Abstract

Die N-terminale Acetylierung (NTA) gehört zu den häufigsten Proteinmodifikationen in Eukaryoten. Die Funktion der NTA ist vielfältig und vom jeweiligen Protein abhängig. Bisher wurde belegt, dass die N-terminale Modifikation die Funktion und Stabilität eines Proteins sowie Protein-Protein Interaktion beeinflussen kann. Im Rahmen dieser Arbeit sollte untersucht werden, ob in der Modellpflanze A. thaliana Proteine N-terminal acetyliert werden. Ein Homologievergleich mit den aus Hefe bekannten Nat-Komplexen (NatA, NatB und NatE) identifizierte folgende Kandidatenproteine: AtNAA10p (At5g13780) und AtNAA15p (At1g80410.1) für den NatA-, AtNAA20p (At1g03150) für den NatB- und AtNAA50p (At5g11340) für den NatE-Komplex. Die enzymatische Charakterisierung der rekombinant exprimierten Enzyme AtNAA10 und AtNAA50, bestätigte die auf Proteinsequenz basierte Vorhersage der enzymatischen Aktivität. Die transiente Expression von AtNAA10p, AtNAA15p und AtNAA50p in Fusion mit fluoreszierenden Marker-Proteinen in Tabakzellen zeigte die erwartete Lokalisierung im Cytosol und Zellkern. Überaschenderweise wurde das Fusionsprotein AtNAA20:EYFP neben dem Cytosol auch in Mitochondrien in Arabidopsis detektiert, was auf den funktionellen Unterschied zwischen AtNAA20p und den orthologen Proteinen in Hefe und Menschen hindeutet, die im Cytosol vorkommen. Das HsNaa20p wird auch im Zellkern exprimiert. Die Identifikation einer T-DNA-Insertionslinie (naa20-1) für AtNAA20 ermöglichte die Bestätigung der katalytischen Funktion von AtNAA20p in vivo. Die hier gezeigten Ergebnisse deuten ein vergleichbares Substratspektrums des NatB-Komplexes aus Hefe, Menschen und höheren Pflanzen an. Homozygote Pflanzen beider identifizierten T-DNA-Insertionslinien für AtNAA20 waren deutlich im Wachstum gehemmt, durchliefen aber eine normale Entwicklung, die mit der Produktion von vitalem Saatgut endete. Im Gegensatz hierzu führte der knock out der NAA50p nach erfolgreicher Keimung zu einer anormalen Entwicklung während des vegetativen Wachstums. Die homozygoten Pflanzen sind auch nicht fertil. Ein Verlust der NAA10 oder NAA15 Funktion führt zu einem Abort der Embryogenese im globulärem Stadium, das sich durch die erstmalige Differenzierung von Organen auszeichnet. Es konnte somit gezeigt werden, das NTA von Proteinen durch die Nat-Komplexe A, B und E nicht nur in Hefe und Menschen sondern auch in Pflanzen konserviert ist, wobei sich funktionelle Alleinstellungsmerkmale der NTA in planta andeuten. Des Weiteren konnte belegt werden, dass die N-terminale Acetylierung von Proteinen essentiell für die Entwicklung von höheren Pflanzen ist.

Translation of abstract (English)

The N-terminal acetylation (NTA) is one of the most common modifications of eukaryotic proteins. The function of the NTA is manifold and dependent on the respective protein. It was already indicated that the N-terminal acetylation may affect biological function and stability of a protein or protein-protein interaction. The present study investigated whether some proteins will be acetylated in the model plant Arabidopsis thaliana. The search for orthologous proteins of Nat complexes in Arabidopsis, which were known in yeast and human, revealed the following candidates: AtNAA10p (At5g13780) and AtNAA15p (At1g80410.1) for the NatA, AtNAA20p (At1g03150) for the NatB and AtNAA50p (At5g11340) for the NatE complex. Based on the protein sequence, the N-acetyltransferase activity for the NAA10 and NAA50 proteins was predicted. The Expression of the N-acetyltransferase activity was confirmed using acetylation assay with recombinant proteins, respectively. In order to identify subcellular localisation of the AtNAA10, AtNAA15 and AtNAA50 proteins a fluorescent protein fusion was used. The transient expression in tobacco cells revealed the expected cytosolic and nucleus localisation. Surprisingly, the fusion protein AtNAA20:EYFP was detected in the cytosol as well as in the mitochondria in Arabidopsis. This suggests the functional differences between AtNAA20p and the orthologous proteins from yeast and human, which were localised in the cytosol. The HsNaa20p was expressed in the nucleus too. The Identification of a T-DNA insertion line (naa20-1) for AtNAA20 allowed the confirmation of the catalytic function of AtNAA20p in vivo. The present results showed a comparable substrate spectrum of the NatB complexes in yeast, human and higher plants. Homozygous plants of both T-DNA insertion lines for AtNAA20 showed retarded growth, but developed normally and produced seeds. Except for effective Germination showed the knock out of the NAA50p in contrast abnormal development during vegetative phase. Homozygous plants are also not fertile. A loss of the NAA10 or NAA15 function induced abort of embryogenesis in the globular stage; a stage characterized by the first-time differentiation of organs. We were able to demonstrate that the NTA of proteins of Nat complexes A, B and E is conserved not only in yeast and human but also in higher plants. However, these present results suggest some unique functional differences of NTA in planta. Furthermore, this study indicated that NTA is essential for development of higher plants.
