Abstract

Bacillus thuringiensis japonensis Stamm Buibui (BTJ) zeigt Melolontha melolontha eine geringe Wirkung, während Anomala-Arten und Popillia japonica empfindlich sind. Zur Identifizierung der Ursachen für die unterschiedliche Wirksamkeit, wurde die Darmphysiologie der Larven von P. japonica und M. melolontha untersucht. Ergebnisse: Das Spektrum der Darmproteasen: Im Darmsaft von M. melolontha wurden jeweils 3 trypsin- und elastaseartige Serinproteasen, 1 Aminopeptidase und 2 weitere Metalloproteasen charakterisiert. Hinzu kommt eine membranständige Aminopeptidase. Im Vergleich zu weiteren Scarabaeiden variieren die Anzahl, die Molekularmassen und die Aktivitäten einzelner Proteasen, aber einen qualitativen Unterschied durch weitere Protease-Klassen gibt es nicht. Das Redoxpotential im Darmmilieu: Im Mitteldarm von M. melolontha herrschen weitaus reduzierendere Bedingungen als bei P. japonica. Durch mikrobielle Fermentationsprozesse sinkt der O2-Partialdruck im Zentrum des Mitteldarms im Gegensatz zur Peripherie ab. Durch ein geringeres Oberflächen/Volumen-Verhältnis ist das Redoxpotential bei M. melolontha insgesamt negativer. Dieses Redoxpotential trägt zur Löslichkeit des 130 kDa Protoxins bei. Auch Detergenzien, wie sie im Darmsaft vorkommen, unterstützen die Lösung des Toxins.. Das gelöste Protoxin wird in vitro von einer mit dem Toxinkristall assoziierten sog. endogenen Metalloprotease von BTJ zum aktiven 65 kDa Toxin prozessiert. Aus der Kombination dieser Gegebenheiten und entsprechender in vitro und in vivo Versuche mit dem Cry8C Toxin kann für M. melolontha folgender Resistenzmechanismus abgeleitet (vorgeschlagen) werden: Während beim Cry8C-sensitiven Japankäfer das Toxin nur zu 55 kDa weiter abgebaut wird, erfolgt bei M. melolontha ein Abbau zu Fragmenten <10 kDa, weil es durch die stärker reduzierenden Bedingungen zu einer Entfaltung des Toxins kommt, wodurch zusätzlich interne Schnittstellen freigelegt werden.

Translation of abstract (English)

Bacillus thuringiensis japonensis strain Buibui (BTJ) is only poorly toxic to Melolontha melolontha while Anomala species Popillia japonica are sensitive. For identification of the reasons which lead to different susceptibility the gut physiology of the larvae of the BTJ sensitive P. japonica and of the resistant M. melolontha were analysed leading to the following results. Variety of the midgut proteases: In the midgut juice of M. melolontha 3 trypsin-like and 3 elastase-like proteases, an aminopeptidase, and two other metalloproteases were identified and characterized. In addition, there was a membrane-bound aminopeptidase. Numbers, molecular masses and activities of other analyzed scarabs varied, but there was no difference in the spectrum of proteases. The gut oxidation reduction potential (ORP): In the midgut of M. melolontha the conditions are more reducing than in the midgut of P. japonica. The O2 partial pressure in the centre of the midgut where microbial fermentation can be found is much lower than in the periphery. Because of the smaller surface/volume ration the ORP of the midgut juice of M. melolontha is stronger reducing than in P. japonica. The ORP is responsible for the solubilization of the 130 kDa protoxin. Detergents which are present in the midgut juice promote this process. In addition, the protoxin can be dissolved in vitro by chaotropic ions. The soluble protoxin is processed to an active 65 kDa toxin by an endogenous metalloprotease of BTJ which is associated with the crystal. Based upon the combination of these conditions and proteolytic assays with Cry8C the following mechanism of resistance can be proposed for M. melolontha: While the toxin is processed to 55 kDa by P. japonica the degradation to fragments <10 kDa in M. melolontha by the same trypsin-like enzymes is due to the stronger reducing conditions which lead to unfolding of the toxin. Consequently, additional internal cleavage sites are offered for proteolysis.
