Abstract

Integrierte Sedimentuntersuchungen bieten durch den kombinierten Einsatz von Bioassays, chemischer Analytik und Bestandaufnahmen in situ umfassende Informationen zum Schädigungspotential von Sedimenten, überprüfen zugleich die ökologische Relevanz der Laboruntersuchungen, geben aber keine Informationen über die biologisch wirksamen Substanzklassen. Mittels Bioassay-dirigierten Fraktionierungstechniken können problematische Substanzklassen identifiziert werden. Der direkte Transfer der Ergebnisse auf das Ökosystem ist jedoch problematisch. Daher wird in diesem Projekt versucht, beide Konzepte miteinander zu verbinden, da für eine umfassende Risikoabschätzung sowohl die Überprüfung der ökologischen Relevanz von Laborexperimenten als auch eine Identifizierung der biologisch wirksamen Fremdstoffe notwendig ist. Es werden (1) das Untersuchungskonzept, das Biotests zur akuten Toxizität, dioxinähnlichen, genotoxischen und endokrinen Wirksamkeit beinhaltet, (2) Befunde der integrierten Untersuchungen und (3) Ansätze zu einer Bewertung vorgestellt. Bei einer Flachwasserzone am Neckar erwies sich insbesondere der Fischeitest (mit Danio rerio) mit den Expositionspfaden natives Sediment, Porenwasser und organischer Extrakt als geeignet, das ökotoxikologische Schädigungspotential zu charakterisieren und Hinweise zur Bioverfügbarkeit der partikulär gebundenen Schadstoffe für Fische zu geben. Mittels Bioassay-dirigierten Fraktionierungstechniken (Flüssig/Flüssig-Fraktionierung und säulenchromatographische Auftrennung mit Aluminiumoxid und HPLC) konnten am Beispiel eines mit Klinikabwässern belasteten Fließgewässers weiterführende Informationen zur Identifikation jener Substanzen gewonnen werden, die im Bioassay eine mutagene Wirkung hervorriefen. Die Ergebnisse einer vergleichenden Bewertung werden mithilfe eines einfachen ortsabhängigen Klassifikationssystems für kontaminierte Fließgewässer vorgestellt und einer Bewertung auf der Basis von Fuzzy Logic gegenübergestellt.

Translation of abstract (English)

The Sediment Quality Triad (SQT) approach with toxicity tests, chemical analyses and community structure studies provides comprehensive insight into the ecological state of sediments. This approach elucidates the ecological relevance of toxicity tests and chemical analyses, but does not allow an identification of the harmful substances. In contrast, effect-directed fractionation procedures may help to identify noxious substances, however, without considering their bioavailibilty. This study was designed to apply the triad approach in combination with bioassay-directed fractionation methods in order to allow an integrated risk assessment, which simultaneously includes a revelation of the ecological relevance of in vitro results and the identification of the harmful substances. The assessment strategy based on bioassays for the determination of acute toxicity, endocrine potential, dioxin-like activity and genotoxicity is presented. Furthermore, results of the SQT and an attempt to comprehensively evaluate the results are illustrated. At a newly designed fish spawning site close to Eberbach, early life-stage tests with Danio rerio and the exposure routes whole sediment, pore water and organic extract allowed a closer characterization of the damage potential. Moreover, lipophilic extracts of a sediment site close to a wastewater treatment plant of a hospital were separated by means of fluid/fluid extraction according to pH, column chromatography and HPLC into different fractions. Mutagenic potentials of the fractions were determined in order to identify the harmful substances. Furthermore, a site-dependent classification system for contaminated streams is compared to a model using fuzzy-logic.
