Abstract

APRIL (engl.: a proliferation inducing ligand) und BAFF (engl.:B cell activating factor of the tumor necrosis factor (TNF) family) sind zwei eng verwandte Liganden innerhalb der TNF-Familie. Beide Liganden teilen sich zwei Rezeptoren, TACI (engl.: transmembrane activator and CAML interactor) und BCMA (engl.: B cell maturation antigen), die vor allem auf B-Zellen exprimiert werden und vor kurzem konnten Glykosaminoglykan (GAG)-Ketten von Proteoglykanen als dritter APRIL-spezifischer Bindungspartner auf Tumorzellen, B-Zellen und T-Zellen identifiziert werden. APRIL hat eine zweiseitige Rezeptorspezifität und kann daher gleichzeitig über seine Heparinbindedomäne mit negativ geladenen GAG-Ketten von Proteoglykanen und über seine TNF-Homologiedomäne (THD) mit den Rezeptoren TACI bzw. BCMA interagieren. Unstimulierte Monozyten sterben via Apoptose, während Stimuli, die deren Differenzierung zu Makrophagen oder Dendritischen Zellen induzieren, die apoptotischen Prozesse inhibieren. Diese Arbeit zeigt erstmals eine kritische Rolle von APRIL für das Überleben und möglicherweise für die Differenzierung primärer humaner Monozyten. Monozyten binden APRIL nach einem Tag in Kultur und in vitro aus Monozyten differenzierte DCs zeigen eine starke APRIL-Bindung, die nach Aktivierung der DCs mit LPS bzw. CD40L weiter ansteigt. Die Bindung korreliert mit der Expression von negativ geladenen GAG-Ketten, da Behandlung dieser Zellen mit Heparitinase oder Chlorat (ein genereller Inhibitor der Sulfatierung) die APRIL-Bindung stark reduziert. Sulfatierte Glykane (Heparin, Heparansulfat/HS) inhibieren die APRIL-Bindung, durch Kompetition mit der APRIL-Bindung an zelluläre HS-Ketten. Im Rahmen dieser Arbeit konnte gezeigt werden, dass immobilisiertes APRIL, vor allem im Komplex mit BCMA Adhäsion von Monozyten auslöst. Dieses Adhäsionssignal wird durch Bindung von APRIL an GAG-Ketten auf der Oberfläche dieser Zellen induziert, da das Adhäsionssignal der Heparinbindedomän-Mutante von APRIL stark reduziert war. Die morphologischen Veränderungen implizieren eine Rolle von APRIL bei der Differenzierung von Monozyten zu Makrophagen. Im zweiten Teil dieser Arbeit wurden GAG-Ketten von Proteoglykanen als ein weiterer Bindungspartner von TACI identifiziert. Durch Untersuchung der bekannten Kristallstruktur und der linearen Sequenz von TACI konnten basische Motive identifiziert werden, die möglicherweise die Bindung von TACI an GAG-Ketten von Proteoglykanen vermitteln. Die hier durchgeführte Studie zeigt erstmals, dass GAG-Ketten von Proteoglykanen einen Interaktionspartner von TACI auf Monozyten und Dendritischen Zellen darstellen. TACI induziert Adhäsion von Monozyten, allerdings führt das durch TACI vermittelte Adhäsionssignal interessanterweise zu deren Tod. Proteoglykane werden von den meisten Zellen kontinuierlich in den Kulturüberstand sezerniert. Rekombinant hergestelltes TACI und aus Monozyten-Kulturüberstand aufgereinigtes APRIL haben Heparansulfat-Proteoglykane (HSPGs) gebunden und sind daher möglicherweise auch in vivo mit HSPGs komplexiert. Die Ergebnisse dieser Arbeit zeigen, dass APRIL über seine Heparinbindedomäne Proteoglykane auf extravasierten Monozyten bindet und dadurch Zell-Zell-Anlagerung von TACI- und/oder BCMA-exprimierenden Lymphozyten bewirken kann. APRIL induziert wesentliche Überlebens- und Differenzierungssignale sowohl in APRIL präsentierenden Monozyten als auch in TACI- bzw. BCMA-exprimierenden Zellen. Die in dieser Arbeit erzielten Ergebnisse bedeuten, dass neben dem B-Zellsystem auch die Funktion von Monozyten und Dendritischen Zellen entscheidend durch die Interaktionen von APRIL mit dessen Rezeptoren beeinflusst wird.

Translation of abstract (English)

APRIL (a proliferation inducing ligand) and BAFF (B cell activating factor of the tumor necrosis factor (TNF)-family) are two closely related members of the TNF-family. Both ligands share two receptors namely TACI (transmembrane activator and CAML interactor) and BCMA (B cell maturation antigen) which are predominantely expressed on B-cells. Recently glycosaminoglycan (GAG) side chains of proteoglycans have been identified as a third APRIL-specific binding partner on tumor cells, B-cells and T-cells. APRIL has a two-sided receptor specificity and can both interact with TACI and BCMA via its TNF-homology domain (THD) and bind negatively charged glycosaminoglycan chains of proteoglycans via its heparin-binding domain. Unstimulated monocytes die in vitro via apoptosis unless their differentiation to macrophages and dendritic cells is induced by various known and yet to be identified factors. We show for the first time that APRIL has a critical role in inducing a survival and probably a differentiation signal in monocytes. Monocytes start to bind APRIL after one day in culture and binding of APRIL increases in in vitro differentiated dendritic cells after stimulation with CD40L or LPS. This binding correlates with the appearance of negatively charged GAG-chains as treatment of these cells with heparitinase or chlorate (a general inhibitor of sulfation) strongly reduces APRIL binding. Sulfated glycans (heparin, heparansulfate/HS) inhibit binding of APRIL by competing with the binding of APRIL to cellular HS-chains. This thesis shows that immobilized APRIL, particularly when complexed with BCMA, induces adhesion of monocytes by binding to GAG-chains on the surface of these cells. The heparin-binding mutant of APRIL has a strongly reduced adhesion capacity. These morphological changes implicate a role for APRIL in the differentiation process of monocytes to macrophages. In the second part of this work GAG-chains of proteoglycans could be identified as a binding-partner for TACI. Analysis of the known crystal-structure and the linear amino acid sequence of TACI revealed basic motifs that possibly mediate binding of TACI to negatively charged GAG-chains of proteoglycans. Our study shows for the first time, that GAG-chains of proteoglycans are a binding-partner of TACI on monocytes and dendritic cells. TACI induces adhesion of monocytes. However this adhesion signal mediates death. Most cells constitutively shed proteoglycans into the culture medium. Recombinant TACI and APRIL that was purified from monocyte culture supernatant copurify with heparansulfate-proteoglycans (HSPGs). This implicates that APRIL and TACI may also be found complexed with HSPGs in vivo. We propose that APRIL binds to proteoglycans on extravasated monocytes via its heparin-binding domain and selectively triggers cell-cell-adhesion of lymphocytes which express TACI and/or BCMA. APRIL possibly induces a survival- and differentiation-signal into both the APRIL presenting monocytes as well as into TACI and BCMA expressing cells. The results of this work show, that in addition to the B-cell system the function of monocytes and dendritic cells is critically affected by the interaction of APRIL with its respective receptors.
