Stefan Knoth Titel: Regulation der Genexpression bei der erythroiden Differenzierung Betreuer: Prof. Dr. Stefan Wölfl Zusammenfassung Viele Erkrankungen des blutbildenden Systems, insbesondere Leukämien, haben ihre Ursache in einer Störung der Differenzierungvorgänge, die ausgehend von undifferenzierten Stammzellen des Knochenmarks zu allen Arten von hochspezialisierten Blutzellen führen. Es ist bekannt, dass sich bei der Chronischen Myeloischen Leukämie (CML) mit verschiedenen Substanzen wie AraC oder Butyrat die Proliferation unterdrücken und eine weitere Differenzierung induzieren lässt. Bei CML-Zellen geht man davon aus, dass durch die genannten Chemikalien die Differenzierung in Richtung der erythroiden Reihe stimuliert wird, da verstärkt Hämoglobine synthetisiert werden. Wegen ihrer einfachen Handhabung in Kultur wurden diese Zellsysteme in der Vergangenheit häufig als Modelle zur Untersuchung der erythroiden Differenzierung verwendet. Die therapeutikastimulierte erythroide Differenzierung von CML-Zellen ist also in zweierlei Hinsicht interessant, einerseits aus medizinischer Sicht zur Bewertung der Vorgänge im behandelten Patienten und andererseits hinsichtlich der Eignung dieser Systeme als Modelle zur Erforschung der Erythropoese. In der vorliegenden Arbeit wurden Veränderungen des Transkriptoms als Maß für Differenzierungsprozesse herangezogen. Die Entwicklung von Erythrozyten aus Vorläuferzellen wurde in vier Zellkultursystemen untersucht, die als Modelle für diesen Differenzierungsweg allgemein anerkannt sind. Als Modell für die normale Erythropoese dienten CD34-positive hämatopoetische Vorläuferzellen, die mit Erythropoietin stimuliert wurden. Die drei anderen Modelle basieren auf der CML-Zelllinie K562, wobei die Zellen mit AraC, Natriumbutyrat oder Hämin behandelt wurden. Bei allen Modellen wurden Zellproben in Form von Zeitreihen entnommen. Mit Hilfe der aus den Zellen präparierten total-RNA wurden Hybridisierungsproben hergestellt und auf AFFYMETRIX-U133A-Microarrays hybridisiert. Alle untersuchten Zeitpunkte wurden definierten Stadien der Erythropoese zugeordnet. Dazu wurden FACS-Daten bekannter Oberflächenmarker, morphologische Kriterien sowie Array-Daten einer Vielzahl weiterer erythrozytenspezifischer Gene benutzt. Die überwiegende Mehrheit der Zellen im CD34+-Experiment erreichte die letzten kernhaltigen Stadien Proerythroblast, Erythroblast und Normoblast, vollendete aber die erythroide Differenzierung nicht. Bei den K562-Experimenten ermöglichte der Vergleich der kompletten Datensätze mit den Datensätzen des CD34+-Experiments durch hierarchisches Clusterns ebenfalls eine Zuordnung zu definierten Stadien der Erythropoese. Anschließend wurden die Datensätze nach differentiell exprimierten Genen durchsucht. Im Ergebnis wurden bei den EPO-stimulierten CD34+-Zellen 2363 Gene, bei den AraC-stimulierten K562 1987 Gene, bei den Butyrat-stimulierten K562 1941 Gene und bei den Hämin-stimulierten K562 850 Gene als differentiell exprimiert eingestuft. Um festzustellen, welche Gene in allen oder zumindest mehreren Modellen reguliert werden, wurden die Schnittmengen berechnet. Wegen der geringen Übereinstimmung der Modelle und unter der Annahme, dass das CD34+-Modell die in-vivo-Bedingungen am besten repräsentiert, wurde festgestellt, dass die therapeutikastimulierten K562 als Modelle der Erythropoese ungeeignet sind. Daher wurde im Weiteren nur das CD34+-Modell untersucht. Mittels k-means-clustering wurden regulierte Gene mit ähnlichen Expressionsverläufen innerhalb einer Zeitreihe zu Gruppen (Cluster) geordnet. Dadurch konnten verschiedene Aktivierungsmuster unterschieden werden. Innerhalb der Cluster wurde nach Genen gesucht, deren Produkte an der biochemischen Signaltransduktion beteiligt sind und in einer funktionellen Beziehung zueinander stehen. Die Bedeutung dieser Veränderungen für die regulatorischen und metabolischen Reaktionswege in der Zelle wurde erörtert. Die therapeutikainduzierte Differenzierung von K562 ist zwar kein ideales Modell der Erythropoese, ist aber von großem Wert bei der Beschreibung der biologischen Vorgänge, die bei einer Chemotherapie im Patienten stattfinden. In diesem Zusammenhang wurde konkret nach regulatorischen Proteinen gesucht, deren Expressionsmuster sich in den K562-Modellen deutlich vom CD34+-Modell unterscheiden und die daher neben BCR-ABL für die Ausprägung des CML-Phänotyps wichtig sein könnten. Es wurde eine Auswahl von 18 Genen vorgestellt und für 14 von ihnen wurden die Array-Daten durch quantitative RT-PCR bestätigt. Mit Hilfe eines noch in Entwicklung befindlichen Microarrays mit phosphorylierungs-spezifischen Antikörpern war am Modell der CD34+-Zellen die Signalkaskade ausgehend von der Stimulierung mit Erythropoietin (EPO) zu verfolgen. Es wurde gezeigt, dass durch EPO die Phosphorylierung von AKT und p70S6-Kinase ausgelöst wird. Diese Ergebnisse konnten durch Immunoblots bestätigt werden.

