Abstract

Ausgangspunkt der Doktorarbeit war die funktionelle Charakterisierung von 1- und 2-COP im sekretorischen Weg. Dazu wurden Unterschiede von 1- und 2-COP hinsichtlich verschiedener Interaktionspartner und ihrer Lokalisierung entdeckt. Es wurden Bindungsstudien mit den g1- und g2-COP appendage Domänen durchgeführt. Appendage Domänen wurden erstmalig bei den a/b-Adaptinen, Bestandteile der Clathrin-Adaptorkomplexe (APs), identifiziert. Neben Aminosäuresequenz-Vergleichen zwischen AP-Komplexen und dem b/d/g/z-COPI Subkomplex (Schledzewski et al. 1999), der unter nicht physiologischen Salz-Bedingungen vom a/b’/e-COPI Subkomplex dissoziiert werden kann, legte die Strukturaufklärung der g1-COP appendage Domäne (Hoffman et al. 2003; Watson et al. 2004) eine Analogie zwischen COPI- und Clathrin-vermitteltem Transport nahe. Während eine Vielzahl von appendage-Bindern im Clathrin-System beschrieben sind (Praefcke et al. 2004), waren im COPI-System ausschließlich ArfGAP2 und ArfGAP3 als g1-COP-Binder bekannt (Watson et al. 2004). In dieser Arbeit konnten ArfGAP1, ArfGAP3 und Annexin A11 als präferentielle g2-COP- und Zfp289 als g1-COP- Bindepartner identifiziert werden. Zfp289 ist als Protein mit Arf1-GAP Aktivität beschrieben worden (Singh et al. 2001) und ist möglicherweise identisch mit ArfGAP2. Während eine Beteiligung der kleinen GTPase Arf1 und deren ArfGAPs an der COPI-Biogenese unstrittig ist (Palmer et al. 1993), ist der Einfluß von Annexin A11 gegenwärtig unverstanden. Eine Beteiligung von Ca2+ sowohl bei dem COPI-vermittelten Transport (Chen et al. 2002) als auch an der Rekrutierung von Annexin A11 an Membranen (Lecona et al. 2003) legt dieser Interaktion zumindest eine funktionelle Relevanz nahe. Weiter konnten g1z2- und g2z1-COP enthaltende COPI-Vesikel immunisoliert werden. Unterstützt wird die Präsenz von diesen distinkten Vesikeln durch eine Analyse der ultrastrukturellen Lokalisierung von g1-, g2- und z2-COP: Während g1- und z2-COP präferentiell im cis-Golgi lokalisierten, wies g2-COP eine trans-Golgi Lokalisation auf. Dies legt eine Funktion von g1z2-Coatomer im cis- und von g2z1-Coatomer im trans-Golgi Bereich nahe. Wie auch im Clathrin-System wird daher beim COPI-vermittelten Transport Heterogenität durch Variation der Hüllproteine erlangt. Ferner könnten die den AP Komplexen entsprechenden Komponenten von Coatomer, g1- und g2- bzw. z1- und z2-COP, eine Direktionalität des Transports determinieren. Letztlich konnten zwei Zelllinien etabliert werden, die einen induzierbaren, individuellen g1- oder g2-COP knock-down erlaubten. Der knock-down der einzelnen g-COP Isoformen konnte, wie auch ein RNAi-basierter knock-down beider Isoformen gleichzeitig, nur transient erreicht werden. Dies unterstützt eine essentielle und unterschiedliche Beteiligung von g1- und g2-COP am sekretorischen Weg.

Translation of abstract (English)

The present work aimed at the functional characterisation of g1- and g2-COP within the secretory pathway. To this end, differences of g1- and g2-COP with respect to protein interactions and their localisation were investigated. First, binding studies with g1- and g2-COP appendage domains were performed. Originally, appendage domains were identified in a/b-adaptins, components of the clathrin adaptor complexes (APs). An analogy between COPI- and clathrin-mediated transport is shown by aminoacid sequence comparison between clathrin’s AP complexes and the b/d/g/z-COPI subcomplex (Schledzewski et al. 1999), which can be dissociated under non-physiological salt conditions from the a/b’/e-COPI subcomplex. Additionally, the g1-COP appendage domain structure showed a striking similarity to a-adaptin (Hoffman et al. 2003; Watson et al. 2004). A variety of proteins binding to appendage domains of the clathrin system is known (Praefcke et al. 2004). In contrast, only ArfGAP2 and ArfGAP3 are known to bind to g1-COP in the COPI system (Watson et al. 2004). In this thesis, ArfGAP1, ArfGAP3 and annexin A11 are identified as preferential g2-COP binding partners, and Zfp289 of g1-COP. Zfp289 was described as an Arf1-GAP (Singh et al. 2001) and likely represents ArfGAP2. While a participation of the small GTPase Arf1 and its ArfGAPs in COPI-biogenesis is established (Palmer et al. 1993), a contribution of annexin A11 in vesicle formation is not understood. A role played by Ca2+ in COPI-mediated transport (Chen et al. 2002) and in recruitment of annexin A11 to membranes (Lecona et al. 2003) could functionally link annexin to COPI transport. Second, g1z2- und g2z1-COP containing COPI vesicles could be immunisolated. This finding is supported by the determination of the ultrastructural localisation of g1-, g2- and z2-COP within the Golgi apparatus: g1- and z2-COP are predominantely found in the cis- and g2-COP in the trans-Golgi. This implies a function of g1z2 coatomer in the cis- and of g2z1 coatomer in the trans-Golgi region. With respect to individual clathrin transport proteins, it’s tempting to compare the major coatomer isoforms with the various adapter proteins in the clathrin system, known to serve specific transport steps. Comparable to the clathrin system, heterogenity of vesicles in the COPI system is achieved by variation of the coat proteins. Additionally, the AP complex resembling components of coatomer, g1-, g2-, z1- and z2-COP, could determine a directionality of transport. Third, two cell lines were established that allow an individual knock-down of either g-COP isoform. In addition, a simultaneous RNAi-based knock-down of both isoforms was performed. The knock-down of the individual g-COP isoforms, as the simultaneous knock-down of both isoforms, could be achieved transiently only. This points to an essential and differential role of both g-COP isoforms within the secretory pathway.
