Abstract

Die Alzheimer Demenz (AD) ist eine degenerative Erkrankung des Nervensystems, bei deren Pathologie das Amyloid Vorläuferprotein (APP) eine wichtige Rolle spielt. Für ein besseres Verständnis standen daher Fragen nach dem intrazellulären Transport und der APP Prozessierung und dessen mögliche Funktion im Mittelpunkt. Zur Analyse des APP Transports wurden Mutanten hergestellt und mit Hilfe des SFV Expressionssystems in Neuronen exprimiert. Ihre zelluläre Verteilung wurde durch immunzytochemische Methoden bestimmt. Außerdem wurden APP-GFP Fusionsproteine hergestellt um die Analyse zu erleichtern. In transfizierten Zelllinien wurde durch die Fusion jedoch die Dimerisierung von APP und die Ab Produktion gehemmt. Da die veränderten Eigenschaften von APP, die bei der Dimerisierung und Prozessierung gefunden wurden, auch den APP Transport beeinflussen könnten, wurden die Fusionsproteine in weiteren Studien nicht mehr verwendet. Die Funktion von APP bei der Signalübertragung wurde mit konstitutiven APP Dimeren untersucht. Diese wurden mit einer künstlichen Disulfidbrückenbindung hergestellt. Bei Verwendung eines GTPase Testsystems wurde kein Einfluss der Dimerisierung auf die Signalübertragung detektiert. Die konstitutive Dimerisierung bewirkte eine Erhöhung der Ab Menge. In Verbindung mit den FAD Mutationen Swedish und London wurde die Dimerisierung und die Ab Produktion noch einmal verstärkt. Die Ergebnisse dieser Arbeit lassen vermuten, dass sowohl die N-Glykosylierung als auch die Ab Domäne eine Funktion für die APP Dimerisierung haben. Diese Signale könnten den Transport und die Prozessierung von APP derart beeinflussen, dass es mit zunehmender Dimerisierung zu einer verstärkten Ab Produktion kommt. Das könnte der relevante Faktor bei der Pathogenese der presenilen AD darstellen.

Translation of abstract (English)

Alzheimer Dementia (AD) is a degenerative disease of the nervous system. The Amyloid Precursor Protein (APP) plays an important role in its pathology. This dissertation focuses on the intracellular transport and processing of APP, and its potential function in signal transduction. To analyse the transport of APP, mutants expressed in hippocampal neurons using the Semliki Forest virus system. The distribution of the mutants was determined by immunocytochemical methods. Due to experimental difficulties, no unambiguous results were obtained, so alternative methods were tested. To facilitate the analysis in the context of transport studies APP-GFP fusion proteins were produced. Experiments in stably transfected cell lines showed inhibition of both dimerisation of APP and Ab production due to the fusion with the GFP sequence tag. Altered behaviour of APP concerning its dimerisation and processing could influence APP transport, so these fusion proteins were not used in further transport studies. The potential function of APP in signal transduction via trimeric G proteins was investigated by means of constitutive APP dimers. These were generated employing artificial disulfide bonds. In a GTPase test system dimerisation did not have an influence on signal transduction. However, constitutive dimerisation led to an increase in the amount of Ab. In combination with the FAD mutations Swedish and London both dimerisation and Ab production were increased again. The experimental results suggest an important role of both the N-glycosylation and the Ab domain for APP dimerisation. These, and possibly other dimerisation signals could influence APP transport and processing in such a way that Ab production raises with increasing dimerisation of APP. This characteristic could be the relevant factor for the pathogenesis of presenile AD.
