Abstract

Für die autonomen Parvoviren MVMp und H-1 Virus wurden onkotrope und onkolytische Eigenschaften beschrieben, also eine präferentielle Replikation dieser Viren mit einhergehender Lyse in transformierten Zellen. Eine Transformation von Zellen mit Onkoproteinen wie aktiviertem Ras Protein führt zu einer Steigerung der Expression vom frühen P4 Promotor und zu einer verstärkten Toxizität des Nichtstrukturproteins NS1. Die Verwendung der P4-ns1-P38 Kassette aus H-1 Virus in einem AAV-2 Vektor sollte diesem Hybridvektor TRH1 neue Eigenschaften verleihen. Die Eigenschaften des Hybridvektors TRH1 wurden einerseits hinsichtlich der Transduktion charakterisiert, andererseits bezüglich einer selektiven Expression in transformierten Zellen. Der Hybridvektor TRH1-GFP zeigte in HeLa Zellen eine erhöhte Basistransduktion gegenüber herkömmlichen AAV-2 Vektoren, vermutlich aufgrund einer stärkeren Expression. Eine Transduktion mit AAV-2 Vektoren kann durch verschiedene Behandlungen der Zellen oder Koinfektion mit Adenovirus 5 stimuliert werden. Eine Transduktion mit TRH1-GFP kann neben einer Koinfektion mit Adenovirus 5 durch UV- und Gamma-Bestrahlung und einer Behandlung der Zellen mit Chemotherapeutika wie Cisplatin, Mitomycin und Carmustine (BCNU) gesteigert werden. Somit verhält sich der Hybridvektor TRH1-GFP wie ein reiner AAV-2 Vektor. Eine Stimulierung der Transduktion mit AAV-2 Vektoren durch Chemotherapeutika oder Gamma-Bestrahlung bietet die Möglichkeit einer Kombinationstherapie. Die hier beschriebene Möglichkeit einer Stimulierung durch BCNU erweitert das Spektrum kombinierbarer Medikamente mit einer AAV Vektor vermittelten Gentherapie. In einem Vergleich der Infizierbarkeit verschiedener Tumorzellinien mit dem Hybridvektor TRH1-GFP gegenüber einem rekombinanten H-1 Vektor, rH1-GFP, konnten die Tumorzellinien erwartungsgemäß mit unterschiedlicher Effizienz transduziert werden. Die Verwendung des Hybridvektors TRH1-GFP in einem AAV-2 Kapsid vermittelt also ein anderes Spektrum infizierbarer Zellen gegenüber einem rekombinanten H-1 Vektor und erweitert so den Einsatzbereich für die Verwendung der P4-ns1-P38 Kassette. In vitro konnte für die Hybridvektoren TRH1-GFP und TRH1-TK, aber auch für herkömmliche AAV-2 Vektoren, eine selektive Expression in transformierten Zellen unter Verwendung von Zellpaaren gezeigt werden. In einem in vivo Tumormodell wurde die Expression verschiedener parvoviraler Vektoren untersucht. Es wurde ein Vektor verwendet, der sich von H-1 Virus ableitete (rH1-hRluc), ein herkömmlicher AAV-2 Vektor (CMV-hRluc) und der Hybridvektor TRH1-hRluc. Die Expression der verschiedenen Vektoren im Leberzellkarzinom (MH3924A) wurde mit der im Leberparenchym verglichen. Alle verwendeten parvoviralen Vektoren zeigten eine stimulierte Expression im Tumorgewebe gegenüber der im Leberparenchym. Jedoch erhöhte die Verwendung der P4-ns1-P38 Kassette im Hybridvektor TRH1-hRluc verglichen mit CMV-hRluc in diesem Tumormodell nicht die Selektivität der Expression im Leberzellkarzinom gegenüber der im Leberparenchym. Die Expression von Vektoren der autonom replizierenden Parvoviren in normalen gegenüber transformierten Zellen wurde bisher nur in vitro, jedoch nicht in vivo untersucht. Der in diesem in vivo Tumormodell ermittelte Faktor einer Stimulierung der Expression des Hybridvektors TRH1-hRluc im Leberzellkarzinom war vergleichbar mit dem Faktor bei der Verwendung des Vektors rH1-hRluc.

Translation of abstract (English)

The autonomously replicating parvoviruses MVMp and H-1 virus are oncolytic viruses with oncotropic properties, i.e. preferential replication in and lysis of transformed cells. Oncogenic transformation of cells by oncoproteins like activated Ras protein stimulates expression from the early viral promotor P4, leading to increased levels of non-structural protein 1, NS1, and furthermore, enhances cytotoxicity mediated by this NS1 protein. Transfer and usage of the P4-ns1-P38 cassette derived from H-1 virus into an AAV-2 vector should confer new qualities to this hybridvector TRH1. On the one hand transduction properties of this hybridvector TRH1 have been characterized and on the other hand specificity of expression in transformed cells has been proved. In HeLa cells hybridvector TRH1-GFP showed enhanced levels of transduction compared to conventional AAV-2 vectors, presumably due to enhanced expression. In general transduction with AAV-2 vectors can be stimulated by different treatments or coinfection with adenovirus 5. Besides a coinfection with adenovirus 5, transduction with TRH1-GFP was augmented by irradiation of the cells with UV- or X-rays and treatment of the cells with chemotherapeutic drugs such as Cisplatin, Mitomycin or Carmustine (BCNU). Concerning transduction properties hybridvector TRH1-GFP behaved like a pure AAV-2 vector. There is a chance in the treatment of cancer to combine AAV-2 vector mediated gene therapy with an anti-cancer therapy which also enhances transduction efficiency. BCNU as described here broadens the field of drugs for combinatorial therapies. The ability of the hybridvector TRH1-GFP to transduce a panel of transformed cells of different origin was compared to the ability of a recombinant H-1 vector, rH1-GFP. As expected these vectors showed a different pattern in their ability to transduce those cells. Infections with the hybridvector TRH1-GFP are mediated at least in early infection steps by the AAV-2 capsid, which expands the spectrum of cells becoming transduced by parvoviral vectors carrying the P4-ns1-P38 cassette. In vitro both hybridvectors TRH1-GFP and TRH1-TK and conventional AAV-2 vectors were shown to express selectively in transformed cells, making experimental use of cell pairs of transformed and normal cells. In vivo expression of different parvoviral vectors was investigated. Therefore expression profiles from a recombinant H-1 vector, rH1-hRluc, a conventional AAV-2 vector, CMV-hRluc, and hybridvector TRH1-hRluc were compared in hepatocellular carcinoma (MH3924A) bearing rats. Levels of expression of the different vectors in liver tumors were compared to the corresponding expression in normal liver tissue. Expression of all parvoviral vectors was increased in tumors compared to normal liver tissue. However, usage of the P4-ns1-P38 cassette within hybridvector TRH1-hRluc did not yield a benefit compared to CMV-hRluc concerning selectivity of expression in tumor tissue. Selectivity of expression, applying vectors derived from the autonomously replicating parvoviruses MVMp or H-1 virus to transformed cells versus normal cells, was shown in vitro but had not been previously investigated in vivo. Stimulation of expression of hybridvector TRH1-hRluc in the liver tumor as determined here was comparable to the stimulation of expression of rH1-hRluc.
