Abstract

Die vorliegenden Studie beschäftigt sich mit der Untersuchungen von ausgewählten Stoff-wechselenzymen zweier einheimischer Fischarten in Leber und Gehirn als Biomarker zur Erfassung von Umweltgiften. Zwischen 1995 und 2000 wurden im Großraum Stuttgart die unterschiedlich stark belasteten Fließgewässer Krähenbach/Aich und Körsch beprobt. Die enzymatischen Daten wurden zu ultrastrukturellen, pathologischen Daten sowie zu Ergebnis-sen der Biotransformation in Beziehung gesetzt. Beide Gewässer konnten klar nach ihrem Belastungszustand differenziert werden. Um die Aussagefähigkeit der Markerantworten aus den Bypassexpositionen zu bewerten wurden zwischen 1998 - 1999 Ergebnisse des passiven und aktiven Biomonitoring verglichen. Parallel zu den Halbfreilanduntersuchungen aus dem Freiland entnommenen Bachforellen und Bachschmerlen zeigten Übereinstimmungen von 61,4 bzw. 71,8 Zur Charakterisierung des Zeitfensters in dem sich Induktionsantworten bei Biomarkern ma-nifestieren, wurden Bachforellen und Bachschmerlen in einem chronischen Belastungsexpe-riment einem ausgewählten PAH-Cocktail exponiert. Nach einem zusätzlichen Be-lastungspeak erwiesen die Stoffwechselenzyme als schnell integrierende und regenerierende Marker. 48 h nach dem Belastungspeak waren erhöhte Induktionsantworten der Stoffwechsel-enzyme nachzuweisen, die mit adaptiven ultrastrukturellen Veränderungen zu korrelieren waren. In Early Life Stage-Untersuchungen mit dem Zebrabärbling wurden teratogene Wirkungen des PAH-Cocktail nachgewiesen werden, wobei die Stoffwechselenzyme in einem vergleich-baren Sensitivitätsbereich mit wichtigen biologischen Endpunkten der Embryotoxikologie wie etwa Behinderung, Missbildung oder Letalität anzusiedeln sind. Die Stoffwechselenzyme stellen sensitive und schnell integrierende Marker dar, die in Kom-bination mit ultrastrukturellen und histopathologischen Biomarkern einen wichtigen Anteil bei der Charakterisierung von Umweltgiften in unseren Gewässern liefern können.

Translation of abstract (English)

The presented study is part of a co-operation research project and is concerned with investigations of selected metabolic enzymes in liver and brain of two fish species, brown trout and loach, in biomarker research to determine environmental contamination. In the area of Stuttgart (Germany) two differently polluted small rivers Krähenbach/Aich and Körsch had been investigated between 1995 and 2000. Fish were exposed under semi-field conditions and enzymatic data were correlated to further biochemical, ultrastructural and pathological parameter. Both rivers could clearly be discriminated according to their chemical load. To evaluate the sensitivity of the biomarker response, data of semi-field (active biomonitoring) and field (passive biomonitoring) exposure were compared in 1998 and 1999. Trout and loach kept under semi-field conditions and fish caught in the field showed a correspondence of 61,4 and 71,8 , respectively. In order to eludicate the induction and duration of a biomarker response after chronic exposure followed by an additional exposure-peak, trout and loach were exposed to a selected PAH-cocktail. The results indicated that the metabolic enzymes represent fast integrating and regenerating markers, which could be correlated to changes in liver ultrastructure. The PAH-cocktail was further applied in early life stage tests with the zebrafish to investigate possible teratogenic effects and to correlate the results to the range of sensitivity of the metabolic enzymes. The metabolic enzymes represent sensitive and fast integrating biomarkers, which in combination with ultrastructural and histopathological parameter deliver an important contribution in the risk assessment of environmental contamination.
