Abstract

VERGLEICHENDE ANALYSE VERSCHIEDENER VERPACKUNGSSYSTEME FÜR HERPESVIRALE AMPLICON-VEKTOREN AM BEISPIEL VON ANTIGENEN DES HUMANEN PAPILLOMVIRUS-16 Amplicon-Vektoren sind aufgrund ihrer (i) enormen Transgenkapazität von bis zu 152 kbp, (ii) ihres breiten Wirtsspektrums und (iii) der hohen Toleranz des natürlichen Wirts Mensch vielversprechende Kandidaten für den Einsatz in der Gentherapie. HSV-1 Amplicon-Vektoren bestehen aus einem Amplicon-Plasmid, welches in die HSV-1 Virushülle verpackt ist. Das Amplicon-Plasmid ist Träger des jeweiligen Transgens. Zur Verpackung der Amplicon-Plasmide wurden zwei virale Helfersysteme, nämlich (i) die Virusrekombinanten HSV-1 LaL und HSV-1 LaLDJ und (ii) ein HSV-1 Bakterielles Artifizielles Chromosom (HSV-1 BAC) verwendet. Ein optimales Verpackungssystem für Amplicon-Vektoren sollte hohe Amplicon-Titer bei gleichzeitig niedriger Helferviruskontamination liefern. In der vorliegenden Arbeit wurden anhand des Modellantigens L1, dem Hauptkapsidprotein des humanen Papillomvirus-16 (HPV-16), die zwei verschiedenen Verpackungssysteme bezüglich Verpackungseffizienz, Helfervirus-kontamination und Transgenexpression untersucht. HPV-16 wird als ein wichtiger Faktor bei der Entstehung des Gebärmutterhalskrebses angesehen und L1 ist einer der Hauptansatzpunkte für Vakzinierungsversuche gegen HPV-16. In Bezug auf die Verpackungseffizienz war das HSV-1 LaL-System am effektivsten, wies aber auch gleichzeitig die höchste Helferviruskontamination auf. Mit Hilfe des HSV-1 LaLDJ-Systems wurden dagegen keine Amplicons erhalten. Das HSV-1 BAC-System zeigte bei akzeptabler Verpackungseffizienz keinerlei nachweisbare Helfervirus-kontamination. Im direkten Vergleich induzierten die mit HSV-1 LaL kontaminierten Amplicons eine stärkere L1-Transgenexpression als die mit HSV-1 BAC verpackten Amplicons. Eine mögliche Erklärung dafür ist, dass die kontaminierenden Helferviren Replikation und Verpackung unterstützen. Ein weiterer Schwerpunkt dieser Arbeit lag auf der Entwicklung von HSV/HPV-Hybridvektoren. Als Modell diente das HPV-16 E7-Protein, das eine wichtige Rolle in der HPV-16 induzierten Karzinogenese spielt und einen häufigen Angriffspunkt für immuntherapeutische Maßnahmen darstellt. Die generierten Hybrid-Vektoren sollen im Vergleich zu bereits etablierten Systemen eine verbesserte Immunantwort gegen E7 induzieren. Dazu wurde das herpesvirale Tegumentprotein VP22 und das HSV-1 Protein der Virushülle Glykoprotein C (gC) mit dem E7-Protein fusioniert. Detaillierte Untersuchungen der generierten Amplicon-Vektoren zeigten, dass die Fusionsproteine korrekt in den Amplicon-Vektor integriert wurden. Die Ergebnisse dieser Arbeit könnten zur Verbesserung und Optimierung des Verpackungsprozesses von Amplicon-Vektoren im Hinblick auf eine gentherapeutische Anwendung von Nutzen sein. HSV/HPV-Hybrid-Vektoren sind ein mögliches Werkzeug zur Modifizierung des Tropismus und zum Protein-Targeting.

Translation of abstract (English)

COMPARATIVE ANALYSIS OF DIFFERENT PACKAGING SYSTEMS FOR HERPESVIRAL AMPLICON VECTORS USING MODEL ANTIGENS OF THE HUMAN PAPILLOMAVIRUS-16 Amplicon-vectors are promising candidates for gene therapy issues, because they offer (i) an enormous transgene capacity up to 152 kbp, (ii) a broad tropism and (iii) they are well tolerated by the human host. HSV-1 amplicon vectors consist of the amplicon plasmid, carrying the gene of interest packaged into a HSV-1 virion. For the encapsidation of the amplicon plasmid, two viral helper systems (i) the viral “helper” recombinants HSV-1 LaL and HSV-1 LaLDJ and (ii) an HSV-1 Bacterial Artificial Chromosome (HSV-1 BAC) were used. An optimal packaging system should offer high amplicon titers and low contamination with helper virus. In the present work the two different packaging systems were tested regarding the amplicon titers, the load of helper and the expression of a transgene, in this case L1, the major capsid protein of the human papillomavirus-16 (HPV16). HPV-16 is a well known high risk factor for the development of cervical cancer and the L1-protein is a frequently used target for vaccination studies against HPV-16. Considering the packaging efficiency the HSV1-LaL system was the most effective, but also the one with the highest helper virus contamination. HSV-1LaLDJ proved to be pretty ineffective because no amplicons could be produced using this virus-recombinant. The HSV-1 BAC system showed no detectable helper contamination and the titer was found to be acceptable. Regarding the transgene expression, the contaminated pAL1LaL amplicons were under the same conditions significantly more effective than the ones packaged with BAC. One possible explanation could be a supporting function of the present helper viruses considering replication and packaging. Another main topic of this work was the development of HSV/HPV-hybrid vectors. The HPV-16 E7 protein served as a model, because it plays a major role in the HPV-16 induced carcinogenesis and it is a main target for immune therapeutic arrangements. The generated hybrid vectors should improve the immune response to E7 compared to already established systems. For this end the herpesviral tegument protein VP22 and the HSV-1 envelope glycoprotein C (gC) were fused to the HPV-16 E7-protein. Detailed analysis of the generated amplicons proved the correct integration of the fusion proteins into the amplicon vector. Taken together, the results of the present work could help optimising the packaging processes of amplicon vectors towards the use in gene therapy. The design and development of HSV/HPV-hybrid-vectors are suited for specific modifications of tropism and protein targeting.
