Abstract

Durch heterologe Komplementation von E.coli Knockout -Mutanten wurde das für Dihydroorotase kodierende Gen aus Solanum tuberosum und Arabidopsis thaliana kloniert. Beide Sequenzen kodieren für das in vivo funktionelle Protein. Im Falle der Kartoffel DHOase wurde durch RACE-PCR die vollst‰ndige untranslatierte 5'-Region (5'-UTR) ermittelt. Mit Hilfe spezieller Suchalgorithmen wurden mit dieser Information Rückschlüsse auf die kontrovers diskutierte subzelluläre Lokalisation des Enzyms in vivo gezogen. Die bisherige Meinung, DHOase sei im Plastiden lokalisiert, erscheint hiernach wenig wahrscheinlich. Durch transiente Genexpression, vermittelt durch Partikelbeschuss, konnten zwei unterschiedliche DHOase:GFP Fusionsproteine mit Confocaler Laser Scanning Mikroskopie (CLSM) im Cytosol von Arabidopsis Epidermiszellen nachgewiesen werden. Somit muß die chloroplastidäre Lokalisation der DHOase in Frage gestellt werden und diese statt dessen als cytosolisch postuliert werden. Die Entwicklung eines hochsensitiven Fluorimetrischen Enzymassays ermöglichte die nichtradioaktive Bestimmung der DHOase Aktivität in pflanzlichem Gewebe. Erstmals konnte somit ohne aufwendige Reinigungsprozeduren pflanzliche DHOase in Bezug auf pH-Optimum und Michaelis-Menten Konstante charakterisiert werden. Zur Untersuchung der Bedeutung der Pyrimidin de-novo Synthese wurden transgene Pflanzen mit reduzierter DHOase Expression hergestellt. Vermittelt durch die Antisense-Technik konnten Pflanzen mit verschieden stark ausgeprägter Reduktion der DHOase Expession isoliert werden. Diese wurden in Hinblick auf Metabolismus,Wachstum, Biomasse und Zelldichte untersucht. Es zeigte sich, dass bei starker Reduktion der DHOase Expression die Zellteilung deutlich vermindert ist und in Folge des geringeren Wachstums Kohlehydrate anstauen. Folglich kann durch endogene oder exogene Inhibierung der DHOase das Wachstum der Pflanze effektiv unterbunden werden. In zwei weiteren Ansätzen wurden Kartoffelpflanzen mit dem für DHOase kodierenden Gen aus E.coli transformiert, welches unter Kontrolle des CaMV 35S-, bzw. des B33-Patatin Promotors steht. In Transformanten beider Linien wurden bis zu einhundertfach erhöhte DHOase Aktivitäten gemessen. Eine Steigerung des Pflanzenwachstums oder des Knollenertrages infolge der erhöhten DHOase Expression wurde jedoch nicht beobachtet.

Translation of abstract (English)

The genes, coding for DHOase in Arabidopsis thaliana and Solanum tuberosum, were cloned by means of heterologous complementation of E.coli knockout mutants. Both sequences are coding for the functional protein in vivo. Furthermore the complete 5' untranslated region (5'-utr) of potato DHOase was revealed by RACE-PCR. That information, processed by special search algorithms, gave hints to the subcellular localisation of DHOase, which has been discussed controversial in the past. According to latest results the localization of DHOase in chloroplasts seems to be less likely. Two different DHOase:GFP fusions for transient gene expression were transformed into epidermal cells of Arabidopsis by particle bombardment. GFP signals were detected by confocal laser scanning microscopy (CLSM) in the cytosol. Thus the chloroplastidary localization of DHOase has to be reconsidered and instead postulated to be cytosolic. Development of a highly sensitive fluorimetric enzyme assay gave way to measure DHOase activity in plant tissue by non-radiactive means. For the first time DHOase has been characterized in regard to pH-optimum and Michaelis-Menten constant without the need of extensive purification procedures. Mediated by antisense suppression transgenic plants with reduced levels of DHOase expression were made to investigate the importance of pyrimidine de-novo synthesis. These plants were analyzed in regard to metabolism, growth, biomass and cell density. It was shown that a high reduction of DHOase expression led to a decrease in cell division and as a consequence of that to an accumulation of carbohydrates. Thus plant growth can be effectively blocked by endogenous or exogenous inhibition of DHOase. In two further approaches plants were transformed with the E.coli DHOase coding gene under control of the CaMV 35S promoter and the B33-Patatine promoter respectively. In both sets transformants showed DHOase activities up to a hundred fold higher than the wildtype. However an increase in plant growth or tuber production was not observed.
