Abstract

Die Erforschung der Chromatinstruktur in Interphasezellkernen basiert im Wesentlichen auf der in situ Hybridisierung von fluoreszenzmarkierten Sonden (=FISH), lichtoptischen Mikroskopieverfahren und digitaler Bildanalyse. Für die Quantifizierung der Bildinformation wurden neue effiziente Algorithmen entwickelt und anschließend auf verschiedene biologische Systeme angewendet. Zur quantitativen Analyse der Topologie und Morphologie von spezifisch fluoreszenzmarkierten Chromatinregionen wurde ein Softwarepaket entwickelt, welches folgende Parameter mißt: absolute und relative Positionen, normierte und absolute Distanzen, Schwerpunktswinkel von Chromatinregionen und die Minkowskifunktionale von Chromosomenterritorien. Die Anpassung eines Ellipsoidmodells an Zellkerne wurde mittels Lagranger Optimierung realisiert. Zusätzlich wurde ein Algorithmus entwickelt, welcher das Kugel-, Ellipsoidvolumen und die Kompaktierung von markierten Genen anhand der Halbwertsbreiten einer an die Intensitätsverteilung angepaßten zweidimensionalen Gaußverteilung bestimmt. Das Softwarepaket wurde an den Chromosomenterritorien 18 und 19 in Fibroblastenzellkernen getestet. Die Anwendung auf Zellkerne des Cervix-Gewebes erlaubte es erstmals ein vollständiges topologisches Modell des Chromosomenterritoriums 18 und des BCL2 Gens mit der Differenzierung und Expression zu verknüpfen. Das Kugel-, Ellipsoidvolumen und die Kompaktierung des SNRPN Gens wurden abgeschätzt. Es konnte aufgezeigt werden, dass herkömmliche Meßmethoden unzureichend sind, daß aber auch FISH-Artefakte das Ergebnis beeinussen können. Daher wurde eine neue Methode zur vitalen kombinatorischen Oligonukleotid-FISH (=vCF) eingeführt. Während bei allen Standard-FISH Methoden destruktive Veränderungen der Chromatinstruktur auftreten können, ermöglicht die vCF eine schonende in situ Hybridisierung in lebenden T-Lymphozyten.

Translation of abstract (English)

Investigations of chromatin structure in interphase nuclei are mainly based on in situ hybridisation of fluorescently labeled probes (=FISH), lightoptical microscopy and digital image analysis. For image quantification novel algorithms were developed and applied to several biological systems. For quantitative analysis of the topology and morphology of specific fluorescently labeled chromatin regions a software-package was developed, that measures the following parameters: absolute and relative positions, normalised and absolute distances, barycenter angles of chromatin regions and the minkowski functionals of chromosome territories. The adaptation of an ellipsoid model to all nuclei was realised by lagrange optimisation. Additionaly an algorithm was developed to determine the spherical, ellipsoidal volume and the compaction of labeled genes using a two-dimensional gaussian fit adapted the intensity distribution. The software-package was tested on chromosomes 18 and 19 in fibroblast nuclei. The application to nuclei in cervix-tissues allows for the first time to connect a complete topological model of chromosome 18 and BCL2 gene with differentiation and expression. The spherical, ellipsoidal volume and compaction of the SNRPN gene were estimated. It could be shown that conventional methods are insufficient, and that FISH artefacts may influence the result. Therefore, a novel labelling technique of vital cells using combinatorial oligo nucleotides (=vCF) was introduced. Whereas all the standard- FISH-methods possibly cause destructive changes of chromatin structure, vCF permits a gentle in situ hybridisation in living cells.
