Abstract

Papillomviren sind DNA-Viren, die in einer Vielzahl von Vertebratenspezies gefunden wurden. Während die meisten Infektionen inapparent verlaufen, gibt es einige humane Papillomvirus-Typen (HPV) mit onkogenem Potential. Von besonderer Bedeutung ist dabei die Assoziation mit dem Zervixkarzinom, der zweithäufigsten Krebsart bei Frauen. Das Kapsid der Papillomviren ist aus zwei verschiedenen Kapsidproteinen aufgebaut, dem Haupt-Kapsidprotein L1 sowie dem Neben-Kapsidprotein. Die Kapsidproteine schützen nicht nur die DNA in der extrazellulären Phase des Virus und ermöglichen die Bindung des Virus an die Wirtszelle - L2 scheint auch regulatorische Funktionen zu haben. L2 spielt möglicherweise eine entscheidende Rolle bei der DNA-Verpackung und der Reifung infektiöser Virionen. In der hier vorgelegten Arbeit wurden zelluläre Interaktionspartner von L2 identifiziert. Zu diesem Zweck wurde eine humane Keratinozyten-cDNA-Bibliothek mittels des Yeast Two Hybrid-Verfahrens nach Interaktionspartnern von HPV 11 L2 durchsucht. Auf diese Weise gefundene potentielle Interaktionspartner wurden auf eine Wechselwirkung mit den L2-Proteinen anderer HPV-Typen untersucht (HPV1, HPV16). Proteine, die dabei mit mehr als einem L2-Protein interagierten, wurden auf die Bindung von HPV 16 L2 in vitro getestet und ihre subzelluläre Lokalisation analysiert. Ein mit L2 in punktartigen Kerndomänen kolokalisierendes Protein, PLINP benannt, wurde u.a. durch die Herstellung von Deletionsmutanten eingehender untersucht. Es konnte so die L2-bindende Domäne sowie der für die Lokalisation in Kerndomänen verantwortliche Bereich eingegrenzt werden. Da für BPV 1 L2 eine Lokalisation in PODs, einer ebenfalls in einem Punktmuster vorliegenden Kerndomäne, beschrieben war, wurde die Lokalisation von L2 und PLINP im Verhältnis zu PODs untersucht. Eine Assoziation dieser beiden Proteine mit PODs scheint zwar vorzuliegen, nicht aber eine ausgeprägte Kolokalisation wie zuvor für BPV 1 L2 beschrieben.

Translation of abstract (English)

Papillomaviruses are a heterogenous family of DNA viruses, that were found in various vertebrate species. While most of the infections are inapparent, some Human Papillomaviruses (HPV) are oncogenic and are associated with the cervix carcinoma, the second most fatal cancer in women. The papillomavirus capsid is formed by two proteins, the major capsid protein L1 (360 copies) and the minor capsid protein L2 (12 copies). The capsid proteins do not only protect the viral DNA in the extracellular phase of the virus and allow binding to the host cell - especially L2 seems to have regulatory functions (e.g. interaction with the viral non-structural protein E2). L2 might have a role in DNA-packaging and maturation of infectious virions. In this work cellular interaction partners of the minor capsid protein L2 were identified. Therefore a human keratinocyte-cDNA-library was screened by yeast two hybrid screening, using HPV11 L2 as a bait. Potential interacting proteins were analyzed for their interaction with L2 proteins of different HPV-types (HPV1 L2 and HPV16 L2). Proteins interacting with more than one L2-protein were checked for their in vitro binding of HPV16 L2 and their subcellular localization was determined. One protein colocalizing with L2 in nuclear dots was named PLINP (papillomavirus L2 interacting nuclear protein). PLINP was analyzed in more detail e.g. by generation of deletion mutants. The L2-binding domain as well as the region responsible for (sub-)nuclear localization could be identified. As for BPV1 L2 a localization in PODs, a subnuclear domain with punctuate pattern, was observed, the localization of HPV16 L2 and PLINP to PODs was analyzed. The two proteins seem to be associated with PODs, but do not display a clear-cut colocalization as does BPV1 L2.
