Abstract

Das zentrale Ziel der vorliegenden Arbeit war die Untersuchung einer durch ein potentielles HIV-Vakzin induzierten Immunantwort, welches auf konformationellen Env-Epitopen basiert, die durch die Interaktion mit dem zellulären Rezeptorkomplex (CD4/CXCR-4) induziert werden. In unserer Arbeitsgruppe wurde bereits gezeigt, dass Pseudovirionen (PV) mit eingebauten HIV-Env- bzw. CD4/CXCR-4-Molekülen eine Env-vermittelte Fusion miteinander eingehen können. In dieser Arbeit wurde ein Env- und CD4/CXCR-4-PV-Gemisch in einer Zwischenphase der Fusion fixiert, während derer induzierte Env-Epitope exponiert werden, und die induzierte Immunantwort nach Immunisierungen von Versuchstieren mit diesem PV-Vakzin analysiert. Am Anfang dieser Arbeit wurden Versuche zur Charakterisierung und Verbesserung der Zusammensetzung und Eigenschaften der für das PV-Vakzin verwendeten Pseudovirionen durchgeführt. Wesentliche Verbesserungen wurden hinsichtlich der Effizienz der PV-Herstellung und bezüglich der PV-Reinheit erzielt. Zahlreiche Immunisierungsexperimente in nicht transgenen Mäusen zeigten, dass die Immunisierung mit Pseudovirionen allein keine starke Immunantwort induziert, und dass eine prime-Immunisierung mit rekombinanten Env-Proteinen notwendig ist, um das Immunsystem effizient auf die geringen Mengen konformationell relevanter Env-Moleküle im PV-Vakzin zu lenken. Neben der Induktion von Env-Antikörpern in den Seren erlaubte das veränderte Protokoll auch die Generierung Env-spezifischer monoklonaler Antikörper (Mabs). Da das PV-Vakzin auch CD4 und CXCR-4 enthielt, wurden weitere Immunisierungen in dagegen toleranten huCD4/huCXCR-4 transgenen Ratten durchgeführt. In einem initialen Immunisierungsexperiment wurden durch das PV-Vakzin neutralisierende Antikörper im Serum induziert und außerdem ein Env-spezifischer monoklonaler Antikörper (R I.23 B/3) generiert, der ein heterologes HIV 1 Primärisolat (J27) neutralisieren kann. Unseres Wissens ist dies der erste nach Vakzinierungen in Versuchstieren hergestellte Mab, der ein heterologes HIV-1 Primärisolat neutralisieren kann. Obwohl die Serumantikörper der vakzinierten Ratte kein Vermögen zur Kreuzneutralisation zeigten, war der Titer gegen homologes Virus höher als in den Seren der Ratten, die mit großen Mengen an rekombinanten Env-Proteinen geboostet wurden. Außerdem zeigte das Tier, welches dieselbe Menge an Env-PV wie im PV-Vakzin erhalten hatte (allerdings ohne ein Mischen mit CD4/CXCR-4-PV), keine neutralisierende Immunantwort. Dies bedeutet möglicherweise, dass konformationelle Epitope des PV-Vakzins für die Induktion der neutralisierenden Immunantwort verantwortlich sind und die geringen Mengen der darin enthaltenen Env-Proteine kompensieren. Diese Immunantworten wecken die Hoffnung, dass Optimierungen des PV-Vakzins, insbesondere die Erhöhung des Env-Einbaus in PV, zur Induktion einer qualitativ besseren Immunantwort führen und somit die Grundlage für weitere Forschungsansätze zur Anwendung im Menschen darstellen könnten.

Translation of abstract (English)

The major aim of this thesis was to analyze the responses elicited by a potential HIV vaccine based on conformational Env epitopes, induced on interaction with the cellular receptor complex (CD4/CXCR-4). Previous experiments in our laboratory have shown that pseudovirions (PVs), with incorporated HIV-Env or CD4/CXCR-4, respectively, can undergo Env-mediated fusion with one another. Thus in the present strategy, mixtures of these Env- and CD4/CXCR-4-PVs were fixed at an intermediate fusion stage, when induced Env conformations are exposed, and the immune responses induced after immunization of experimental animals were analysed. In the early stages of this thesis, experiments to establish and to improve the composition and properties of the respective pseudovirions, which constitute the potential vaccine, were performed. Major improvements with respect to time and ease of PV-production and PV-purity were achieved. Several series of immunizations in non-transgenic mice revealed that inoculation with PVs alone was not sufficient to induce a robust immune response and that a prime with recombinant Env-protein was necessary in order to focus the immune system on the small amount of conformationally relevant Env in the PV-vaccine. In addition to the induction of Env-specific serum antibodies, this modified protocol now allowed the generation of Env specific monoclonal antibodies (Mabs). Since CD4 and CXCR-4 are components of the PV-vaccine, further experiments in huCD4/huCXCR-4 transgenic rats, which are tolerant to these components, were carried out. In an initial experiment, neutralizing serum antibodies were induced by the vaccine and an Env-specific rat monoclonal antibody (R I.23 B/3), capable of neutralizing a heterologous primary HIV-1 strain (J27), was generated. To our knowledge, no further Mabs, elicited by a vaccine in an animal model, and capable of neutralizing a primary HIV, have been described. Although the serum antibodies in the vaccinated rat did not exhibit broadly neutralizing activity, nevertheless, the titer against homologous virus was higher than in control sera of rats boosted with high amounts of recombinant Env protein. Furthermore, the animal which had received the same amount of Env-PV as in the PV-vaccine (but not mixed with CD4/CXCR-4-PV) did not exhibit a neutralization response. This could indicate that induced epitopes in the PV-vaccine are instrumental in the induction of the neutralization response and compensate for the low amount of total Env protein in the PV-preparation. These immune responses raise the hope that improvements in the PV-vaccine composition in particular by increasing the extent of Env incorporation may lead to the induction of a superior immune response and lay the basis for further developments for human use.