Stefan Knoth Titel: Regulation of Gene Expression during Erythroide Differentiation Superviser: Prof. Dr. Stefan Wölfl Many diseases of the hematopoietic system, especially leukemias, originate from a dysfunction of differentiation processes. During these differentiation processes primitive hematopoietic stem cells develop into mature blood cells of all lineages. The chronic myeloic leukemia (CML) in blast crisis is characterized by the appearance of immature blast cells which no longer differentiate. Moreover it is known that cytostatic substances as cytosine arabinoside or butyrate induce in blast cells a resuming of the aborted differentiation beyond their antiproliferativ action. In CML the mentioned chemicals are believed to trigger differentiation into the erythroid lineage because of the raised synthesis of hemoglobins. So the chemical induced erythroid differentiation of CML cells is interesting for two reasons. First the action of the drugs on malign cells within a treated patient is simulated. Second the applicability of these systems as models for the differentiation of erythrocytes still has to be confirmed. Up to now little is known about the details of chemical induced red blood cell differentiation and its comparability with normal erythropoiesis. Moreover the molecular basics of normal erythropoiesis are characterized incompletely as well. The development of erythrocytes from progenitor cells was examined in four cell culture systems which are generally accepted as models of red blood cell differentiation. CD34+ hematopoietic progenitors stimulated with erythropoietin have been used as model for the normal, healthy erythropoiesis. Three other models base upon the CML cell line K562 which was stimulated with cytosine arabinoside, butyrate or hemin. In all four models cell probes were taken as time courses. The total RNA was prepared and hybridization probes were synthesized and hybridized upon AFFYMETRIX U133 A microarrays. The examined points in time of all models were assigned to defined stadiums of erythropoiesis. For this purpose the expression of erythroid markers was analyzed by cytometry and retrieved from the microarray data. Additionally morphological data and hierarchic clustering were used. The majority of CD34+-cells reached the last nucleated stadiums proerythroblast, erythroblast and normoblast but did not complete the differentiation. In case of all three K562-experiments the comparison of the complete data sets with the datasets of the CD34+-experiment by hierarchic clustering enabled also an assignment. After that the data sets of all experiments were searched for differential expressed genes. In the CD34+-experiment 2363 genes were identified. In K562 stimulated by cytosine arabinoside 1987 genes were discovered, in butyrat-stimulated K562 1941 genes and in hemin-stimulated K562 850 genes. All possible intersections of the differential expressed genes of all experiments were calculated to identify those genes which were differentially expressed in more than one model. Surprisingly only 66 genes were regulated in all models. Because of the assumption that the CD34+-model mimics the in-vivo-conditions best, the further work was focused on EPO-stimulated CD34+-cells. Genes with similar expression profiles were clustered by k-means-clustering. Within the clusters rather then in similarly shaped clusters was searched for genes whose products are involved in functional related biological processes. This search based on the assumption that similar expression profiles are caused by similar regulatory mechanisms. The meaning of these changes for the regulatory and metabolic pathways of the cells was discussed. Genes were identified whose products probably play a central role for the conversion of the extracellular stimuli into the cell response. A focus lay on the identification of differentially expressed transcription factors and their patterns of activation because cooperating scientists planned to model transcriptional networks with this information. Besides the bad applicability of the chemical induced differentiation of K562 as models for erythropoiesis these systems are useful to investigate the biological processes in patients treated with these drugs. In this context was searched for genes of regulatory proteins whose expression profiles were well distinguishable in K562- and CD34+-experiments. Besides of BCR-ABL such proteins could be important for the establishment and maintenance of the phenotype CML. A choice of 18 genes was presented whereas for 14 of them the array data were confirmed by quantitative RT-PCR. Changes of the phosphorylation of meaningful signaling proteins in erythropoietin stimulated CD34+-cells were analyzed by the use of a novel protein microarray. This array has phosphorylation-specific antibodies as probe molecules. It is shown that erythropoietin triggers the phosphorylation of AKT and p70S6K. These results were confirmed by immunoblots.
