Abstract

In der vorliegenden Arbeit wurde die Interaktion der proteasomalen Untereinheit alpha4 mit Parkin untersucht. Frühere Untersuchungen in einem Yeast-Two-Hybrid-Screen mit einer humanen fötalen cDNA-Hirn-Bibliothek hatten Hinweise auf eine solche Wechselwirkung ergeben. Um die Bindung zwischen Parkin und der alpha4-Untereinheit zu verifizieren, und um erste Hinweise auf die mögliche Bindungsdomäne in Parkin zu erhalten, wurde ein weiterer Yeast-Two-Hybrid-Assay mit verschiedenen Parkin-Konstrukten, inklusive mutierten Proteinen von Parkin, durchgeführt. Dieser Assay bestätigte die Interaktion zwischen Parkin und alpha4, konnte jedoch keine Hinweise auf die Bindungsdomäne liefern, da sämtliche C-terminalen Konstrukte in einem Vortest (Reporteraktivitätsassay) als autoaktivierend reagierten und somit nicht bei der anschließenden Interaktions Mapping-Analyse eingesetzt werden konnten. Als weitere Bindungsstudien wurden Co-Lokalisationsstudien, Co-Immunpräzipitationen. Pull Down-Experimente und Gelfiltrationen durchgeführt. Die Ergebnisse der übrigen Versuche ließen folgende Rückschlüsse bezüglich der Interaktion zwischen der alpha4-Untereinheit des 26S Proteasoms und Parkin zu: Zum einen konnte anhand der Ergebnisse ein Bindungsbereich eingegrenzt werden. Eine Bindung von alpha4 mit dem amino-terminalen Bereich von Parkin (Aminosäuren 1-191) kann ausgeschlossen werden. Dagegen wurde aber immer mit einem der carboxy-terminalen Fragmente von Parkin (Aminosäuren 192-465) eine Interaktion mit der alpha4-Untereinheit nachgewiesen. Dies lässt darauf schließen, dass die Bindungsdomäne in diesem carboxy-terminalen Bereich von Parkin liegt. Zum anderen zeigten die Ergebnisse, dass weder die Mutation in der Ring 1-Domäne (Mutante C289) noch in der Ring 2-Domäne (Mutante C418R) die Interaktion aufzuheben vermag. Die Ergebnisse der verschiedenen Experimente für die einzelnen Parkin-Konstrukte waren teilweise unterschiedlich. Daher ist anzunehmen, dass sie Interaktion sehr schwach und/oder transient ist. Die Interaktion von alpha4 mit Parkin ist ein bisher unbekanntes Phänomen, da bisher nur Interaktionen bekannt waren, bei denen Parkin mit Untereinheiten der 19S-Komplexe (Rpn10, Rpt6, Rpn1 oder Rpn2) des 26S Proteasoms interagiert. Die Untereinheit alpha4 gehört aber zu den Untereinheiten des 20S-Kernkompexes des 26S Proteasoms. Im Unterschied zu den Interaktionen mit Untereinheiten des 19S Komplexes, erfolgt hier die Bindung nicht am N-Terminus von Parkin, sondern an seinem C-Terminus. Zudem ist alpha4 kein Substrat für die Parkin-abhängige Ubiquitylierung. Demzufolge könnten mögliche Funktionen dieser Interaktion Substrat-Präsentation an das 26S Proteasom sein, oder Regulierung der proteasomalen Aktivität.

Translation of abstract (English)

In this work the interaction between the proteasomal subunit alpha4 and Parkin was analysed. Previous analysis in a Yeast-Two-Hybrid Screen with an human fetal cDNA brain library suggested such an interaction. To verify the interaction between alpha4 and Parkin, and to get first hints on the potential binding region in Parkin, an additional Yeast-Two-Hybrid assay was performed using different Parkin constructs, including mutated proteins of Parkin. The assay indeed approved the interaction between Parkin and the proteasomal subunit alpha4, but did not revealed any hints on the binding domain since all C-terminal constructs of Parkin were shown to be autoactivating in a preceding reporter activity assay. Therefore those C-terminal constructs could not be set in the following interaction mapping analysis. For other binding studies, co-localisations, co-immunoprecipitations, Pull Down assays and gel filtration were performed. The results of the other experiments allowed the following conclusions in terms of the interaction between the proteasomal subunit alpha4 and Parkin: On the basis of these results the position of the binding domain was narrowed down: The binding of alpha4 with the amino-terminal domain of Parkin (amino acids 1-191) was ruled out. In contrast to this, using the carboxy-terminal constructs of Parkin (amino acids 192-465) an interaction with the alpha4 subunit was consistently shown. This led to the conclusion that the binding domain must be located in the carboxy-terminal part of Parkin. Moreover the results showed that neither the mutation in the domain of ring 1 (mutant C289) nor the mutation in ring 2 domain (mutant C418R) was able to abolish the interaction. The results of the experiments using the different Parkin constructs varied slightly. Thus it is assumed that the interaction is very weak and/or transient. So far the interaction of alpha4 with Parkin is a unknown phenomenon, because until now the only interactions known for Parkin are with subunits of the 19S cap complex of the 26S proteasome (Rpn10, Rpt6, Rpn1 or Rpn2). However, the subunit alpha4 belongs to the 20S core complex of the 26S proteasome. Unlike the interactions with the subunits of the 19S complex, where the binding takes place at the amino-terminus, in the case of the interaction with the alpha4 subunit the binding region is located at the carboxy-terminus of Parkin. Alpha4 is also not a substrate for Parkin-dependent ubiquitylation. Putative functions of the interaction might therefore be substrate presentation to the 26S proteasome or regulation of proteasomal activity.
