Abstract

Im Rahmen dieser Doktorarbeit wurden selbstaggregierende Monolagen (SAM) proteinresistenter ethylenglykolterminierter (EG) Alkanthiole auf Metallsubstraten mit Hilfe verschiedener Meßmethoden, wie der Summenfrequenzspektroskopie (SFS), der Neutronenreflektometrie, aber auch der Ellipsometrie und der Infrarotspekroskopie charakterisiert. Da seit Beginn der 80er Jahre aus den Arbeiten von Jeon und Andrade et al. bekannt ist, daß die starke Wechselwirkung des Wassers über Wasserstoffbrückenbindungen mit Poly(ethylenglykol) (PEG) eine entscheidende Rolle für die Proteinrepulsion von PEG-beschichteten Oberflächen nach dem Mechanismus der sogenannten „sterischen Repulsion“ spielt, wurde in dieser Arbeit besonders die Wechselwirkung des Wassers mit PEG- bzw. oligo(ethylenglykol)terminierten (OEG) Alkanthiol-SAMs untersucht. [Ref. 1-3] Bei der Optimierung der PEG- und OEG-SAM-Präparation auf Gold wurde nachgewiesen, daß die Gegenwart von Sauerstoff in der Reaktionslösung die Bildung dichtgepackter OEG-SAMs fördert und bei PEG-SAMs erst ermöglicht. Dies wurde dadurch interpretiert, daß der bei der Reaktion von Thiol mit Gold frei werdende Wasserstoff mit dem Sauerstoff reagiert und sich dadurch günstig auf die Reaktionsenthalpie auswirkt. Des weiteren wurden die Banden der SFS-Spektren von SAMs bestehend aus methoxyhexa(ethylenglykol)terminiertem Undekanthiol (EG6OMe) auf Gold zugeordnet und für die Interpretation anderer Messungen verwendet. Zu diesen zählen Experimente an EG6OMe-Alkanthiolmischfilmen, die bei Erhöhung der Temperatur einen Phasenübergang aufweisen, durch den sie ihre Proteinresistenz verlieren. Außerdem wurde die These aus früheren Experimenten von Herrwerth et al. bestätigt, daß die Proteinresistenz von dichtgepackten, kristallinen OEG-SAMs hauptsächlich von deren lateralen Packungsdichten und folglich von deren Hydratisierbarkeit abhängt. [Ref. 4] Bei der Untersuchung von EG-Filmen in wassergesättigter Stickstoffatmosphäre wurde eine Schichtdickenzunahme durch Bildung einer 2 3 nm dicken Wasserschicht auf PEG-SAMs und einer etwas dünneren Wasserschicht auf OEG-SAMs gemessen, die bei anderen hydrophilen Oberflächenbeschichtungen nicht gefunden werden konnte. Ferner war es möglich die feste Bindung von Wassermolekülen an die Endgruppen von OEG-SAMs mit Hilfe von SFS indirekt durch Untersuchung der Konformationsänderungen der Filme vor und nach Wasserkontakt nachzuweisen. Diese Resultate wurden als Hinweis auf die Bildung einer geordneten Wasserstruktur gedeutet, die nach Arbeiten von Kushmerick et al. einen möglichen Mechanismus der Proteinresistenz von dichtgepackten OEG-SAMs darstellt. [Ref. 5] Die feste Bindung bestätigt allerdings auch Arbeiten von Kreuzer et al. [Ref. 6] und Chan et al. [Ref. 7], die die Bildung und Immobilisierung von Hydroxidionen als Grund für die Proteinabstoßung sehen. Translation of abstract (English)

During the work for this dissertation self-assembled monolayers (SAM) of protein resistant, ethylene glycol (EG)-terminated alkanethiols on metal substrates have been characterized using different measuring methods like sum frequency spectroscopy (SFS), neutron reflectivity, but also ellipsometry and infrared spectroscopy. Since the beginning 1980s it is well-known from the studies of Jeon and Andrade et al. that, according to their mechanism of steric repulsion, the strong interaction of water via hydrogen bonds with poly(ethylene glycol) (PEG) plays a vital role for the protein resistance of PEG-coated surfaces. Therefore this work focused mainly on investigating the interaction of water with SAMs consisting of PEG- and oligo(ethylene glycol) (OEG)-terminated alkanethiols respectively. [Ref. 1-3] During the optimisation of PEG and OEG SAM preparation on gold surfaces it was proved, that the presence of oxygen in the reaction solution promotes the formation of densely packed OEG SAMs and is a necessity for forming densely packed PEG SAMs. This result has been interpreted as a reaction of oxygen with hydrogen originating from the reaction of thiol with gold, which is favourable for the enthalpy of the reaction. In addition the bands from SFS spectra on methoxy hexa(ethylene glycol) (EG6OMe)-terminated undecanethiol SAMs on gold were assigned and used for the interpretation of other measurements, such as the experiment on EG6OMe/alkanethiol mixed monolayers, that lose their ability to resist the adsorption of proteins due to a phase transition when heating the film. Furthermore the assumption from earlier experiments from Herrwerth et al., that the protein resistance of densely packed, crystalline OEG SAMs depends mainly on their lateral packing density and hence on their ability to be hydrated, could be verified. [Ref. 4] A series of experiments in water saturated nitrogen atmosphere could show an increase in film thicknesses of 2-3 nm of OEG and PEG SAMs. Other hydrophilic surface coatings did not show a similar behaviour. In addition it was possible to prove indirectly the strong binding of water molecules to the OEG endgroups by investigating the conformational changes of films before and after water contact using SFS. These results could be interpreted as a clue for the formation of an ordered water structure that represents, according to the work of Kushmerick et al., a possible mechanism of protein resistance for the densely packed OEG SAMs. [Ref. 5] But the strong binding of water to the endgroups of an OEG film also approves studies, carried out by Kreuzer et al. [Ref. 6] and Chan et al. [Ref. 7], which suggest that the formation and immobilisation of hydroxide ions from the bound water are the main reasons the protein repelling properties of OEG SAMs. Both mechanisms are discussed in this thesis.
