Abstract

Das humane T-lymphotrope Virus-1 (HTLV-1) ist der ätiologische Erreger der adulten T-Zell Leukämie (ATL), einer neoplastischen Erkrankung von T-Lymphozyten. Das virale Onkoprotein Tax spielt eine bedeutende Rolle bei der malignen Transformation der CD4+-Lymphozyten. Ein charakteristisches Merkmal HTLV-1-infizierter Zellen und ATL Zellen ist die Resistenz gegen Chemotherapeutika und andere Apoptosestimuli, was die Behandlungsmöglichkeiten einer akuten ATL begrenzt und eine schlechte Prognose zur Folge hat. In dieser Studie wurde der molekulare Mechanismus der Tax-vermittelten Apoptoseresistenz HTLV-1-infizierter Zellen untersucht. In diesem Zusammenhang wurde die Bedeutung anti-apoptotischer Proteine bereits gut charakterisiert. Dagegen ist über die Bedeutung der pro-apoptotischen Proteine für die Regulierung der Apoptoseresistenz HTLV-1-infizierter Zellen nur wenig bekannt. Analysiert wurde daher der Einfluss des HTLV-1 Tax Proteins auf die Expression pro-apoptotischer Bcl-2 Familienmitglieder. Hierbei wurde gezeigt, dass ATL Zellen ein vermindertes Niveau der BH3-only Proteine Bid und Bim exprimieren. Untersuchungen des molekularen Mechanismus konnten zeigen, dass die Expression des viralen Onkoproteins Tax mit der verminderten Expression von Bid und Bim korreliert. Zudem induziert Tax die Überexpression des Transkriptionsfaktors HIF-1alpha (Hypoxie-induzierter Faktor), der als Repressor die Transkription von Bid und Bim inhibiert. Die Relevanz der BH3-only Proteine für die Apoptoseresistenz wurde mit Hilfe eines siRNA-vermittelten knock downs von Bid in Jurkat-Zellen, was sowohl zu einer verminderten Rezeptor- als auch Mitochondrien-induzierten Apoptose führte, nachgewiesen. Dagegen führte die ektopische Expression von Bid in HTLV-1-infizierten Zellen zu einer Erhöhung der Apoptosesensitivität. Zudem konnte eine pharmakologische Inhibition von HIF-1alpha sowohl die Expression von Bid und Bim als auch die Sensitivität der HTLV-1-infizierten Zellen für Apoptose wiederherstellen. Die Ergebnisse dieser Arbeit zeigen erstmals die Bedeutung pro-apoptotischer Proteine für die HTLV-1-vermittelte Apoptoseresistenz auf. Darüber hinaus könnte die Wiederherstellung der Bid und Bim Expression, z. B. durch HIF-1alpha Inhibitoren, eine vielversprechende Strategie für die Behandlung HTLV-1-infizierter ATL-Patienten darstellen.

Translation of abstract (English)

The human T-lymphotropic virus 1 (HTLV-1) is the etiological agent of adult T-cell leukemia (ATL), an aggressive T-cell malignancy caused by viral oncoprotein Tax-mediated malignant transformation of CD4+ T-cells. One characteristic feature of HTLV-1-infected cells and ATL cells is their resistance towards chemotherapeutics and other apoptotic stimuli, complicating treatment and contributing to poor prognosis. In this study, the molecular mechanism by which HTLV-1 Tax confers resistance towards apoptosis was investigated. The role of anti-apoptotic proteins in this context has been well studied. However, little is known about the contribution of pro-apoptotic Bcl-2 proteins in regulation of apoptosis resistance in HTLV-1-infected cells. Therefore, the effect of HTLV-1 Tax protein on expression of pro-apoptotic Bcl-2 family proteins was investigated. HTLV-1-infected ATL cells were found to express reduced levels of Bid and Bim. Investigation of the molecular mechanisms by which HTLV-1 down-regulates Bid and Bim expression revealed that the viral oncoprotein Tax is the causative factor for the diminished expression of Bid and Bim. In addition, Tax induces overexpression of the transcription factor HIF-1alpha which, in turn, functions as a transcriptional suppressor of Bid and Bim. The importance of BH3-only proteins for apoptosis resistance was evidenced by knockdown of Bid by means of siRNA in leukemic Jurkat T-cells which resulted in a reduction in receptor- as well as mitochondria-mediated apoptosis, whereas ectopic expression of Bid in HTLV-1-infected cells sensitized towards apoptosis. Furthermore, pharmacological inhibition of HIF-1alpha was shown to restore expression of Bid and Bim and, consequently, sensitivity of HTLV-1-infected cells towards apoptosis. Collectively, this study provides the first evidence for the involvement of the pro-apoptotic proteins Bid and Bim in HTLV-1-mediated apoptosis resistance. Moreover, these findings suggest that enhancing Bid and Bim expression, e.g. by HIF-1alpha inhibitors, may be a promising approach for treatment of HTLV-1-associated ATL.
