Abstract

Das Adaptormolekül FADD/MORT1 ist essentiell für die Signaltransduktion der Todesrezeptoren CD95, TNF-RI, DR3, DR4 und DR5, spielt eine wichtige Rolle beim Wiedereintritt von ruhenden T Zellen in den Zellzyklus und wird an Serinresten phosphoryliert. Untersuchungen mit dem Phosphorylierungs-spezifischen Antikörper 1C4 wiesen Serin 194 als einzige Phosphorylierungsstelle in FADD nach. Zellzyklusarreste und Elutrationen ergaben, daß die Phosphorylierung Zellzyklus-abhängig in G2/M erfolgt. In vitro Kinase Tests und Immunpräzipitationen führten zur Detektion einer ca. 70 kDa großen Kinase, die mit der C-terminalen Hälfte der DED von FADD assoziiert. Die Kinase selbst lokalisiert in der Nukleusfraktion. Experimente mit T Zellen aus C-FADD transgenen Mäusen und C-FADD stabil exprimierenden Fibroblasten hatten gezeigt, daß C-FADD die Proliferation inhibieren kann. Um zu testen, ob die Phosphorylierung von C-FADD diesen Zellzyklusarrest bewirkt, wurden die Brustkarzinomzellinie MCF-7 und die spontan immortalisierte epitheliale Brustzellinie MCF-10A mit den adenoviralen Vektoren AdV-C-FADD Wildtyp, AdV-C-FADD S194A und AdV-C-FADD S194E infiziert. Die Expression des Wildtyps führte nur in der Zellinie MCF-10A zu einem Zellzyklusarrest in G2/M, was mit der Phosphorylierung von FADD in G2/M korrelierte. Auch die adenovirale Transduktion von naiven CAR transgenen T Zellen mit FADD Wildtyp und Punktmutanten ergab, daß der Proliferationsarrest von der Phosphorylierung des Wildtyp Proteins abhängig ist. Um die Rolle von FADD in der DISC Bildung von CD95 Typ I Zellen zu untersuchen, wurde in BJAB C-FADD Transfektanten Apoptose mittels CD95 induziert. Während im DISC von C-FADD S194A Transfektanten nach Stimulation von CD95 kaum endogenes FADD und keine Caspase-8 gefunden werden konnte, waren bei C-FADD S194E endogenes FADD und Caspase-8 schon mit dem unstimulierten Rezeptor assoziiert. Untersuchungen mit den Transfektanten zeigten, daß C-FADD Wildtyp und S194E in der Lage sind, mit Aktin zu assoziieren. Die Behandlung von Zellen mit dem Kinaseninhibitor Staurosporin verhinderte die Bildung des DISC gänzlich und reduzierte die Phosphorylierung von FADD, was darauf hindeutet, daß die Phosphorylierung von FADD für die Signaltransduktion des CD95 Rezeptors in Typ I Zellen notwendig ist.

Translation of abstract (English)

The adaptor molecule FADD/MORT1 is essential for the signal transduction of the death receptors CD95, TNF-RI, DR3, DR4 and DR5, plays an important role in the re-entry of resting T cells into the cell cycle and is phosphorylated on serine residues. Experiments with the phosphorylation specific antibody 1C4 revealed serine 194 as single phosphorylation site in FADD. Cell cycle arrests and elutriation experiments showed that the phosphorylation occurs in a cell cycle dependent manner in G2/M. In vitro kinase assays and immunoprecipitations led to the detection of a ca. 70 kDa kinase that associates with the C-terminal half of the DED of FADD. The kinase localizes in the nukleus fraction. Experiments with T cells from C-FADD transgenic mice and stably C-FADD expressing fibroblasts had shown that C-FADD can inhibit proliferation. To test if it is the phosphorylation of C-FADD that induces this cell cycle arrest, the breast carcinoma cell line MCF-7 and the spontaneously immortalised epithelial breast cell line MCF-10A were transduced with the adenoviral vectors AdV-C-FADD wild type, AdV-C-FADD S194A and AdV-C-FADD S194E. Expression of the wild type protein in MCF-10A cells led to a cell cycle arrest in G2/M, which correlated with the phosphorylation of FADD in G2/M. Adenoviral transduction of naive CAR transgenic T cells with C-FADD wild type and point mutants confirmed that the cell cycle arrest is dependent on the phosphorylation of the wild type protein. To investigate the role of FADD in the formation of the CD95 DISC in type I cells we induced apoptosis through CD95 in stably C-FADD expressing BJAB transfectants. After stimulation of CD95 the DISC of C-FADD S194A transfectants did not contain any endogenous FADD or caspase-8, while in C-FADD S194E transfectants endogenous FADD and caspase-8 were already associated with the unstimulated receptor. Experiments with these transfectants showed that C-FADD wild type and S194E associate with actin. Treatment of cells with the kinase inhibitor staurosporine inhibited the formation of the DISC completely and reduced the phosphorylation of FADD. This suggests that the phosphorylation of FADD is necessary for the signal transduction of the CD95 receptor in type I cells.
