Abstract

Einfluss von Tensiden auf die Resorption von Substraten der ABC-Transportproteine 1.Standardisierung des Caco-2-Modells: Das Caco-2-Modell ist das am weitesten verbreitete Resorptionsmodell. Zur Verbesserung der Vergleichsmöglichkeiten von Ergebnissen, die in verschiedenen Laboratorien erhalten werden, wurden in einem Ringversuch unter Teilnahme von fünf Einrichtungen die Permeabilitätseigenschaften von ausgewählten Substanzen unter Einhaltung vorher festgelegter Versuchsvorschriften bestimmt. Der Vergleich zeigte keine Übereinstimmung der Absolutwerte der Permeabilitätskoeffizienten. Es wurde jedoch eine einheitliche Einstufung in bezug auf die Permeabilitätseigenschaften der Substanzen vorgenommen. Zusätzlich wurde eine unterschiedliche Expression der aktiven Transportproteine, p-gp und MRP2 festgestellt. Dies stellt auch das größte Problem der Standardisierung des Modells dar. 2. Einfluss von Tensiden auf die Resorption von Arzneistoffen: Zahlreiche Arzneistoffe haben eine schlechte Bioverfügbarkeit, weil ihre intestinale Resorption durch die Transportaktivität von Exportproteinen, z.B. p-gp und MRP2, eingeschränkt ist. Der Einfluss einer Reihe von Tensiden auf die Transportaktivität beider Proteine wurde im Caco-2-Modell untersucht. Viele Tenside inhibierten den Transport durch p-gp, wobei die Poloxamere, z.B. Pluronic P85, die größte Wirkung zeigten. Einige Tenside hemmten den Transport durch MRP2. Hierbei waren Cremophor RH40 und Solutol HS15 am effektivsten. Nach Bestimmung der Kritischen Mizellbildungskonzentration zeigte sich, dass die effektivsten Tensidkonzentrationen im Bereich der CMC liegen, was für eine Aktivität der einzelnen Tensidmoleküle und nicht der Mizellen spricht. Die untersuchten Tenside verursachten eine Erniedrigung der Membranfluidität der Zellen. Dies kann zur Hemmung der Aktivität der Transportproteine beitragen. Es konnte jedoch kein Unterschied zwischen mehr oder weniger wirksamen Tensiden festgestellt werden. Die Tenside zeigten in den wirksamen Konzentrationen keine Toxizität, was in zwei Toxizitätsstudien gezeigt werden konnte. Eine Resorptionsverbesserung von Arzneistoffen, die von p-gp und MRP2 transportiert werden, scheint durch Zugabe der wiksamen Tenside möglich zu sein.

Translation of abstract (English)

Influence of Surfactants on Absorption of Substrates of ABC-Transportproteins 1.Standardisation of the Caco-2 model: The Caco-2 model is the most common model for investigation of intestinal drug absorption. In a study with participation of five laboratories the permeability characteristics of a set of chosen drugs was determined under previously defined standard operating procedures to improve the possibility of interlaboratory comparison of results obtained with this model. The comparison showed a variation in absolute permeability coefficients, but all groups determined a common ranking in the permeability of the chosen drugs. In addition, altering expression of transport proteins, p-gp and MRP2, could be observed which seems to be the greatest problem in the standardisation of the Caco-2 model. 2. Influence of surfactants on absorption of drugs: Many drugs have a low oral bioavailabilty because their intestinal absorption is limited by the transport activity of export proteins. e.g. p-gp and MRP2. The influence of a range of surfactants on transport activity of both proteins was investigated using the Caco-2 model. Many surfactants caused an inhibition of p-gp, with poloxameres, e.g. Pluronic P85, showing the greatest effect. A few surfactants caused an inhibition of MRP2, hereby Cremophor RH40 and Solutol HS15 showed the most pronounced effect. After determination of the critical micellization concentration of surfactants it could be observed that the most effective surfactant concentration was in the range of the CMC suggesting inhibiting activity of single molecules rather than micelles. Investigation of influence of all surfactants on membrane fluidity showed a decrease of membrane fluidity. This might affect transport activity of p-gp and MRP2, but a difference between more or less active surfactants could not be determined. Surfactants did not cause any toxicity in effective concentrations as was shown in two toxicity assays. Absorption enhancement of drugs transported by p-gp and MRP2 seems to be possible by adding effective surfactants.
