Abstract

Förster-Resonanzenergietransfer (FRET) ist ein Effekt, bei dem die Energie eines durch einen Laser angeregten Fluoreszenzfarbstoffs (der Donor) strahlungsfrei auf einen zweiten (den Akzeptor) übertragen wird. Da die Effizienz dieser Energieübertrag vom gegenseitigen Abstand der beiden Farbstoffe abhängt, können mit dieser Technik Distanzen im Bereich von 2 - 8 nm ermittelt werden. Markiert man zum Beispiel ein Enzym an passenden Stellen mit einem Donor- und einem Akzeptorfarbstoff, so lassen sich Konformationsänderungen des Moleküls feststellen. Führt man FRET-Experimente an einzelnen Molekülen durch, so lassen sich Subpopulationen unterschiedlicher Konformationen ermitteln. Ein Nachteil herkömmlicher Einzelmolekül-FRET-Messungen liegt darin, dass kaum unterschieden werden kann zwischen einer nur mit dem Donor markierten Probe und einer doppelt markierten, bei der die Farbstoffe zu weit auseinander liegen, als dass ein Energietransfer stattfinden kann. Dieser Missstand lässt sich beheben, indem zusätzlich zum donoranregenden Laser in alternierender Weise ein weiterer Laser dazu benutzt wird, der den Akzeptor direkt anzuregen. Dadurch kann die Existenz eines Akzeptorfarbstoffs unabhängig vom Donor festgestellt werden.

Im Rahmen dieser Arbeit wurde ein vorhandenes Einzelmolekül-FRET-Spektrometer um alternierende Laseranregung (ALEX) erweitert. Dazu war eine Steuerung zu installieren, die die Intensitäten der beiden Anregungslaser moduliert. Gleichzeitig musste im Messsignal vermerkt werden, durch welche Anregungswellenlänge ein detektiertes Photon erzeugt wurde. Für die Analyse der aufgezeichneten Daten aus ALEX-Messungen wurde ein Programm geschrieben. Dieses System wurde charakterisiert und der Einfluss unterschiedlicher Mess- und Analyseparameter auf das Ergebnis untersucht.

Translation of abstract (English)

Förster resonance energy transfer (FRET) is an effect where the energy of an excited fluorescent dye (the donor) is nonradiatively transferred to a second dye (the acceptor). Because the efficiency of this energy transfer depends on the displacement of the two dyes relative to one another, it is possible to measure distances in the range of 2 - 8 nm. For example, if the donor and the acceptor are attached to specific sites of an enzyme one can observe conformational changes of the molecule. Performing FRET experiments on single molecules allows one to detect subpopulations within an ensemble of sample molecules. A drawback of traditional single molecule FRET measurements, however, is the difficulty to distinguish samples labeled with only a donor dye from doubly labeled ones, where the fluorophores are too distant from each other to make energy transfer. This problem can be solved by adding a second excitation laser in an alternating manner which activates the acceptor directly. Thereby the existence of an acceptor dye can be probed independently of the donor.

In the context of this thesis, an existing single molecule FRET spectrometer was upgraded with alternating laser excitation (ALEX). A control system for the intensities of both excitation lasers was installed. The alternation of the laser beams had to be synchronized with the measured signal to know the excitation source for each single detected photon. A program was written to analyse the ALEX data. Finally the setup was experimentally characterized and the influence of different measurement and analysis parameters is presented.
