Abstract

Lymphatische Neoplasien stellen eine heterogene Gruppe proliferativer Erkrankungen der Lymphozyten dar. Zwar konnten die zugrundeliegenden Pathomechanismen in einigen wenigen Fällen bereits entschlüsselt werden, doch sind sie bei der Mehrzahl der lymphatischen Leukämien und Lymphome unbekannt. So besteht auch für das bisher unheilbare Mantelzell-Lymphom (MCL) großer Bedarf an der Entschlüsselung der zugrundeliegenden molekularen Vorgänge, die letztlich zur Entstehung des Lymphoms führen. Erste Anhaltspunkte lieferte bereits die Aufdeckung der chromosomalen Translokation t(11;14)(q13;q32), welche letztendlich zu der für das MCL charakteristischen Überexpression von Cyclin D1 führt. Ferner konnte in 75 % der MCL-Fälle eine Inaktivierung der Proteinkinase ATM nachgewiesen werden. Ziel dieser Arbeit war es, durch Kombination der genannten Aberrationen im murinen System ein Modell zu schaffen, an dem die Pathomechanismen von B-Zell-Lymphomen und insbesondere von MCL untersucht werden können. Zu diesem Zweck wurden zwei unterschiedliche Ansätze gewählt. Zum einen wurden die genetischen Aberrationen durch Kreuzung eines Atm-knock-out und eines Ccnd1 transgenen Mausstammes zusammengeführt, zum anderen wurden mit Ccnd1-rekombinanten Retroviren infizierte, Atm-defiziente hämatopoetische Stammzellen zur Rekonstitution des hämatopoetischen Systems letal bestrahlter Mäuse genutzt. In keinem der Ansätze konnte nach langfristiger Beobachtung und eingehender Untersuchung die Entstehung eines B-Zell-Lymphoms festgestellt werden. Hingegen wurden Hinweise gefunden, wonach die molekularen Folgen des Atm-Verlusts dem Effekt der Ccnd1-Überexpression entgegenwirken. Darüber hinaus entwickelten alle Atm-defizienten Mäuse unabhängig von Ccnd1-Status thymische T-Zell-Lymphome. Anhand immunphänotypischer und molekulargenetischer Untersuchungen wurde die weitestgehend unverstandene Genese dieser durch Atm-Verlust verursachten Tumoren untersucht. Die neoplastischen Zellen der T-Zell-Lymphome wurden umfassend charakterisiert und konnten als nicht aktivierte, CD4 CD8 doppelt positive T-Lymphozyten identifiziert werden, die eine ausgeprägte Deregulation der entwicklungsspezifischen Oberflächenmoleküle CD25 und T-Zell-Rezeptor β (TCRβ) zeigen. Erstmalig konnten in dieser Arbeit sowohl der oligoklonale Ursprung dieser Tumorzellen als auch deren inkorrekte Rekombination des TCR β Gens nachgewiesen werden. Die Untersuchung der Atm-/- T-Zell-Lymphome mit hochauflösenden aCGH-microarrays ließ charakteristische genomische Imbalancen (Chromosom 12, 14 und 15) erkennen, von welchen der TCRα-Lokus auf Chromosom 14 grundsätzlich in sämtlichen untersuchten Proben betroffen war. Basierend auf der Analyse von Atm-/- Thymi ohne Lymphom konnten genomische Tumorvorstufen als Stadien beschrieben werden, deren mittels microarray-basierter Expressionsanalyse erstellte Transkriptionsprofile grundlegend von den Profilen genomisch unveränderter Thymi abweichen. Die weitere Auswertung der gewonnen Expressionsdaten ergab neue Erkenntnisse über die transkriptionelle Deregulation tumorassoziierter Gene, wie z. B. Tuba1 und Aurka. Die Mehrzahl der hier untersuchten T-Zell-Lymphome wies darüber hinaus einen genomischen Notch1-Zugewinn auf, der sich in der Deregulation Notch1-regulierter Gene, insbesondere c-Myc, Dtx1 und Hes1, auswirkte. Durch bioinformatorische Auswertung der gewonnenen Expressionsdaten konnte ein Netzwerk deregulierter Gene postuliert werden, welches auf eine Dysregulation der CD95 vermittelten Apoptose in der betroffenen T-Zellpopulation der Lymphome hinweist.

Translation of abstract (English)

Lymphatic neoplasias represent proliferative diseases of lymphocytes. While the pathomechanisms for some of these diseases have been extensively investigated and elucidated, for most of the lymphatic leukemias and lymphomas they are still unknown. Also for the so called mantle cell lymphoma (MCL), a high-malign and as yet incurable B-cell lymphoma, the molecular processes that underlie tumorgenesis are unclear. Nevertheless, first evidences came from two important findings several years ago. One of them specify a chromosomal translocation, t(11;14)(q13;q32), resulting in the characteristic overexpression of the cell cycle regulator protein cyclin D1. The other one, which is described for about 75% of all MCL, is the inactivation of the protein kinase ATM gene. Based on these findings the first aim of this study was to establish a mouse model system, representing both above named genetic aberrations, to investigate the pathomechanisms of B-cell-lymphomas including MCL. To this purpose two independent approaches were chosen. While one model was generated by cross-breeding an Atm knock out mouse with a cyclin D1 transgenic mouse, another model was build by transplantation of retroviral infected Atm-deficient bone marrow stem cells into lethally irradiated mice to reconstitute the haematopoietic system overexpressing cyclin D1. As a result of these breeding experiments in none of the mice a MCL was detected, however extensive analysis of the animals provided evidences that the molecular mechanisms resulting from Atm-deficiency on one hand side, and cyclin D1 overexpression on the other hand side, would counteract each other. Moreover, all Atm-deficient mice developed severe thymic T-cell lymphoma. Using immuno-phenotypic as well as molecular-genetic assays, Atm knock out mice were used to investigate essentially unknown molecular processes involved in Atm-deficiency dependent, thymic T-cell lymphoma. The experimental results led to the identification of neoplastic cells as CD4 CD8 double positive, non-activated T-lymphocytes, characterized by a severe deregulation of T-cell development specific cell surface markers, CD25 and T-cell receptor β (TcRβ). Indeed, for the first time these abnormal T-cells were shown to be of oligoclonal origin as well to be severely impaired in TcRβ gene recombination. Furthermore, high resolution array-based comparative genome analysis (aCGH) of Atm-deficient T-cell lymphoma revealed distinct chromosomal imbalances (chromosome 12, 14 and 15). Of these imbalances the one concerning the TcRα genetic locus on chromosome 14, was basically found in all samples examined. Based on the analysis of Atm-deficient mice that did not yet develop thymic lymphoma by aCGH, 5 genomic pre-tumoral stages could be defined. Using microarray-based expression profiling analysis, major differences were identified in expression profiles of these Atm-deficient non-lymphoma cells. Further analysis of gene expression data led to the detection of transcriptional deregulated, tumor-associated genes, as for example Tuba1 and Aurka. Moreover a vast majority of analysed T-cell lymphoma cell samples showed an additional genomic amplification of Notch1 that subsequently was demonstrated to be followed by a transcriptional deregulation of Notch1-dependent genes, particulary c-Myc, Dtx1 and Hes1. Finally, a functional network of deregulated genes could be constructed based on all generated expression data. The network points to a deregulation of CD95 dependent apoptosis signalling in the respective Atm-deficient, thymic lymphoma T-cell population.
