This enables life’s great degree of organization. To allow nutrients and\r\ninformation to pass this barrier, cells use various transmembrane proteins for material\r\nand signal transduction. When creating artificial cells, synthetic biology has\r\nto account for these facts. This thesis presents the development of such a stable\r\nand easy-to-manipulate protocell model for cellular adhesion experiments. First\r\nadhesion of small unilamellar vesicles containing reconstituted blood-platelet integrin\r\nto functionalized surfaces were studied using quartz crystal microbalance. Due\r\nto restrictions of giant unilamellar vesicles (GUV), a novel compartment system,\r\nnamed droplet-stabilized giant unilamellar vesicles (dsGUV), was created combining\r\nthe versatility of microfluidics, the stability of surfactant-stabilized water-in-oil\r\ndroplets, with the high level of bio-mimicry of lipid membranes. Through the use\r\nof microfluidic pico-injection technology, components can be sequentially added to\r\nthe compartment, pathing the way for more complex and diverse model systems.\r\nIn this work, reconstitution of integrin into dsGUVs is presented.
