Abstract

Die juvenile Form der X-chromosomal vererbten Adrenoleukodystrophie ist durch eine progressive, entzündliche Demyelinisierung der cerebralen weißen Substanz bei gleichzeitiger Nebenniereninsuffizienz gekennzeichnet. Der zugrunde liegende genetische Defekt ist die Mutation eines peroxisomalen Membranproteins unbekannter Funktion, des Adrenoleukodystrophie-Proteins (ALDP). Versuche, ein Mausmodell für die juvenile Form der Krankheit zu generieren, waren erfolglos, da neurologische Symptome ausblieben. Die Deletion des ALDP Gens führte lediglich zu biochemischen Parallen der Krankheit. Da die Krankheit beim Menschen üblicherweise nicht vor dem fünften Lebensjahr ausbricht und dann besonders zwei sehr lipidhaltige Gewebe der Patienten betroffen sind (der adrenale Cortex und das Myelin), stellten wir die Arbeitshypothese auf, dass aus der primären ALDP-Defizienz ein sekundärer Peroxisomendefekt in fettreichen Geweben resultiert. Um ein Mausmodell für die X-ALD zu erhalten, an dem sich die pathologischen Vorgänge im Nervensystem untersuchen lassen, sollte diese Hypothese im Rahmen der vorliegenden Arbeit untersucht und dazu eine Mauslinie mit defekter Peroxisomenbiogenese in myelinbildenden Zellen generiert werden. Die oligodendrozytenspezifische Peroxisomendysfunktion wurde durch Anwendung des Cre/LoxP-Systems erreicht. Hierzu wurde eine Mauslinie, bei der die Cre-Rekombinase unter Regulation des CNP-Promotors steht, mit einer Mauslinie verpaart, bei der essentielle Bereiche des peroxisomalen Biogenesefaktor Peroxin 5- (PEX5) Gens mit LoxP-Sequenzen flankiert worden waren. Anhand von Zellkulturanalysen konditionaler PEX5 null-mutanter Mäuse (CNP-Cre*floxPEX5) konnte die Expression von Cre und eine damit verbundene Defizienz von PEX5 in Oligodendrozyten nachgewiesen werden. Immuncytochemische und immunhistochemische Untersuchungen zeigten Katalase-defiziente Peroxisomen in Oligodendrozyten und im Myelin der mutanten Tiere. Massenspektrometrische Lipidanalysen des Myelins ließen den Schluss auf peroxisomale Dysfunktion zu. Homozygote konditionale PEX5-defiziente Mäuse sind lebensfähig und bis zu dem Alter von 3 bis 4 Monaten phänotypisch unauffällig. Im Alter zwischen 3 und 6 Monaten erscheinen erste neurologische Auffälligkeiten in Form einer motorischen Hinterlaufschwäche. Die Krankheit verläuft progressiv über die vollkommene Paralyse der Hinterläufe und dem Erreichen eines vegetativen Stadiums bis zum Tod. Es liegt eine progressive, inflammatorische, symmetrische Demyelinisierung und Neurodegeneration zu Grunde an der Mikroglia, Astrozyten und Leukozyten beteiligt sind. Die CNP-Cre*floxPEX5 Mauslinie liefert ein Mausmodell, das in seiner Ausprägung mit der juvenilen Form der Xchromosomal erblichen Adrenoleukodystrophie vergleichbar ist.

Translation of abstract (English)

The juvenile form of X-linked adrenoleukodystrophy (X-ALD) displays a progressive inflammatory demyelination of cerebral white matter accompanied by adrenal gland insufficiency. The disease is caused by mutations of the adrenoleukodystrophy protein (ALDP). This is a peroxisomal membrane protein of unknown function. Attempts to create a mouse model for the juvenile form of X-ALD by inactivating the ALDP gene failed, in as much as ALDP deficient mice did not exhibit CNS demyelination. These mice showed an abnormal accumulation of very long chain fatty acids (VLCFA) that is also found in human patients. In humans X-ALD appears between four to ten years of age. Two of the most lipid rich tissues (myelin and adrenal glands) are affected. We hypothesized that ALDP deficiency, as a primary defect, leads to secondary loss of additional peroxisomal functions. This second step would take place preferentially in cells of lipid-rich tissues. To test for this hypothesis we generated a mouse line with defective peroxisomal biogenesis restricted to myelin forming glia, using the Cre-LoxP-system. We intercrossed a mouse line expressing Cre recombinase under control of CNP regulatory elements with a second mouse line that harboured a “floxed” peroxisomal biogenesis factor peroxin 5 (PEX5) gene. Immunocytochemical analysis of the offspring (CNP-Cre*floxPEX5) verified oligodendrocyte-specific Cre-mediated PEX5 deficiency. Further immunocytochemical and immunohistochemical examination displayed the lack of catalase positive peroxisomes in oligodendrocytes and in myelin of these mice. Mass spectrometric lipid analysis of myelin demonstrated peroxisomal dysfunction. Homozygous conditional PEX5 deficient mice are viable and, up to 4 months of age, phenotypically normal. Between 4 to 6 months of age, first neurological abnormalities become obvious as indicated by motor-impairment of the hind limbs. The disease progresses gradually and comprises complete paralysis of the hind limbs, followed by a vegetative phase and death. The underlying cause is a progressive, inflammatory, symmetrical cerebral demyelination and neurodegeneration with involvement of microglia, reactive astrocytes, and invading leukocytes. CNP-Cre*floxPEX5 mice develop a leukodystrophy, that clinically and histologically resembles the severely progressive and inflammatory childhood form of X-ALD and thus, provide a bona fide mouse phenocopy for this neurological disease.
