Abstract

Gamma-Protocadherine (gamma-pcdh), glykosilierte Typ I Transmembranproteine mit einer extrazellulären Domäne aus sechs Cadherin-artigen Modulen, sind vermutlich an Zelladhäsion beteiligt und in Säugetieren überlebensnotwendig. In der Maus gibt es 22 gamma-pcdh-Isoformen, deren Ektodomäne, Transmembrandomäne und ein kleiner Teil der intrazellulären Sequenz gemeinsam von variablen Exons mit eigenem Promotor kodiert werden. Am 3´- Ende dieses seriell angeordneten „Genklusters“ befinden sich drei, als konstante Region bezeichnete Exons, welche den Rest der zytoplasmatischen Sequenz, die invariante C-terminale Domäne (gamma-ICD), kodieren. Durch „cis-Splicen“ der von diesem „Genkluster“ erzeugten Primärtranskripte können auf diese Weise Isoformen exprimiert werden, die alle die gamma-ICD enthalten. Die vorliegende Arbeit beschäftigt sich mit funktionellen Aspekten der gamma-ICD. Es wurden gentechnisch veränderte Mäuse analysiert, welche nach Entfernung des konstanten Exon 1 (deltaC1) keine funktionelle gamma-ICD mehr exprimieren können. Die neonathal sterbenden, homozygoten deltaC1/deltaC1-Tiere zeigten einen nahezu vollständigen Verlust der Lokusexpression. In den überlebenden, heterozygoten + /deltaC1-Mäusen wurden vom verbleibenden, intakten Allel Expressionsstärken von 50% der Wildtypmengen erwartet. Der Abfall der Transkript- und Proteinmengen auf 25% relativ zum Wildtyp war deshalb auffällig. Auffällig war auch die in Populationen von Nervenzellen auf Schnitten von adultem Maushirn beobachtete, gamma-ICD-spezifische Färbung des Zellkerns. Dies lässt eine Funktion der gamma-ICD bei der Genregulation im Zellkern vermuten. Für weitere Analysen wurden daher einfachere Zellkultursysteme verwendet, in welchen verschiedene, rekombinante gamma-pcdh-Proteinvarianten transient exprimiert werden konnten. Zunächst wurde damit die intrazelluläre Verteilung der gamma-pcdh in COS-1 Zellen bestimmt. Immunofluoreszensfärbungen ergaben, dass die Verteilung der N- und C-terminalen gamma-pcdh-Domänen unterschiedlich war und dass sich -wie in Neuronen- das gamma-ICD enthaltende Fragment im Zellkern befand. Sowohl in transfizierten COS-1 Zellen, welche mit gamma-Sekretaseinhibitoren behandelt wurden, als auch in gamma-Sekretase-inaktiven PS1/2 K.O. Zellen fanden sich zwingende Belege für die Prozessierung der gamma-pcdh durch „Regulierte Intramembran Proteolyse“ (RIP). Die intrazelluläre Domäne wird offenbar durch die gamma-Sekretase freigesetzt und dann in den Zellkern importiert. Analysen der Transkript- und Proteinmengen von transient (myc)gamma-ICD überexprimierenden HEK-293 Zellen zeigten eine Expressionssteigerung der endogenen Lokusexpression. Über zunehmende Luziferaseintensitäten von Reportervektoren ließ sich auch eine (myc)gamma-ICD-vermittelte Aktivitätssteigerung mehrerer gamma-pcdh-Promotor ermitteln. Darüber hinaus ließ sich in SH-SY5Y Zellen durch die Hemmung der gamma-Sekretaseaktivität auch die endogene Menge an gamma-pcdh reduzieren, so dass die gamma-pcdh-Lokusexpression über RIP reguliert wird. Diese Ergebnisse beschreiben den ersten Signaltransduktionsweg der gamma-pcdh und identifizieren damit ein neues, wichtiges Substrat des gamma-Sekretasekomplexes. Letztendlich bietet dieser Signaltransduktionsweg auch eine attraktive Erklärungsmöglichkeit für die in heterozygoten + /deltaC1-Mäusen beobachtete Reduktion der gamma-pcdh-Transkript- und Proteinmengen.

Translation of abstract (English)

Gamma-protocadherins (gamma-pcdhs), type I membrane-spanning glycoproteins with an extracellular domain of six cadherin-like modules are probably involved in cell adhesion and required for survival in the mammal. There are 22 gamma-pcdh-isoforms in the mouse, with the ectodomain, one transmembrane domain, and a small portion of the intracellular sequence together encoded by one variable exon, each with its own promotor. Three constant region exons are located at the 3’ end of the tandemly arranged gene cluster which encode the invariant C-terminal domain (gamma-ICD). Cis-splicing of the cluster’s primary transcripts produces the gamma-ICD containing isoforms. The present work focused on functional aspects of the gamma-ICD. First, gene-targeted mice were analyzed, that can not express a functional gamma-ICD, due to removal of constant exon 1 (deltaC1). Homozygous deltaC1/deltaC1 animals showed almost complete loss of gamma-pcdh locus expression and died shortly after birth. Remarkably, although 50% expression was expected from the presence of the remaining wild-type allele in viable +/deltaC1 heterozygous mice, gamma-pcdh transcript and protein levels dropped to 25% relative to wildtype. Notably, subsets of neurons in sections from adult mouse brain exhibited nuclear gamma-ICD-staining. Thus, gamma-ICD function might include transcriptional regulation in the nucleus. So, for further analysis, simpler cell culture systems were used and various recombinant gamma-pcdh proteins were transiently expressed. First, the subcellular distribution of gamma-pcdh was traced in COS-1 cells. Immunofluorescense staining revealed that N- and C-terminal portions of the gamma-pcdhs were localized differently, with the gamma-ICD containing fragment found in the nucleus, reminiscent of neurons. Both, transfected COS-1 cells treated with gamma-secretase inhibitors, and gamma-secretase inactive PS1/2 K.O. cells, made a strong case for gamma-pcdhs undergoing „Regulated Intramembrane Proteolysis“ (RIP). Obviously, the intracellular domain is released by gamma-secretase for import into the cell nucleus. Upon transcript- and protein analysis in HEK-293 cells, transiently overexpressed (myc)gamma-ICD promotes gamma-pcdh locus expression. Additionally, increased luciferase levels of reporter constructs demonstrated (myc)gamma-ICD-mediated, enhanced gamma-pcdh promoter activity. Moreover, inhibition of gamma-secretase function reduced endogenous gamma-pcdh levels in SH-SY5Y cells, showing that gamma-pcdh locus expression is regulated by RIP. These results reveal a novel intracellular signaling pathway for gamma-pcdhs and identify a novel vital target for the gamma-secretase complex. Finally, this signalling pathway represents an attractive explanation for the observed reduction of gamma-pcdh transcript- and protein levels in +/deltaC1 heterozygous mice.
