes interactions with other proteins likely. In summary RBP138 represents a novel human RNA-binding protein containing a novel RNA-binding domain. Concerning its function it might be speculated that RBP138 is involved in the cytoplasmic transport or localization of mRNA molecules.
