Abstract

Etwa 1 % aller Lebendgeburten leiden unter angeborenen Herzfehlern, von denen viele untherapiert zum Tod durch Herzinsuffizienz führen. Um die Früherkennung von angeborenen Herzfehlern zu erleichtern und Therapieansätze zu finden, wurden Tiermodelle entwickelt, die es ermöglichen, angeborene Herzfehler detailliert zu erforschen. Weiches herzm245 (whz) ist eine rezessive, embryonal letale Zebrafischmutante, die im Rahmen der funktionellen Genomanalyse durch ENUMutagenese erzeugt wurde. Homozygot-mutante whz-Embryonen entwickeln im Verlauf ihrer Herzentwicklung eine kontraktile Dysfunktion, die zum vollständigen Verlust der Blutzirkulation und damit zum embryonalen Tod führt. Durch histologische und molekularbiologische Untersuchungen von mutanten whz-Embryonen wurde in der vorliegenden Arbeit gezeigt, dass die Entwicklung des atrioventrikulären Kanals (AVC) bereits im frühesten durch molekulare Marker nachweisbaren Entwicklungsstadium inhibiert ist. So entwickeln mutante whz- Embryonen weder AV-Klappen, noch ein spezialisiertes AV-Überleitungssystem, das im Wildtyp (WT) die sequentielle Kontraktion von Atrium und Ventrikel steuert. Die positionelle Klonierung ergab, dass der whz-Phänotyp durch einen Gendefekt in tbx20, einem Tbox-Transkriptionsfaktor, hervorgerufen wird, der zum partiellen Verlust der transkriptorischen Aktivatorfunktion führt. Die loss of function- Mutation in tbx20 wurde durch einen genetischen rescue von whz durch WT-tbx20 bestätigt. Die durchgeführten Analysen kamen zu dem Schluss, dass im Zebrafisch die Entwicklung des AVC durch die synergistische transkriptorische Aktivität von Tbx20 mit Nkx2.5, Gata5 und Tbx5 reguliert wird und dass bmp4 ein direktes Zielgen von Tbx20 darstellt. Des Weiteren wurde im Rahmen dieser Arbeit im Zebrafisch eine zweite Isoform von Tbx20, Tbx20b, isoliert, die nicht die Entwicklung des AVCs oder des Herzens, sondern die Angiogenese reguliert. Darüber hinaus wurde festgestellt, dass der FGF-Signalweg sowohl die morphologische als auch die funktionelle Entwicklung des AVCs reguliert und dort möglicherweise durch Tbx20 induziert wird.

Translation of abstract (English)

Nearly 1% of live births suffer from congenital heart diseases, frequently leading to lethal heart failure in untreated patients. To allow early diagnosis of congenital heart diseases and the development of new therapeutical approaches, animal models have been developed allowing accurate investigations of cardiac disorders. Weiches herzm245 (whz) represents a recessive, embryonic lethal zebrafish heart mutant, generated by ENU-mutagenesis in the setting of functional genome analysis. Hearts of homozygous mutant whz embryos develop early contractile dysfunction resulting in the total abolition of blood circulation and consequently leading to embryonic death. In this thesis, the complete loss of atrio-ventricular canal (AVC) development in mutant whz embryos was demonstrated by both histomorphological and molecular analyses. It was shown that whz embryos fail to develop atrio-ventricular valves and an appropriate atrio-ventricular conduction system, responsible for sequential contractions of atrium and ventricle under physiological conditions. It was demonstrated by positional cloning that a “loss of function” mutation in tbx20, a member of the Tbx20 transcription factor family, causes the characteristic whz phenotype due to the loss of the transcriptional activator function. Accordingly, homozygous mutant whz embryos could be genetically rescued by transient ectopic wildtype-tbx20 expression. Several analyses revealed that synergistic functions of Tbx20 with Nkx2.5, Gata5 and Tbx5, respectively, regulate the development of the AVC in zebrafish and that Tbx20 is a direct upstream regulator of bmp4. A second Tbx20 isoform, Tbx20b, lacking transcriptional domains was isolated and functionally analysed. In contrast to full Tbx20, this isoform plays an important role in the regulation of angiogenesis rather than in the development of the AVC. Furthermore, it was shown that the FGF pathway is linked to atrio-ventricular development, probably induced by Tbx20. In conclusion, Tbx20 activation is an early event in atrio-ventricular valve development as well as in the formation of a physiological atrio-ventricular conduction system since its loss of function leads to the total failure of these heart structures resulting in early cardiac death.
