Abstract

Die vorliegende Arbeit hatte das Ziel, Plasmamembranproteine zu finden, die spezifisch für die kutane T-Zell Lymphome (CTCL), Sézary Syndrom (SzS) und Mycosis fungoides (MF), sind. Zu ihrer Identifizierung wurde zunächst über differentielle Zentrifugation ein Oberflächenpräparat der CTCL-Zelllinie HuT78 hergestellt. Es diente als Antigengemisch für die Immunisierung von Kaninchen. Die gewonnenen polyvalenten Antiseren reagierten in ELISA- und Western Blot Analysen spezifisch mit Proteinen des Oberflächenpräparats. Ferner zeigte eine immunzytochemische Analyse, dass sie die Oberfläche nativer HuT78-Zellen erkannten. Für die eigentliche Identifizierung von Plasmamembranproteinen wurden die Kaninchen-Antiseren zunächst in der Immunpräzipitation eingesetzt. Dabei wurde unter den präzipitierten Proteinen auch das Vorläuferprotein des plasmamembranständigen ICAM-1 identifiziert. Ihren zweiten Einsatz hatten die Kaninchenseren in einer serologischen Analyse, bei der eine cDNA-Phagenbank der CTCL-Zelllinie SeAx nach reaktiven Klonen durchsucht wurde. Hierbei wurden neben Plasmamembran- auch viele Nicht-Plasmamembranproteine identifiziert. Aufgrund ihrer Lokalisation und Funktion bei diversen Tumoren wurden aus 42 identifizierten Proteinen fünf für eingehendere Expressions-Analysen ausgewählt: Ku86, ein Mitglied eines DNA-Reparatur-Komplexes; hZimp10, ein Ko-Aktivator des Androgen-Rezeptors; FOXM1, ein in Embryonalgewebe ubiquitär auftretender Transkriptionsfaktor; MT5-MMP, eine im Abbau von extrazellulärer Matrix involvierte Proteinase und das Adhäsionsprotein Integrin alpha 4. Western Blot Analysen zeigten, dass Ku86 bei HuT78-Zellen auch Teil von „Lipid rafts“, cholesterinreichen Membrandomänen mit signalübertragenden Proteinen, sein kann. Real-time PCR Analysen zielten darauf ab, die Expression von hZimp10, FOXM1, MT5-MMP und Integrin alpha 4 in Haut- und PBMC-Proben von CTCL-Patienten mit entsprechenden Kontroll-Proben zu vergleichen. Dabei wurde eine gegenüber Normalzellen erniedrigte Transkripthöhe von hZimp10 bei CTCL beobachtet. Desweiteren konnte eine Expression von FOXM1 ausschließlich in Proben von SzS-Patienten gezeigt werden. Im Vergleich zu Nicht-Tumorproben wurden in CTCL-Proben für Integrin alpha 4 gleiche Transkripthöhen und für MT5-MMP in den Hautproben der MF-Patienten erniedrigte Transkripthöhen bestimmt. Auf Proteinebene wurde darüber hinaus für das kutane T-Zell Lymphom ein von Normalzellen abweichendes Reifungsmuster von MT5-MMP beobachtet. Vor allem waren die löslichen, aktiven Formen der Matrix Metalloproteinase in niedrigeren Mengen vorhanden bzw. gänzlich abwesend in den CTCL-Proben. Aus der Arbeit resultierend, könnten eingehendere „Lipid raft“-Analysen von CTCL-Zellen zum Auffinden von tumorspezifischen Plasmamembranproteinen beitragen.

Translation of abstract (English)

This thesis attempted to identify specific plasma membrane proteins of the cutaneous T-cell lymphomas (CTCL), the Sézary syndrome (SzS) and mycosis fungoides (MF), which could serve as targets for therapeutic antibodies. Therefore, a plasma membrane enriched fraction of CTCL cell line HuT78 was achieved by differential centrifugation which was then used for immunization of rabbits. Reactivity of the antisera was checked in an ELISA-system as well as by Western blot, proving reactivity against the plasma membrane fraction. Furthermore, immunocytochemical analyses could show that the rabbit sera reacted against the plasma membrane of native HuT78 cells. For the identification of plasma membrane proteins the antisera were first applied in immunoprecipitation. The identification of precipitated proteins revealed the precursor of the plasma membrane protein ICAM-1. Secondly, a serological screening of a CTCL cell line cDNA library by using the rabbit antisera was performed by a modified SEREX method. This resulted in plasma membrane and numerous non-plasma membrane proteins. According to literature researches, 5 of 42 different proteins were preselected due to their localization and function in various tumor types for further expression analyses: Ku86, a member of a DNA-repair complex; hZimp10, a co-activator of androgen receptor; FOXM1, a transcription factor ubiquitously expressed in embryonic tissue; MT5-MMP, a proteinase responsible for degradation of extracellular matrix and the adhesion protein Integrin alpha 4. Lipid rafts analysis revealed a novel localization of Ku86 in these cholesterol-rich subdomains of CTCL cells, which harbor important receptors for signal transduction. Real-time PCR analyses were performed to compare the transcription levels of hZimp10, FOXM1, MT5-MMP and integrin alpha 4 in skin- and PBMC-probes of CTCL-patients with those of healthy persons and patients with benign inflammatory skin diseases. The nuclear protein hZimp10 showed a decreased expression level in CTCL, whereas transcription factor FOXM1 was found to be only expressed in tumor cells of Sézary syndrome patients. The plasma membrane protein integrin alpha 4 showed no different transcript amounts in CTCL. MT5-MMP was found to be decreased in probes of mycosis fungoides patients compared to those of healthy persons. Furthermore, by Western blotting there is evidence that MT5-MMP exhibits an aberrant maturation profile in CTCL. The results showed decreased amounts of soluble, active forms of MT5-MMP in CTCL compared the healthy controls. With regard to the results, a more detailed analysis of lipid rafts of CTCL cells could help finding tumorspecific plasma membrane proteins.
