Abstract

Es wurde eine Multimethode zur Bestimmung von verschiedenen endokrin wirksamen Substanzen (EWS) in Wasser erarbeitet. Mit einem Analysengang können Steroidhormone, Alkylphenole, Bisphenol A, Diethystilbestrol, Hexestrol, Phthalate, PCB und mehrere Pestizide zusammen bestimmt werden. Die Festphasenextraktion, die in dieser Arbeit entwickelte On-Line-Derivatisierung, die Kapillar-Gaschromatographie und die Ion-Trap-Massenspektrometrie wurden verwendet. Niedrige Nachweisgrenzen und gute Wiederfindungen wurden erreicht. Die Gehalte an EWS wurden in verschiedenen Fließgewässern in Südwestdeutschland, während eines Hochwasserereignisses des Neckars sowie in Wasser eines größeren Wasserversorgungsunternehmens quantifiziert. Der Vergleich mit einem Biotest zeigte eine Korrelation zwischen den Konzentrationen an EWS und den Messergebnissen des Biotests. Auch Sedimente wurden untersucht und einige dieser Kontaminanten dort in höheren Konzentrationen gefunden. Die gewonnenen Daten sind plausibel und vergleichbar mit Literaturdaten. Nach derzeitigem Stand der Kenntnisse bergen die Gehalte des Trinkwassers an EWS keine konkrete Gefahr für die menschliche Reproduktionsfähigkeit. Die gemessenen oder der Literatur entnommenen Konzentrationen an Estradiol, Ethinylestradiol, Nonylphenol, Phthalaten, Bisphenol A und DDT in den Oberflächengewässern sind jedoch stellenweise in der Lage, endokrine Effekte in Fischen und anderen aquatischen Organismen zu bewirken. Zusätzlich wurden Literaturdaten von anthropogenen EWS in der Nahrung den natürlichen Gehalten an Phytoestrogenen in Lebensmitteln sowie der endogenen Produktion an Östrogenen gegenübergestellt. Demnach kann aus dem Gehalt der Nahrung an anthropogenen EWS weder eine Gefährdung der menschlichen Reproduktionsfähigkeit noch ein Zusammenhang mit dem Anstieg an genitalen Missbildungen gefolgert werden.

Translation of abstract (English)

A multimethod for the determination of endocrine disrupting compounds (EDC) of different chemical structures was developed. Steroid hormones, alkylphenoles, bisphenol A, diethystilbestrol, hexestrol, phthalates, PCB and several pesticides have been determined together in one analytical process. Techniques used consist of: solid phase extraction with self-made cartridges, the automatic on-line-derivatization (silylation), developed in this study, capillary gaschromatography and ion-trap mass sprectrometry. LOD´s in the low to mid ng/l range and good to satisfactory recovery rates were achieved. The concentrations of EDC were determined in several surface waters in southwestern Germany, a flood event of the Neckar river and in water of a major water municipal supply company. The results showed a good correlation to those of a biotest, based on the production vitellogenine-mRNA. The sediments were also examined and some of the contaminants were found in higher concentrations. All results were plausible and consistent with literature data. Based on the current knowledge, no concrete danger for human reproduction can be expected from the analytical results of this work or from results of the cited literature. Nevertheless, the determined concentrations of estradiol, ethynylestradiol, nonylphenol, phthalates, bisphenol A and DDT in surface waters presented in this work and by other authors may induce endocrine disruption in fish and other aquatic organisms. Finally, literature data of unintentional intake of anthropogenic EDC in food was studied and compared to natural contents of EDC in food (phytoestrogens) and the human endogenous production of estrogenes. Thus, neither a hazard for human reproduction nor a correlation with the increasing rate of genital malformations can be expected from the content of anthropogenic EDC in food.
