"The interactions between tumor cells and the microenvironment play a crucial role in tumor\r\nprogression. Tumor cells secrete growth factors, cytokines and proteases and are thus able\r\nto affect the gene expression of stromal cells. Activate\r\nd stromal cells like fibroblasts, immune\r\ncells and endothelial cells are able to promote tumor progression by releasing growth factors,\r\ncytokines and proteases. One of the factors which play a central role in this tumor\r\n-\r\nand\r\nstroma\r\n-\r\ninteraction is interleuk\r\nin\r\n-\r\n6. In many tumor entities, the IL\r\n-\r\n6 signal\r\npathway is highly\r\ndysregulated\r\nby overexpression of IL\r\n-\r\n6 and otherwise by permanent activation of the\r\nSTAT3\r\ntranscription factor.\r\nIn this work, we showed that IL\r\n-\r\n6 stimulation resulted in an increased prolifera\r\ntion of HaCaT\r\n-\r\nras A5 benign tumor keratinocytes, but does not affect the fibroblast cell line MSU1.1.\r\nThis increase correlates with the activation of the JAK/STAT signaling pathway. We were\r\nable to demonstrate that the amount of activated STAT3 proteins\r\nafter IL\r\n-\r\n6 stimulation in\r\nHaCaT\r\n-\r\nras A5 cells is strongly up regulated and the inhibition of STAT3 activation also\r\ninhibits the proliferation of HaCaT\r\n-\r\nras A5 cells. This showed the crucial role of the STAT3\r\nactivation in proliferation of HaCaT\r\n-\r\nras A5 cells.\r\nIn contrast, fibroblasts showed a stronger inhibition of the JAK/STAT signal pathway by\r\nSOCS3 in comparison with the HaCaT\r\n-\r\nras A5 cells.\r\nAll data where used to establish a mathematical model of JAK/STAT pathway which is able\r\nto describe the dynamic behav\r\nior of STAT3 and SOCS3 Proteins.\r\nIn addition to the effects shown, IL\r\n-\r\n6 is also able to affect the proliferation in an indirect\r\nmanner. To this end, IL\r\n-\r\n6 stimulation activates a network of growth factors in HaCaT\r\n-\r\nras A5\r\ncells and fibroblasts like HGF, KGF\r\n, VEGF or IL\r\n-\r\n8. While IL\r\n-\r\n6 showed no direct influence of\r\nthe proliferation of fibroblasts, the addition of the condition media of both IL\r\n-\r\n6 stimulated\r\nHaCaT\r\n-\r\nras A5 and as well as IL\r\n-\r\n6 stimulated MSU1.1 resulted in strong up regulation of the\r\ncell numbers o\r\nf the other cell line.\r\nComparative gene expression analyses clearly showed that upon IL\r\n-\r\n6 stimulation of over\r\n16000 analyzed genes in HaCaT\r\n-\r\nras A5 and Fibroblasts only a subset of 19 genes was up\r\nregulated in both cell types. The up regulated genes were f\r\nound to be closely connected with\r\nthe interferon signal pathway which gives a potential hint of the cause of the IL\r\n-\r\n6 induced\r\ninhibition of growth in fibroblasts.\r\nFurther gene expression analyses showed that the proteinase inhibitor SerpinB4, was\r\npermanent\r\nly up regulated in IL\r\n-\r\n6 stimulated HaCaT\r\n-\r\nras A5 cells. The down regulation of\r\nSummary\r\n9\r\nSerpinB4 by siRNA knockdown indicated that SerpinB4 plays an important role in the\r\nproliferation of HaCaT\r\n-\r\nras A5.\r\nThe observation of this work of the detailed regulation of IL\r\n-\r\n6\r\nin tumor and stroma cells\r\nshould give information about the tumor promoting effect of IL\r\n-\r\n6 and will ultimately serve as a\r\nbasis for therapeutic therapies."^^ .
