Abstract

Die Mitglieder der konservierten Vigilin-Proteinfamilie zeichnet sich durch ihren modularen Aufbau aus 14-15 KH-Domänen aus, die eine RNA-Bindung vermitteln können. Aufgrund von Untersuchungen in unterschiedlichen eukaryontischen Modellsystemen wurde ihnen eine Bedeutung in verschiedenen funktionellen Zusammen-hängen wie Zellaktivierung, Proteinsynthese und -Sekretion, mRNA- und tRNA-Metabolismus, Funktion von Heterochromatin sowie Teilung von ER- und Kernmembranen zugeordnet. Ziel dieser Arbeit war die Charakterisierung von Scp160p, des Vertreters der Vigilin-Familie in Saccharomyces cerevisiae. In Hefe führt der Verlust von funktionellem Scp160p irreversibel zu pleiotrophen Phänotypen wie anormalem Erscheinungsbild der Zellen, Verdopplung der chromosomalen DNA, einer verringerten Lebens-fähigkeit der Zellen und einer unregelmäßigen Verteilung genetischer Marker beim Kreuzen von Stämmen. Durch Fluoreszenzmikroskopie und Zellfraktionierung konnte nachgewiesen werden, dass in vivo der Großteil von Scp160p mit ER-Membranen interagiert. Eine kleinere Fraktion findet sich im Zytoplasma. Sowohl an der ER-Membran als auch im Zytosol assoziiert Scp160p mit Polysomen. Diese Interaktion benötigt die Anwesenheit von mRNA, ist sättigbar und in vivo spezifisch für eine Subfraktion von mRNA-Ribosomen-Komplexen. Scp160p wird tubulinabhängig vom Zytoplasma an die Membran transportiert. Nach Depolymerisation von Mikrotubuli ist die Assoziation von Scp160p mit der ER-Membran abhängig von aktiver Translation. Die beiden C-terminalen KH-Domänen von Scp160p werden zur Interaktion mit Polysomen, nicht jedoch zur Komplementation des Chromosomenverdopplungs-Phänotyps der scp160*-Mutante benötigt. Die Aminosäuren 98-142 beinhalten ein Signal, das den aktiven und Ran-abhängigen Kernimport eines 2xGFP-Reporters vermittelt. Im Gesamtprotein wird die N-terminale Kernimport-Aktivität durch Beiträge C-terminaler Domänen abgeschirmt. Unter optimalen Wachstumsbedingungen im Labor gelangt Scp160p daher nicht in den Zellkern. Mikroarray-Experimente zeigten, dass Depletion von Scp160p zu charakteristischen Veränderungen im Transkriptom und zu einer Verschiebung spezifischer mRNA-Signale zwischen einer membranangereicherten und einer zytosolischen Fraktion führt. Diese Veränderungen betreffen insbesondere mRNAs, die für Proteine kodieren, die in den sekretorischen Weg eingehen. Die Ergebnisse der Mikroarray-Experimente geben im Zusammenhang mit der bevorzugten Assoziation von Scp160p mit membrangebundenen Polysomen Hinweise darauf, dass Scp160p als Effizienzfaktor eine Rolle bei der Translation einer definierten Gruppe sekretierter und membrangebundener Proteine spielt.

Translation of abstract (English)

The members of the conserved family of vigilin-like proteins all contain 14-15 tandemly-repeated KH-domains, which are implicated in binding to RNA. Studies in different eukaryotic model organisms proposed, that these proteins function in various cellular pathways including cell activation, protein synthesis and secretion, mRNA- and tRNA-metabolism, function of heterochromatin and the partitioning of ER- and nuclear membranes during cell division. In this work, Scp160p, the vigilin-like protein in Saccharomyces cerevisiae, was characterized. In yeast, loss of functional Scp160p leads to pleiotrophic phenotypes such as anormal cell shape, doubled DNA-content and reduced viability. Additionally, scp160*-strains show irregular segregation of genetic markers when crossed with wild-type strains. Using fluorescence microscopy and cell fractionation it was shown, that the vast majority of Scp160p interacts with ER-membranes in vivo, while a smaller fraction is found in the cytoplasm. In both fractions, Scp160p associates with polysomes. The association with polysomes is dependent on mRNA, saturable and specific for a subfraction of mRNA-ribosome-complexes. Furthermore, transport of Scp160p to the ER-membrane is dependent on a functional tubulin-cytoskeleton. After depolymerization of microtubules, the association of Scp160p with the ER-membrane is dependent on ongoing translation. The two C-terminal KH-domains of Scp160p are needed for the interaction with polysomes but not for the complementation of the DNA-doubling-phenotype of the scp160*-mutant. Amino acids 98-142 harbour a cryptic signal that mediates the active and Ran-dependent nuclear import of a 2xGFP-reporter protein. In full length Scp160p, the N-terminal nuclear import signal is masked due to the presence of the C-terminus. Thus, Scp160p is not found in the nucleus under normal growth conditions. Microarray-experiments showed, that depletion of Scp160p leads to characteristic changes in the transcriptome and to a shift of specific mRNA-signals from a membrane-enriched to a cytosolic fraction. These changes are found especially for mRNAs that code for proteins that enter the secretory pathway. Together with the predominant association of Scp160p with ER-bound polysomes, the microarray results provide strong hints for a role of Scp160p in the regulation of the translational efficiency of a defined group of secretory and membrane-bound proteins.
