Abstract

Die vorliegende Arbeit befasst sich mit der selektiven Akkumulation von T-regulatorischen Zellen (Tregs) im humanen Pankreaskarzinomgewebe durch ihre Adhäsionsmolekül-Expression und dem daraus resultierenden Mikromilieu, welches die Funktion zytotoxischer T-Zellen im Tumorgewebe supprimiert. In dieser Arbeit konnte gezeigt werden, dass es zu einer selektiven Akkumulation von Tregs im Tumorgewebe kommt. Diese erschwerte wahrscheinlich durch Zell-Zellinteraktionen und Ausschüttung von TGF-ß die Erkennung und Zerstörung von Tumorzellen durch tumorinfiltrierende zytotoxische T-Zellen. Zusätzlich konnte bewiesen werden, dass es zu einer gesteigerten Vaskularisierung im Tumorgewebe kam, welche der besseren Versorgung von hoch-proliferativen Tumorzellen diente. Weitere Untersuchungen bewiesen die Tumormilieu-induzierte Modulation von endothelialen- und Treg- CAM-Expressionsmustern, welche in vitro und in vivo zu selektiv verstärkter Adhäsion und Transmigration von Tregs durch Tumorendothel führte. Die Treg selektive Tumorinfiltration wurde in vitro und in vivo unter anderem durch Bindung von CD62L, ß-7 Integrin und CD166 an ihre tumorendothelial exprimierten Bindungspartner MAdCAM-1, VCAM-1, CD62-E und CD166 vermittelt. Die spezifische Blockade dieser Proteinbindungen mittels Antikörpern inhibierte in vitro und in vivo eine Treg-selektive Migration durch Tumorendothel und inhibierte nur schwach die Transmigration durch Kontrollendothel. Die notwendigen inhibitorischen Treg-Effekte in nicht-pathologischen Geweben würden somit bei einer Treg selektiven Migrationsblockade erhalten bleiben. Dies könnte zur Steigerung der zytotoxischen Funktionalität im Tumor führen, ohne eine Auto-Immunität zu induzieren.

Translation of abstract (English)

This work defines selective accumulation of T regulatory cells (Tregs) in human pancreatic carcinoma tissue due to expression of adhesion molecules during Treg migration, establishing a tumour microenvironment which suppresses cytotoxic T cell function against tumour growth. This work demonstrated selective accumulation of Tregs in tumour tissue. This Treg infiltration impeded recognition and destruction of tumour cells by cytotoxic T cells, using mechanisms of cell-cell interaction and/or secretion of TGF-ß. We also showed that there was an increased vascularization in tumour tissue which lead to a nutritional environment supporting highly proliferative tumour cells. In addition, in vitro and in vivo, we showed modulation of Treg and endothelial cell adhesion molecule expression patterns in response to the tumour milieu, leading to selective and increased adhesion and trans-migration of Tregs through tumour endothelium. We showed that the selective infiltration of Tregs into tumour tissues is mediated by the binding of Treg-expressed CD62L, ß7-integrin and CD166 to the tumour endothelial-expressed binding partners MAdCAM-1, VCAM-1, CD62-E and CD166. The in vitro and in vivo blocking of these binding interactions by monoclonal antibody lead to largely selective inhibition of Treg migration through tumour-endothelium but not through nonmalignant endothelium. This forms the basis for increased cytotoxic T cell function in tumour tissue while avoiding the major problem of concomitant activation of autoimmunity in nonmalignant tissue.
