Abstract

Dynamine sind große GTPasen, die in der Zelle an wichtigen Prozessen wie der Endocytose, dem Vesikeltransport, der Morphologieerhaltung und Verteilung von Mitochondrien u.v.m. beteiligt sind. Welche Rolle die Dynamine hierbei im Detail spielen, ist jedoch noch ungeklärt. Im Rahmen dieser Arbeit wurde ein neues Mitglied der Dynaminfamilie aus dem Modellorganismus Dictyostelium discoideum identifiziert und charakterisiert. Dynamin B hat eine berechnete Molekularmasse von 105 kDa. Es besitzt eine für Dynamine typische Domänenstruktur mit einer ca. 300 Aminosäuren umfassenden GTPase-Domäne und einem weniger gut konservierten carboxyterminalen Bereich von ca. 500 Aminosäuren. Zusätzlich besitzt es eine Extension von 136 Aminosäuren am Aminoterminus. Es konnte gezeigt werden, daß die Extension ein mitochondriales Signalpeptid darstellt. Zum einen wird die Extension an einer vorhergesagten Schnittstelle (R-2-Motiv) posttranslational abgespalten. Zum anderen ist sie für die Lokalisation des Proteins an den Mitochondrien notwendig: Dynamin B, das am Carboxyterminus mit YFP markiert wurde, kolokalisiert in hohem Maß mit den Mitochondrien, im Gegensatz zu Dynamin B-YFP, dem die Extension fehlt. Die Immun-Elektronenmikroskopie ergab darüber hinaus, daß das Protein mit der äußeren Mitochondrienmembran assoziiert ist und außerdem mit der Plasmamembran. Durch gezielte Geninaktivierung wurden Dictyostelium-Zellen hergestellt, die Dynamin B nicht mehr produzieren. Die Dynamin B-Nullzellen sind bezüglich Zellmorphologie, Wachstum, Entwicklung, Endocytose, Funktion und Morphologie der Mitochondrien nicht von den Wildtypzellen zu unterscheiden. Sie zeigen jedoch eine dreifach erhöhte Adhäsion gegenüber Oberflächen und anderen Zellen. Möglicherweise wird Dynamin B in die Mitochondrienmatrix importiert, dort prozessiert und anschließend wieder exportiert um zum eigentlichen Wirkort (Plasmamembran, äußere Mitochondrienmembran) zu gelangen.

Translation of abstract (English)

Dynamins are high-molecular weight GTPases which are involved in many important processes such as endocytosis, vesicular transport, maintenance of mitochondrial morphology and distribution of mitochondria. However, the precise role of dynamins in these processes is still unclear. This work deals with the identification and characterization of a new member of the dynamin family from the model organism Dictyostelium discoideum. Dynamin B has a calculated molecular mass of 105 kDa. Its domain structure resembles that of other dynamins. It contains a GTPase domain of 300 amino acids and a less well conserved region of about 500 amino acids at the carboxyterminus. In addition it has an extension of 136 amino acids at the aminoterminus which was shown to be a mitochondrial signal peptide. This extension is posttranslationally processed at a predicted cleavage site (R-2-motif). Furthermore, C-terminally YFP-labelled dynamin B colocalizes with mitochondria to a high degree whereas dynamin B without the extension does not. Immuno-electron microscopy showed in more detail that dynamin B is associated with the outer mitochondrial membrane and, additionally, with the plasma membrane. Dynamin B null cells were generated by gene targeting to study the function of the protein. The dynamin B null cells do not show any obvious alterations concerning cell morphology, growth, development, endocytosis and function and morphology of mitochondria. However, they exhibit a threefold increase in adhesion on surfaces and towards other cells. Therefore dynamin B might be imported into the mitochondrial matrix, be processed and then exported and transported to the site of action (plasmamembrane, outer mitochondrial membrane).
