Abstract

Im Rahmen dieser Doktorarbeit wurden Untersuchungen zu Mutation, Expression und Regulation des Tumorsuppressor-Kandidaten APM-1 durchgeführt. APM-1 wird infolge der Integration von DNA des humanen Papillomvirus Typ 68 (HPV68) in den chromosomalen Genlocus 18q21 der Zervixkarzinom-Zelllinie ME180 aberrant als Bestandteil viral-zellulärer Hybrid-mRNAs transkribiert. Die zellulären APM-1-Transkripte bestehen aus identischen proteincodierenden Anteilen und unterschiedlichen 5’-untranslatierten Regionen (UTRs) mit der Bezeichnung p2, a31 und 15C, was für eine separate Transkriptionsinitiation an drei verschiedenen Promotoren spricht. Das APM-1-Gen codiert für ein Produkt, das mit seiner N-terminalen BTB-Domäne sowie vier Zinkfingern des Krüppel-Typs möglicherweise als Transkriptionsfaktor fungiert. Für funktionelle Studien zur Wachstumsinhibition wurde unter Verwendung des Tet-On-Genexpressionssystems versucht, HeLa-Zellen mit induzierbarer APM-1-Genexpression zu etablieren. Durch Hybridisierungen von cDNA-Makroarrays mit radioaktiv markierten cDNA-Sonden aus APM-1-exprimierenden Tumorzelllinien oder HeLa Tet-On-Klonen wurde nach potenziellen Zielgenen des vermutlichen Transkriptionsfaktors APM-1 gesucht. Sequenzanalysen ergaben, dass funktionell inaktivierende Mutationen im proteincodierenden Bereich des APM-1-Gens nicht vorkommen. Mit Hilfe von RT-PCR-Analysen wurde gezeigt, dass APM-1 in Tumorzelllinien und Biopsieproben heterogen exprimiert wird, von drei verschiedenen Promotoren aktiv transkribiert wird und wahrscheinlich durch Methylierung transkriptionell stillgelegt werden kann. Mit transienten Transfektionsstudien und anschließenden Reportergenanalysen wurde Promotor- und Enhancer-Aktivität in den genomischen Regulationsbereichen vor den UTRs der Klassen p2 und 31 nachgewiesen. Schließlich wurden Hinweise für eine mögliche regulatorische Beziehung zwischen APM-1 und dem Tumorsuppressor-Gen p53 aufgezeigt.

Translation of abstract (English)

During the work for this doctorate thesis the mutational status, expression and regulation of the tumour suppressor candidate APM-1 was analysed. Due to the integration of DNA of the human papillomavirus type 68 (HPV68) into chromosome 18q21 in the cervical carcinoma cell line ME180 the APM-1 gene is aberrantly transcribed as part of viral-cellular hybrid mRNAs. The cellular APM-1 transcripts consist of identical protein coding moieties and different 5’ untranslated regions (UTRs) named p2, a31 and 15C. This speaks in favour of a separate initiation of transcription at three different promoters. The APM-1 gene encodes a product, which has an N-terminal BTB domain as well as four Krueppel type zinc fingers and thus may have the function of a transcription factor. For functional studies on growth inhibition the establishment of inducible APM-1 expression in HeLa cells using the Tet-On gene expression system was attempted. A search for potential target genes of the putative transcription factor APM-1 was done by hybridising radioactively labelled cDNAs taken from APM-1 expressing tumour cell lines or HeLa Tet-On cell clones to cDNA macro arrays. Sequence analyses showed that functional inactivating mutations in the protein coding region of the APM-1 gene do not occur. The results of RT-PCR analyses indicated heterogeneous expression of APM-1 in tumour cell lines and biopsy samples, active transcription from three different promoters, and the possibility for transcriptional silencing probably caused by methylation. Transient transfections followed by reporter gene analysis demonstrated both promoter and enhancer activity in the genomic regulating regions upstream of the p2 and a31 UTRs. Finally, a potential regulatory connection between APM-1 and the tumour suppressor gene p53 was revealed.
