Abstract

Das spaghetti-Gen (spag) von Drosophila melanogaster ist in der chromosomalen Region 60B lokalisiert und kodiert für ein 534 Aminosäuren langes Protein, welches eine TPR-Domäne, bestehend aus drei TPR-Motiven, aufweist. Homozygot spaghetti-mutierte (spag-) Tiere sind nicht in der Lage, sich zu verpuppen. Sie sterben als überalte Larven des dritten Stadiums. Diese Larven zeigen melanotische Pseudotumore, ihre Imaginalscheiben sind atrophisch und die Lymphdrüsen vergrößert. Untersuchungen des Wachstums der Imaginalscheiben der homozygot spag--Larven durch Färbung mit Acridinorange und TUNEL-'assays' ergaben eine hohe Anzahl apoptotischer Zellen. Die Apoptose kann partiell durch ektopische Expression des Baculovirus p35-anti-apoptotischen Faktors verhindert werden. Das Spaghetti-Protein ist predominant in den Embryonen nachweisbar. Western-Blot Analysen von Kernextrakt sowie Immunfluoreszenzaufnahmen von Kernen der Embryonen zeigen, dass das Spaghetti-Protein neben seiner Lokalisierung im Zytoplasma eine Komponente des Zellkerns ist. Das Hitzeschockprotein Hsp90 interagiert mit dem Spaghetti-Protein sowohl im yeast two-hybrid System als auch im GST-'binding' System TPR-Domänen abhängig. Ektopische Expression des Spaghetti-Proteins führt zu Formationen von Extravenen in den Flügeln der Fliege sowie zu abnormaler Differenzierung der scutellaren bristless. Eine genetische Mosaik-Analyse von spag--defizienten Klonen ergab, dass spag--Klone selten auftreten und eine elongierte Form in den Flügeln annehmen. Frühe Induktion der spag-- Klone führt zu massivem Gewebeverlust von Thorax, Augen und Flügeln, wobei insoweit ein nicht-autonomer Effekt beobachtet werden kann, als in der Umgebung vermehrt Zellteilungen stattfinden. Daher kann geschlussfolgert werden, dass das Spaghetti-Protein für das Überleben der Zellen der Imaginalscheiben notwendig, und an der Bildung morphologischer Strukturen der Imaginalscheiben beteiligt ist.

Translation of abstract (English)

A P-Element inserted at position 60B was found to cause lethality of third instar larvae with imaginal disc atrophy, moderate overgrowth of the hematopoietic organs and formation of melanotic pseudo-tumours. This P-Element is inserted in the spaghetti (spag) gene, designated by virtue of the elongated shape of spag- clones present on wings of genetic mosaics. Immuno-fluorescence studies of imaginal wing discs in spag- larvae with TUNEL, Acridinorange or anti-ssDNA antibodies revealed a high rate of apoptosis in the wing discs showing that spag function is required during imaginal disc growth. Ectopic expression of the Baculovirus p35 anti-apoptotic protein was able to partially rescue the structure of the spag- discs. The spag gene encodes a protein of 534 amino acids with three tetratricopeptid repeats (TPR). Analysis with the yeast two-hybrid system showed that HSP90 and the product of the CG5792 gene, a member of the TonB-dependent-receptor-protein family interacts with Spag. To this family of the TonB-dependent-receptor-proteins also belongs the small heatshock proteins Hsp23, Hsp26 and Hsp27 of Drosophila melanogaster. Moreover GST-pull-down assay confirmed that Spag and HSP90 could bind to each other through the TPR-domain, whereas the binding of Spag to the TonB-dependent-receptor-protein does not require the TPR-domain. Mosaics studies showed that spag is required during the development of the imaginal discs, particularly in the hinge region of the wing discs as indicated by strong abnormalities detected in the thorax. Altogether these results indicated that Spag could directly associate with HSP90. Both proteins should participate to a large chaperone complex that is required for imaginal disc cell growth and differentiation.
