"In\r\nthe\r\nprotozoan\r\nparasite\r\nTrypanosoma\r\nbrucei\r\ngene\r\nexpression\r\nis\r\nmostly\r\nregulated\r\non\r\nthe\r\npost-­‐transcriptional\r\nlevel.\r\nC3H\r\nzinc\r\nfingers\r\nhave\r\nshown\r\nto\r\nplay\r\nan\r\nimportant\r\nrole\r\nin\r\nthis\r\nprocess.\r\nSo\r\nfar,\r\nall\r\ndescribed\r\nC3H\r\nzinc\r\nfinger\r\nin\r\nTrypanosoma\r\nbrucei\r\nhave\r\nshown\r\nto\r\nstabilize\r\ntheir\r\ntargets.\r\nThis\r\nwork\r\nfocuses\r\non\r\nthe\r\nzinc\r\nfinger\r\nprotein\r\nZC3H32.\r\nA\r\nlarge-­‐scale\r\nisolation\r\nof\r\nfree\r\nand\r\nmembrane-­‐bound\r\npolyribosomes\r\nfrom\r\nproyclic\r\nand\r\nbloodstream\r\ntrypanosomes,\r\nwhich\r\nis\r\nalso\r\ndescribed\r\nin\r\nthis\r\nthesis,\r\nsuggested\r\nthat\r\nthis\r\nprotein\r\nassociates\r\nwith\r\npolyribosomes.\r\nHowever,\r\nsmall-­‐scale\r\npolyribosome\r\nisolations\r\nfollowed\r\nby\r\nWestern\r\nblotting\r\nrevealed\r\nthat\r\nonly\r\na\r\nminor\r\nfraction\r\nof\r\nit\r\nis\r\nactually\r\nassociated\r\nwith\r\npolyribosomes.\r\nImmunofluorescence\r\nshowed\r\nthat\r\nZC3H32\r\nlocalizes\r\nto\r\nthe\r\ncytoplasm.\r\nA\r\nyeast-­‐two-­‐\r\nhybrid\r\nscreen\r\nidentified\r\nit\r\nas\r\na\r\nputative\r\nbinding\r\npartner\r\nof\r\nMKT1.\r\nThis\r\ninteraction\r\nwas\r\nconfirmed\r\nby\r\nco-­‐immunoprecipitation.\r\nRNAi,\r\nas\r\nwell\r\nas\r\nknock-­‐out\r\nstudies,\r\nshowed\r\nthat\r\nZC3H32\r\nis\r\nessential\r\nin\r\nthe\r\nbloodstream\r\nform.\r\nNorthern\r\nblotting,\r\nas\r\nwell\r\nas\r\na\r\nSILAC\r\nscreen\r\nby\r\nUrbaniak\r\net\r\nal.\r\nindicate\r\nthat\r\nit\r\nis\r\nalso\r\nenriched\r\nin\r\nthis\r\nlife-­‐cycle\r\nstage\r\nas\r\ncompared\r\nto\r\nprocyclics.\r\nArtificial\r\ntethering\r\nof\r\nZC3H32\r\nto\r\na\r\nreporter\r\nRNA\r\nlead\r\nto\r\nthe\r\nRNA’s\r\ndegradation,\r\nsuggesting\r\nthat\r\nthis\r\nprotein\r\nhas\r\na\r\ndestabilizing\r\neffect.\r\nTethering\r\nof\r\nZC3H32\r\nfragments\r\nrevealed\r\nthat\r\nboth\r\nits\r\nN-­‐terminal,\r\nas\r\nwell\r\nas\r\nits\r\nC-­‐terminal\r\nregion\r\nare\r\nable\r\nto\r\ngenerate\r\nthis\r\ndestabilization\r\neffect,\r\nwhile\r\nthe\r\nmiddle\r\nregion,\r\ncontaining\r\nthe\r\nzinc\r\nfinger\r\ndomains,\r\ncan’t.\r\nRNA\r\nisolation\r\nfrom\r\npolyribosomal\r\nfractions\r\nshowed\r\nthat\r\ntethering\r\nof\r\nZC3H32\r\nalso\r\ndecreases\r\nthe\r\ntranslation\r\nof\r\nthe\r\nreporter\r\nRNA.\r\nHigh-­‐throughput\r\nsequencing\r\nof\r\npoly-­‐A+\r\nRNA\r\nfrom\r\na\r\nZC3H32-­‐RNAi\r\ncell\r\nline\r\nrevealed\r\n20\r\nRNAs\r\nthat\r\nwere\r\nup-­‐regulated\r\nupon\r\nZC3H32\r\nknock-­‐down\r\nand\r\nthus\r\nmight\r\nbe\r\nputative\r\ntargets.\r\nThe\r\nupregulation\r\nwas\r\nconfirmed\r\nfor\r\nthree\r\nof\r\nthese\r\ncandidate\r\nRNAs.\r\nThe\r\nmajority\r\nof\r\nthe\r\nputative\r\nZC3H32\r\ntargets\r\nplay\r\na\r\nrole\r\nin\r\nthe\r\ntrypanosome’s\r\nenergy\r\nmetabolism\r\nand\r\n15\r\nof\r\nthem\r\nare\r\nup-­‐regulated\r\nin\r\nprocyclics.\r\nThese\r\nresults\r\nsuggest\r\nthat\r\nZC3H32\r\nmight\r\nbe\r\ninvolved\r\nin\r\nthe\r\nstage-­‐specific\r\nregulation\r\nof\r\nthese\r\nRNAs\r\nin\r\nthe\r\nbloodstream\r\nform."^^ .
