Abstract

In der vorliegenden Arbeit lag der Fokus auf der Identifikation von Substraten und Interaktionspartnern der in die DNA-Schadensantwort eingebundenen Serin/Threonin Kinase Chk1. Chk1 reguliert in eukaryotischen Organismen die Transition der Zelle in verschiedene Zellzyklusphasen durch Phosphorylierung. Auf Grund dessen eignete sich besonders die proteinbiochemische Methode KESTREL zur Substratfindung, bei der Lysate chromatographisch aufgetrennt und in einer anschließenden Kinasereaktion eingesetzt werden. Größtes Problem war die Reproduzierbarkeit der Ergebnisse, insbesondere der Signale nach gelelektrophoretischer Auftrennung der Kinasereaktionen. Auf Grund der in dieser Arbeit ausführlich dargelegten Probleme in der Reproduzierbarkeit, wurde der KESTREL-Assay zur Substratidentifikation eingestellt und ein neuer molekularbiologischer Ansatz (Hefe-2-Hybrid) zur Identifikation von Interaktionspartnern von Chk1 begonnen. Im Rahmen der Diplomarbeit von Friederike Krop konnte mit Hilfe der in dieser Arbeit hergestellten chromatographischen Fraktionen, ein Substrat der Tyrosinkinase Pyk2 identifiziert werden (MAT2A Methionine Adenosyltransferase II, alpha; Krop, 2005). Dies deutet auf ein Chk1 spezifisches Problem im KESTREL Assay hin. Die Suche nach Interaktionspartner mittels des Hefe-2-Hybrid Systems führte zur Identifikation der N-Methylpurine-DNA-Glykosylase als potentieller Interaktions-partner der Chk1 Kinase. Diese Interaktion konnte biochemisch durch Bindungsexperimente an Glutathionagarose bestätigt werden. Weiterführende Versuche zeigten sowohl eine Co-Elution in chromatographischen Fraktionen, als auch eine Co-Lokalisation des MPG und Chk1 Proteins im Zellkern nach Co-Transfektion in der Immunfluoreszenz. Erste Daten deuteten darauf hin, dass sich die Lokalisation von MPG nach Behandlung mit dem Chemotherapeutikum Cisplatin ändert. Die Interaktion wurde somit mit zwei voneinander unabhängigen Methoden nachgewiesen. Diese Experimente lieferten erste Hinweise auf eine Beteiligung der N-Methylpurine-DNA-Glykosylase in die Regulation der Chk1 vermittelten DNA-Schadensantwort.

Translation of abstract (English)

The present thesis is focused on the identification of substrates and interaction partners of the DNA-damage pathway integrated serine/threonine Chk1 kinase. In eukaryotic organisms Chk1 regulates the transition of the cells in different cell cycle phases via phosphorylation. The biochemical method KESTREL was used to identify new Chk1 substrates. In this procedure lysates are separated by chromatography and used in subsequent kinase assays. The reproducibility of the experiments, especially the indentification of radioactive signals after gelelectrophoretic separation of the kinase reactions turned out to be the most significant problem. Based on this the KESTREL approach has been discontinued and a new approach (Yeast Two Hybrid) to identify interaction partner for Chk1 was started. However, with the help of the chromatographic fractions produced for KESTREL a new subtrate of the tyrosine kinase Pyk2 could be identified (MAT2A Methionine Adenosyltransferase II, alpha; Krop, 2005). This pinpoints to a Chk1 specific problem in the KESTREL assay. A new interaction partner, N-Methylpurine-DNA-Glycosylase, was identified by using the Yeast Two Hybrid system. This interaction was confirmed biochemically using GST-pulldown technology. Furthermore, both proteins coeluted in chromatographic fractions, as well as a colocalized in cells. First data indicated that the localization of MPG changed after treatment with chemotherapeutic cislatin. In summary the interaction was confirmed by two independent methods. These experiments could provide a first indication an involvement of the N-Methylpurine-DNA-Glycosylase in the regulation of Chk1 dependent DNA damage signaling.
