Abstract

Der Granulozyten Kolonie-stimulierende Faktor (G-CSF) ist allgemein als hämatopoetischer Wachstumsfaktor bekannt. Vor einigen Jahren wurde ihm neben dieser bekannten Rolle ein zusätzliches, neues Wirkungsspektrum als neuronaler Wachstumsfaktor im Zentralen Nervensystem (ZNS) zugeschrieben. Da chronische, neurologische Erkrankungen eine lebenslange, medikamentöse Behandlung erfordern, wirken sich die mäßige Halbwertszeit und die erheblichen Produktionskosten im Fall von G-CSF nachteilig aus. Pharmakologische Weiterentwicklungen von Proteinen stellen agonistische Peptidliganden dar, wie sie für eine Vielzahl von Wachstumsfaktor-Rezeptoren identifiziert wurden. Aus diesem Grund wurde ein Screening für agonistisch wirkende G-CSF-Peptide, abgeleitet von der Sequenz des humanen G-CSF, durchgeführt. Trotz der hohen Sensitivität des zellbasierten Assays konnte keine agonistische Aktivität der 20 mer Peptide allein oder in einem Multiplex-Ansatz detektiert werden. Möglicherweise sind größere, zusammenhängende Teile des Proteins oder dimerisierende Peptide nötig, um eine Aktivierung des Rezeptors zu erreichen. Neben seinen neuroprotektiven und pro-regenerativen Effekten wurde untersucht, ob diese Wirkung von G-CSF auch auf Schwannzellen und periphere Demyelinisierungen übertragbar ist. Als Modell für periphere Neuropathien wurde die Charcot-Marie-Tooth 1A (CMT1A) Krankheit ausgewählt. Bei dieser Erkrankung handelt es sich um eine der am häufigsten auftretenden hereditären Neuropathien, für die es bis zum heutigen Zeitpunkt keine effektive Behandlung gibt. Der G-CSF Rezeptor wurde von der Schwannzelllinie RSC96 exprimiert und G-CSF aktivierte in der selbigen anti-apoptotische Signalwege. Trembler-J Mäuse, ein Tiermodell für CMT1A, wurden daher mit G-CSF (30 mg/kg/Tag mit Hilfe von osmotischen Pumpen abgegeben) für drei Monate behandelt und unterliefen einer Anzahl von Verhaltenstests. Trotz der Aktivierung anti-apoptotischer Signalwege in vitro konnten keine signifikanten Unterschiede zwischen der behandelten und unbehandelten Tiergruppe in vivo beobachtet werden. G-CSF veränderte bei Konzentrationen, die in anderen Modellen für neurologische Erkrankungen verwendet wurden, nicht den Krankheitsverlauf der Trembler-J Tiere, weswegen angenommen werden muss, dass G-CSF kein offenkundiger Kandidat für die Behandlung von CMT1A und möglicherweise auch von peripheren Neuropathien ist.

Translation of abstract (English)

Granulocyte-colony stimulating factor (G-CSF) is commonly known as hematopoetic growth factor. Some years ago an additional, novel impact as neuronal growth factor in the central nervous system (CNS) was put down to its known role. As chronic diseases require a lifelong, medicinal care, the moderate plasma half-life time and the considerable production costs in the case of G-CSF do affect adversely. Pharmacological further developments of proteins display agonistic peptide ligands like described for a number of growth factor receptors. Because of this reason a screen for peptide agonists derived from the sequence of human G-CSF was conducted. Despite the high sensitivity of the cell based assay no agonistic activity of 20 mer peptides alone or in multiplex approaches could be detected. One concludes that larger coherent parts of the protein or dimerising peptides may be needed to achieve activation of the receptor. Besides its neuroprotective and pro-regenerative effects it was analysed, if this impact of G-CSF could be conferred on Schwann cells and peripheral demyelinisations. As a model for peripheral neuropathies Charcot-Marie-Tooth 1A (CMT1A) disease was chosen. This disease is one of the most common hereditary neuropathies with no effective treatment available yet. The G-CSF receptor was expressed by the RSC96 Schwann cell line, and G-CSF evoked anti- apoptotic pathways in the cell line. Trembler-J mice, an animal model for CMT1A, were therefore treated with G-CSF (30 mg/kg/day delivered by subcutaneous pumps) for three months and followed by a number of behavioural tests. In spite of the activation of anti- apoptotic pathways in vitro, no significant differences between treated and untreated animal groups could be observed in vivo. One concludes that G-CSF at concentrations that have shown effects in other models of chronic neurological conditions did not alter the disease course of the Trembler-J model, and therefore does not appear as an evident drug candidate for CMT1A and potentially peripheral neuropathies.
