Abstract

Die Embryonalentwicklung von Drosophila beginnt mit 13 Kernteilungsrunden in einem Synzytium. Zu Beginn von Zyklus 14 setzt die zygotische Genexpression ein und die Plasmamembran invaginiert zwischen benachbarten Kernen, was zur Bildung eines einzellschichtigen polarisierten zellulären Blastoderms führt. Dieser mit der Zytokinese verwandte Prozess wird als Zellularisierung bezeichnet. slow-as-molasses (slam) ist ein neues, in Drosophiliden konserviertes Gen ohne bekannte funktionelle Domänen, das für die Membraninvagination während der Zellularisierung notwendig ist. Slam lokalisiert an der Front der invaginierenden Membran, dem Furchungskanal (FC) und slam-defiziente Embryonen zeigen einen letalen Zellularisierungsphänotyp. Anhand hypomorpher Allele, die den Zellularisierungsphänotyp nicht zeigen, konnte eine Beteiligung an der späteren Wanderung der Keimzellen gezeigt werden während der Slam nicht exprimiert wird. Trotz der starken Expression zu Beginn der zygotischen Genexpression konnte anhand von slam-Keimbahnklonen eine essentielle maternale Kontribution nach-gewiesen werden, deren Fehlen nur teilweise zygotisch gerettet wird. In Abwesenheit von Slam ist die Lokalisation von Aktin, Nullo und Dia am frühen FC nicht betroffen. Bis auf die initiale Bildung des FC kommt es ohne Slam jedoch zu keiner Membraninvagination und Myosin lokalisiert nicht am FC. Slam rekrutiert RhoGEF2 durch direkte Interaktion mit dessen PDZ-Domäne an den FC und fördert so die lokale Akkumulation von F-Aktin. Slam kann während der gesamten Zellularisierung am FC detektiert werden. FRAP-Analysen zeigten, dass Slam in einem kurzen Zeitfenster zu Beginn der Zellularisierung am FC lokalisiert und anschließend sehr stabil an ihn bindet. slam mRNA und Protein kolokalisieren am FC und in basalen Partikeln, wobei die mRNA ein sehr ähnliches Lokalisations- und Diffusionsverhalten wie das Protein zeigt. Slam kolokalisiert zu Beginn der Zellularisierung mit Myosin10a in den basalen Partikeln sowie später am FC. Die Ergebnisse zeigen, dass slam eine Schlüsselfunktion sowohl bei der Membraninvagination als auch der Stabilisierung des FC während der Zellularisierung zukommt. Es scheint zwei getrennte Signalwege zu geben, die für die Membraninvagination essentiell sind, wovon einer von slam abhängt. Der andere Signalweg ist von Nullo abhängig. Die Initiale Festlegung der späteren Invaginationsstellen funktioniert auch in der Abwesenheit von Slam und Nullo.

Translation of abstract (English)

Development of the Drosophila embryo starts with 13 rounds of nuclear division in a syncytium. At the onset of cycle 14 zygotic gene expression sets in and the plasma membrane invaginates between adjacent nuclei to form a single-layered polarized cellular blastoderm in a process related to cytokinesis called cellularization. slow as molasses (slam) is a novel gene conserved in drosophilids with no known functional domains that is required for plasma membrane invagination during cellularization. Slam localizes to the tip of the invaginating membrane, the furrow canal (FC) and slam deficient embryos show a lethal cellularization phenotype. Using hypomorphic alleles that do not show the severe early cellularization phenotype, Slam has also been reported to play a role in germ cell migration in later development when no slam expression is detected. Despite its expression peak at the onset of zygotic gene expression, generation of slam deficient germline clones revealed a crucial maternal contribution that can only partially be rescued by zygotic expression. In the absence of slam initial accumulation of actin at the future invagination site is not affected as well as localization of Nullo and Dia. Besides the initial formation of the FC membrane invagination is completely abolished in slam deficient embryos and MyosinII does not localize to the future invagination site. Slam recruits RhoGEF2 to the FC and ectopic sites by direct interaction with the PDZ domain thus promoting local actin assembly. The Slam protein can be visualized throughout the whole process of cellularization. FRAP analyses revealed that Slam localizes to the FC in a small time window at the onset of cellularization followed by stable binding. Strikingly slam mRNA and protein colocalize at the FC and in basal particles with the mRNA showing a very similar localization and diffusion behaviour as the protein. Slam colocalizes with Myosin10A in the basal particels at the onset of cellularization and later at the FC. The results show that slam is a key regulator of cellularization having a dual function in membrane invagination and maintainance of the FC. There seem to be two separate pathways required for proper membrane invagination, one being Slam-dependent and the other being independent of Slam requiring Nullo. Initial definition of the invagination site is not affected in the absence of both Slam and Nullo.
