Abstract

Gehirntumoren, allen voran Gliome, stellen eine heterogene Zellpopulation aus transformierten Zellen und einwandernden Leukozyten dar. Bis zu 45% der Tumormasse sind Makrophagen und Mikroglia, die auch als Makrophagen des Gehirns bezeichnet werden. Die Funktion dieser Fresszellen innerhalb des Tumors ist noch weitestgehend unklar und wird als wachstumsfördernd, aber auch tumorbekämpfend angesehen. Die vorliegende Arbeit sollte zur Klärung dieser Frage beitragen. Es sollte untersucht werden, ob murine, primäre Mikroglia in vitro in der Lage sind, nach Aktivierung toxisch auf Gehirntumorzellen zu wirken. Es zeigte sich, dass Mikroglia nach Priming mit IFN-g und Aktivierung mit LPS toxische Komponenten sezernieren, die in drei verschiedenen Gliomzelllinien Apoptose hervorrufen. Die untersuchten Zellen starben - abhängig von der Zelllinie - einen bis vier Tage nach der Behandlung. Der Zelltod wurde weder durch Tumor-Nekrose-Faktor (TNF)-a noch durch Stickstoffmonoxid (NO) ausgelöst; eine Beteiligung von Interferon (IFN)-b konnte nicht eindeutig ausgeschlossen werden. Inwieweit solche toxischen Effekte jedoch in vivo ausgeübt werden können, ist unklar. Durch vom Tumor sezernierte, immunsupprimierende Substanzen wird die Immunkompetenz der Mikroglia wahrscheinlich unterdrückt. Daher müssen Wege gefunden werden, diese potenziell toxischen Zellen in situ zu aktivieren. Eine Möglichkeit bietet der Einsatz onkotroper parvoviraler Vektoren, die über ein bis mehrere Transgene eine Aktivierung des Immunsystems einleiten könnten. Als Vorarbeit dazu wurde im Rahmen dieser Arbeit die Infektion von murinen, primären Gliazellen mit dem murinen Parvovirus MVM untersucht. Es zeigte sich, dass sowohl Astrozyten als auch Mikroglia in der Lage sind, das Virus zu internalisieren. Replikation der viralen DNA und Expression von viralen Proteinen findet in nur 5-10% der Astrozyten, nicht aber in Mikroglia statt. MVM hat keinen Einfluss auf die Aktivierung beider Zelltypen mit LPS und IFN-g; die Zytotoxizität der Mikroglia gegen Hirntumorzellen wird weder positiv noch negativ beeinflusst. Die vorliegende Arbeit bietet eine Grundlage für eine mögliche Gentherapie von Gehirntumoren. Mikroglia könnten innerhalb der Tumormasse aktiviert werden und so Apoptose der transformierten Zellen auslösen. Die Aktivierung der Zellen könnte mit Hilfe parvoviraler Vektoren gezielt unterstützt werden.

Translation of abstract (English)

Brain tumors (glioma) are a heterogeneous cell population composed of transformed cells and infiltrating leukocytes. Up to 45% of the overall tumor mass consists of macrophages and microglia, the macrophages of the brain. The function of these phagocytes - being endowed with tumor-promoting as well as with tumor-destructive capacities - inside the tumor is largely unknown. The aim of this work was to try and decipher the role of microglia in the biology of the tumor. The ability of murine primary microglia to exert toxic effects against brain tumor cells after activation in vitro was investigated. The results show that microglia, after priming with IFN-g and triggering with LPS, indeed secrete toxic compounds that induce apoptosis in three different glioma cell lines. These cells died, depending on the cell line, within one to four days after treatment with culture supernatant of activated microglia. Cell death was neither mediated by Tumor Necrosis Factor (TNF)-a nor by nitric oxide (NO); involvement of Interferon (IFN)-b in inducing apoptosis could not be definitely ruled out. Whether microglia can exert these toxic effects in vivo remains to be analyzed. Tumor-derived, immunosuppressive factors such as transforming growth factor (TGF)-b or Prostaglandin E2 (PGE2) could likely disable immunocompetent microglia. Therefore new ways for activating in situ these potentially toxic cells have to be found. A promising strategy lies in the use of the oncotropic parvoviral vectors, that could – via the transduction of one or more transgenes into tumor cells – initiate or support an immune response against the tumor. The capacities of murine primary astrocytes (the cell type from which glioma derive) and microglia to be infected by the autonomous parvovirus MVM and possible effects of the parvovirus on their immunocompetence were investigated. Astrocytes as well as microglia internalize the virus. However, replication of the viral DNA and expression of viral proteins took place in only 5-10% of the astrocytes, but not in microglia. MVM had no influence on the activation of both cells with LPS and IFN-g; cytotoxic properties of microglia against brain tumor cells were neither positively nor negatively affected. This thesis provides a basis for a possible therapeutical approach for brain tumors involving parvovirus-based gene therapy. According to this in vitro study, microglia might be activated inside the tumor mass and thereby induce apoptosis of transformed cells. Activation of microglia might be selectively supported by means of parvoviral delivery of immunostimuli.
