Abstract
Die Kenntnis über die Mechanismen der Schmerzverarbeitung und ihre Veränderung durch Entzündungen spielt eine wichtige Rolle in der Entwicklung spezifischer Schmerzmedikamente. Schmerzreize werden von den Fasern spezialisierter Nervenzellen des peripheren Nervensystems, den Nozizeptoren detektiert, deren Zellkörper in den Spinalganglien liegen. Die Nozizeptoren selbst stellen somit die erste Verarbeitungsstelle für Schmerzreize dar. Die Erforschung der Beeinflussung ihrer Signaltransduktion durch Entzündungsmediatoren stand deshalb im Mittelpunkt dieser Arbeit. Hier ist es gelungen ein neues Modell auf zellulärer Basis zu entwickeln, das die Induzierung einer Entzündung innerhalb weniger Stunden ermöglicht und sowohl ex vivo als auch in vitro angewendet werden kann. Mit Hilfe des Modells ist es gelungen den Einfluss einer Entzündung auf die Aktivität und Expression verschiedener Transduktionskanäle zu untersuchen. Ebenso wurde die Rolle von Chlorid in der Signaltransduktion von Schmerzzellen analysiert. Es konnte gezeigt werden, dass Entzündngsmediatoren zu einer vertärkten Signaltransduktion und damit möglicherweise auch zu einer gesteigerten Schmerzwahrnehmung führen.
Translation of abstract (English)
The knowlegde about the mechanisms behind the transduction of pain signals and their modification by inflammational processes is important for the development of new specific drugs. Fibers of specialized neurons, so called nociceptors, detect painful stimuli. The cell bodies of these pain cells are located in the dorsal root ganglia. Hence nociceptors are the first site for signal processing, the investigation of inflammation induced impact on their signal transduction was the focus of this work. In this work a new inflammation model on cellular basis was established, which induces an inflammation already after hours. The model can be used ex vivo and in vitro. With this model it was possible to analyze the influence of inflammation mediators on the activity and expression of different transduction channels. Additionally the role of chloride in the signal transduction of nociceptors was investigated. The inflammation model induces an enhanced signal transduction which might lead to an intensified pain perception.
