Abstract

HINTERGRUND: Damit Tumorantigen-spezifische T-Zellen Tumorzellen spezifisch erkennen und zerstören können, müssen sie zuvor durch professionelle Antigen-präsentierende Zellen, wie den Dendritischen Zellen (DC) aktiviert werden. Neuere Therapiekonzepte konzentrieren sich deshalb auf den Einsatz von Tumorantigen-beladenen DC als Vakzine. ZIEL: Das Ziel der vorliegenden Arbeit war es, die Elektroporation von DC mit RNA zu etablieren und die T-Zell-stimulatorischen Eigenschaften RNA-elektroporierter DC zu analysieren. METHODEN UND ERGEBNISSE: Die RNA-Elektroporation wurde für dendritische Zellen, sowie für andere Antigen-präsentierende Zellen etabliert. Die Analyse der T-Zell-stimulatorischen Kapazität RNA-elektroporierter DC erfolgte nach Elektroporation von RNA, kodierend für das immunodominante pp65 Antigen des humanen Cytomegalievirus. Es konnte eine effiziente Stimulation pp65-spezifischer CD8+-T-Zellen erzielt werden. Die Fähigkeit, RNA-elektroporierter DC eine spezifische Stimulation von CD4+-T-Zellen zu vermitteln, wurde im Tumormodell untersucht. DC wurden mit der RNA des Tumorantigens Melanotransferrin (Mtf) elektroporiert und mit T-Zellen, spezifisch für das erst kürzlich identifizierte HLA-DRB1*0401-präsentierte Mtf668-683-Epitop, inkubiert. Eine spezifische Stimulation der T-Zellen durch Mtf-RNA-elektroporierte DC konnte detektiert werden. Weiterhin wurde die Präsenz des Mtf668-683-Epitops in gesunden Donoren und Melanompatienten analysiert. Dabei wurden Epitop-spezifische T-Zellen in beiden Gruppen nachgewiesen. Die erzielten Daten zeigen demnach, dass RNA-elektroporierte DC Antigen-spezifische T-Zellen effizient stimulieren können, so dass ihr Einsatz in Rahmen von Antigen-spezifischen Vakzinierungsstrategien denkbar ist.

Translation of abstract (English)

BACKGROUND: Tumor-antigen specific T-cells need to be activated by Dendritic cells (DC) to gain the ability to recognice and destroy tumorcells efficiently. Therefore new strategies concentrate on the development of vaccines using DC loaded with tumor antigen. The aim of this study was to establish electroporation of DC with RNA thereby analysing the capability to stimulate antigen specific T-cells. METHODES AND RESULTS: RNA electroporation was established for DC as well as other antigen presenting cells. The capacity of RNA electroporated DC to stimulated antigen specific T-cells was analysed by electroporation of RNA encoding for the immunodominant Antigen pp65 of the human Cytomegalievirus. At this an efficient stimulation of pp65-specific CD8+-T-cells could be achieved. The ability of RNA electroporated DC to stimulate CD4+-T-cells was analysed by electroporation of DC with the tumor-antigen Melanotransferrin (Mtf). Those DC were than incubated with T-cells specific for the recently identified HLA-DRB1*0401 restricted Mtf668-683-Epitope resulting in the detection of specific T-cell stimulation. Furthermore healthy individuals as well as melanoma patients were examined for the presence of the Mtf668-683-Epitope. In both groups epitope specific T-cells were detected. In line with this results the propotion can be made, that RNA electroporated DC can efficiently stimulate antigen-specific T-cells. Hence the application in new vaccinationtrials is conceivable.
