Abstract

Die humanen beta-Papillomviren HPV5 und HPV8 infzieren spezifsch Keratinozyten der Haut. Beide Typen wurden mit der Entstehung von nicht-melanozytärem Hautkrebs (NMSC) in Verbindung gebracht. Über das zelltransformierende Potential der beta-HPV Onkogene E6 und E7 ist jedoch wenig bekannt. Das Ziel dieser Arbeit war, neue Interaktoren für E6 und E7 zu identifzieren und die Interaktion mit diesen Proteinen hinsichtlich ihrer Funktion und ihres Transformationspotentials zu untersuchen. Unter Verwendung des Yeast-two-hybrid-Systems und mittels GST-Pulldown wurden insgesamt 35 potentielle Interaktoren, davon 13 für E6 und 22 für E7, isoliert. Zwei der Interaktoren, NuMA und Myosin1C, wurden für eine nähere Analyse ausgewählt. Das Nuclear Mitotic Apparatus Protein 1 (NuMA) fokussiert, im Komplex mit Dynein, die Minusenden der Mikrotubuli an den Polen der mitotischen Spindel. Eine Disfunktion des NuMA/Dynein-Komplexes führt zur Defokussion der Spindel und Fehlausrichtung der Metaphasechromosomen. E6 und E7 von HPV5 und HPV8 und NuMA sind im Interphasenukleus kolokalisiert. Nur E7 assoziiert mit NuMA an den Polen der mitotischen Spindel. Diese Interaktion führt in E7-exprimierenden Keratinozyten weder zum Abbruch, noch zu einer messbaren Verlängerung der Mitosephase. Auch eine aus Zellteilungsfehlern resultierende Änderung der Ploidie konnte nicht festgestellt werden. In E6/E7-exprimierenden Zellen treten jedoch vermehrt Zellen mit überzähligen Zentrosomen auf, was auch auf die Rolle von NuMA in der Zentrosomentrennung zurückgeführt werden könnte. Es wird vermutet, dass die E7-NuMA-Assoziation während normaler Mitosen der Seiteneffekt einer primären Funktion in der asymmetrischen Teilung der Basalzellen ist, die der Replikation von HPV dient. NM1, eine nukleare Isoform von Myosin1C, ist in die Transkription ribosomaler Gene durch die RNA-Polymerase I (Pol I) involviert. Das E7-Protein von HPV8, nicht aber von HPV5, assoziiert mit NM1. Diese Bindung wurde durch Koimmunpräzipitation verifiziert. Expression von HPV8 E7 führt zu einer verminderten Pol I-Aktivität und reduziert damit die Synthese von prä-rRNA. Die Experimente zeigen eine Halbierung der prä-rRNa-Konzentration in E7-exprimierenden Zellen, die sich jedoch nicht maßgeblich auf die Mengen maturierter rRNAs auswirkt. Über die Interaktion mit NM1 kann HPV8 die energieaufwendigen Pol I-Transkriptionsvorgänge einschränken, um möglicherweise die in der Zelle vorhandenen Energierecourcen zum eigenen Vorteil zu verteilen. Die Schlussfolgerung aus den Untersuchungen dieser Arbeit ist, dass die Interaktion von E6 und/oder E7 mit NuMA oder NM1 zwar vermutlich nicht zur Transformation von Keratinozyten beiträgt, aber dennoch in Prozessen involviert ist, die für die Infektions- oder Replikationsmechanismen der beta-HPV bedeutend sind.

Translation of abstract (English)

The human beta-Papillomaviruses HPV5 and HPV8 specically infect keratinocytes of the skin. Both types have been associated with the development of non-melanoma skin cancers (NMSC). So far, little is known about the cell-transforming potential of beta-HPV and its oncogenes E6 and E7. The aim of this thesis was to identify new interaction partners of E6 and E7 and to investigate function and transformation potential of these protein interactions. A total of 35 potential interaction partners, 13 for E6 and 22 for E7, were identied using the yeast-two-hybrid system and GST pull-down. Two of these interaction partners, NuMA and Myosin1C, were analyzed further. In a complex with dynein, the Nuclear Mitotic Apparatus protein 1 (NuMA) is responsible for focusing the minus ends of microtubules at the poles of the mitotic spindle. A dysfunction of this NuMA/dynein-complex results in a defocused spindle and misalignment of metaphase chromosomes. Both E6 and E7 from HPV5 and HPV8 colocalize with NuMA in the nucleus during interphase. However, only E7 associates with NuMA at the poles of the mitotic spindle. In E7 expressing keratinocytes, these interactions neither lead to failure of nor cause a measurable prolongation of mitosis. Also, changes in ploidy due to errors in cell division were not observed. Still, in cell cultures expressing E6/E7 an increased percentage of cells contained abnormal numbers of centrosomes, which could result from the implication of NuMA during centrosome segregation. It has been proposed that the association of E7 with NuMA during normal mitosis is a side effct of a primary function in the asymmetric division of basal cells, which supports the replication of HPV. NM1, a nuclear isoform of myosin1C, is involved in the transcription of ribosomal genes via RNA-Polymerase I (Pol I). The E7 protein from HPV8, but not from HPV5, associates with NM1. This interaction was verified via co-immunoprecipitation. Expression of HPV8 E7 leads to decreased activity of Pol I and thereby reduces the synthesis of pre-rRNA. The experiments demonstrated that the expression of E7 halves pre-rRNA levels in cells, but does not lead to comparable reductions in mature rRNAs. It is possible, that by limiting the energy consuming Pol I transcription processes via interaction with NM1, HPV8 aims to reallocate the cell's energy resources to its advantage. The conclusions resulting from this work are that the interactions of E6 and/or E7 with NuMA or NM1 probably do not contribute to the transformation of keratinocytes. However, these interactions affct processes that are important for beta-HPV infection or replication mechanisms.
