Abstract

Das SGTA-Protein ist evolutionär konserviert und wird ubiquitär exprimiert. Wie seine Isoform SGTB bindet es an das Chaperon HSP70/HSC70 und beeinflusst dessen Proteinfaltungsfunktion. SGTA ist essenziell für die Zellteilung von Tumorzellen und könnte ein „synthetic lethal target“ der Krebstherapie darstellen. Zudem ist SGTA ein zellulärer Interaktionspartner des NS1-Proteins der autonomen Parvoviren. Durch die Untersuchung von Mausgeweben, humanen Kulturzellen sowie mouse embryonic fibroblasts (MEFs) aus wildtyp und SGTA und –B doppelknockout Mäusen wurden in dieser Arbeit SGTA und –B funktional verglichen und initiale Experimente zur Validierung von SGTA als Zielmolekül für die Krebstherapie durchgeführt. Darüber hinaus wurde untersucht, ob SGTA einen Beitrag zum parvoviralen Lebenszyklus leistet.

Translation of abstract (English)

The SGTA protein is evolutionary conserved and ubiquitously expressed. Like its isoform SGTB, it binds to the chaperone HSP70/HSC70 and influences its protein folding ability. SGTA is essential for cell division in tumor cells and may represent a „synthetic lethal target“ for cancer therapy. Moreover, SGTA is a cellular interaction partner of the autonomous parvovirus NS1 protein. The thesis presented here aimed at comparing SGTA and –B functionally through analysis of mouse tissues, human cell lines as well as mouse embryonic fibroblasts (MEFs) from wildtype and SGTA and –B double knockout mice. In addition, initial experiments for the validation of SGTA as a target for cancer therapy were conducted. Finally, it was examined if SGTA contributes to the parvoviral life cycle.
