Abstract

Saure Invertasen haben im pflanzlichen Stoffwechsel eine große Bedeutung. Sie spielen eine wichtige Rolle bei der Regulation sink-Stärke und damit zusammenhängend der Assimilatverteilung in der Pflanze. Dar¬über hinaus beeinflussen sie die Zusammensetzung löslicher Zucker. Nachdem diese auch als Signale für verschiedene Stoffwechselprozesse dienen, kommt den Sauren Invertasen auch eine Funktion bei der pflanzli¬chen Entwicklung sowie verschiedenen Stressreaktionen zu. Eine Komponente der posttranslatio¬nalen Regulation der Invertasen stellen die Invertase-Inhibitoren dar, kleine Proteine, die spezifische Komplexe mit den Enzymen bilden und ihre Aktivität herabsetzen. Sowohl die Vakuolären Invertasen (VIs) der Solanaceen sowie insbesondere der vakuoläre Inhibitor Nt VIF der Modellpflanze Nicotiana tabacum (Tabak) wurden im Rahmen dieser Arbeit molekularbiolo¬gisch und bio¬chemisch näher charakterisiert. Die partiellen Sequenzen von VIs aus Tabak (Nt VI) und Kartoffel (St VI) wurden ver¬vollständigt. Bei etwa konstanter Proteinmenge unterscheidet sich die Akti¬vität der löslichen Invertasen in den vegetativen Geweben bei Tabak erheblich, was auf regulierende Me¬chanismen hinweist. Der Inhibitor Nt VIF wurde in dieser Arbeit erstmals als funktionelles Protein rekombinant hergestellt. Anders als die apoplastische Isoform Nt CIF, der die Invertasen beider Kompartimente gleichermaßen zu hemmen vermag, inhibiert Nt VIF nur die Vakuoläre Invertase. Durch Experimente mit chimären Proteinen, die zu je einer Hälfte aus der einen und anderen Isoform bestehen, konnte ermittelt werden, dass der N-Terminus für die Spezifität verantwortlich ist. Eine Deletion weniger Aminosäuren an diesem Ende führt zur Inaktivierung von Nt VIF. C terminal können über 18 Aminosäuren fehlen, ohne dass die Inhibitonsfähigkeit beeinträchtigt wird. Die Mutation bereits eines der konservierten Cysteine führt ebenso wie die chemische Reduktion am re¬kombinanten Nt VIF zur Inaktivität des Inhibitors. Die postulierte Bedeutung von Disulfidbrücken für die strukturelle Integrität des Proteins konnte dadurch experimentell bestätigt werden. Paradoxerweise fördern viele nicht aktive rekombinante Inhibitoren in dieser wie auch in anderen Arbei¬ten die Aktivität der Invertasen. Die Analysen sprechen für ein Artefakt, das durch nicht nativ gefaltete rekombinante Proteine ausgelöst wird. Sie aggregieren wahrscheinlich mit den endogenen Inhibitoren der Invertase-Präparation und führen so zu einer scheinbaren Erhöhung der Aktivität. Der endogene Nt VIF konnte bisher nicht in Pflanzen nachgewiesen werden, obwohl das Gen transkri¬biert wird. Die Expression wird daher nur in speziellen Gewebebereichen vermutet.

Translation of abstract (English)

Acid invertases play a crucial role in plant metabolism. They strongly influence sink strength and assimilate partitioning. Additionally they are involved in regulation of sugar composition. As soluble sugars function as signals, these enzymes are connected with several developmental processes and stress reactions. One component in posttranslational regulation are invertase inhibitors, small proteins that form specific complexes with invertases and lower their activity. This work investigates vacuolar invertases (VI) of the solanaceae family with special emphasis on the vacuolar inhibitor Nt VIF of the model plant Nicotiana tabacum (tobacco). The partial sequences of a tobacco (Nt VI) and a potato (St VI) invertase have been completed. Although different vegetative tissues show similar protein amounts the activity differs strongly indicating regulatory mechanisms. The inhibitor Nt VIF was overexpressed and renaturated as a functional recombinant protein for the first time and shown to exclusively inhibit the vacuolar invertase. On the other hand the apoplastic isoform Nt CIF inhibits the enzymes of both compartments. Chimeric proteins consisting of the combined halves of both inhibitors showed the significance of the N terminus for enzyme specificity. A relatively short deletion in this region therefore inactivates Nt VIF, whereas a C terminal deletion of more than 18 amino acids is tolerated. Mutation of one of the four strongly conserved cystein residues inactivates the protein completely. Treatment with reducing agents has the same effect confirming the postulated importance of disulfid bridges for structural integrity. Paradoxically, many inactive recombinant inhibitors increase invertase activity. This can be interpreted as an artefact of denatured protein leading to inactivation of endogenous inhibitors through aggregation. Although Northern Blot studies show specific transcription of the endogenous inhibitor Nt VIF, the protein could not be detected so far. This could be due to expression limited to special cell types.
