Abstract

Clofibrat dient zur Senkung von Lipiden und Cholesterin im Blut. Es ist ein Peroxisomenproliferator in Ratten und Mäusen und kann Krebs auslösen. Diese Effekte werden in Primaten nicht beobachtet. Die Wirkung von Clofibrat auf primäre Hepatocyten aus der Regenbogenforelle und die Fischzelllinie RTG-2 wurde untersucht. 1 mM Clofibrat führte zum Absterben der Zellen und die Art des Todes erinnerte an Apoptose. Eine Proliferation der Peroxisomen blieb aus. Der Zelltod wurde licht- und elektronenmikroskopisch näher untersucht. Biochemische Versuche beinhalteten den Nachweis von Caspase 3 und p53 durch Westernblotting sowie das Auftreten einer Verlagerung von Cytochrom c aus den Mitochondrien in das Cytosol. Ferner wurde die Spaltung eines Caspase 3-Substrats in clo-fibratbelasteten Hepatocyten untersucht. Ein Caspase 3-Inhibitor sollte die Veränderungen blockieren. Zum Vergleich wurden Hepatocyten mit UV-Licht bestrahlt oder mit Staurosporin belastet. Die Belastung von Hepatocyten der Regenbogenforelle mit 1 mM Clofibrat führt zu einer Kondensie-rung von Chromatin in der Peripherie des Zellkerns und zu stark veränderten Mitochondrien. Das En-domembransystem erfährt eine Umgestaltung in einzelne Membranzisternen und annulierte Lamellen. Die äußere Kernmembran ist in diesen Prozess mit einbezogen. Es kommt zu einer Stabilisierung von p53 und der Aktivierung Caspase 3-artiger Proteasen. Im Vergleich zu anderen Induktoren der Apop-tose treten gemeinsame Merkmale auf; Unterschiede bestehen vor allem beim Endomembransystem und der Mitochondrienmorphologie. Effekte durch Staurosporin lassen sich mit einem Caspase 3-Inhibitor verhindern. Die Bestrahlung mit ultraviolettem Licht führt zu einer Mischung aus Onkose und Apoptose. Der Zelltod durch Clofibrat lässt sich nicht mit einem Caspase 3-Inhibitor unterbinden. Weitere Mechanismen zur Abtötung der Zellen müssen existieren. Hierbei könnte die Bcl-2-Familie und der Apoptosis Inducing Factor eine Rolle spielen.

Translation of abstract (English)

Clofibrate is used to lower lipid and cholesterol in blood. Clofibrate is a peroxisome proliferator in rats and mice and can induce cancer in those animals. Both effects can not be found in primates. The effects of clofibrate on primary hepatocytes from rainbow trout and the fish cell line RTG-2 were studied. The chosen concentrations lead to dying cells and the type reminded on programmed cell death (apoptosis). A significant proliferation of peroxisomes could not be found. The type of cell death was studied using morphological changes on a light and electronmicroscopical level. Biochemical studies contained western blotting for caspase 3 and p53 as well as the shift of cytochrome c contents from mitochondria to the cytosol. Additionally, cleavage of a caspase 3 substrate in hepatocytes ex-posed to clofibrate was studied. A caspase 3 inhibitor should block the observed changes. To compare the results, hepatocytes were also exposed to UV light and staurosporine. Exposure of hepatocytes to 1 mM clofibrate results in the condensation of chromatin in the periphery of the nucleus and to severely altered mitochondria. The endomembrane system changed into single cisternae and annulated lamellae found in the cytosol. The outer membrane of the nucleus was also part of this membrane network. P53 is stabilized and caspase 3 like proteases are activated. Compared to other inducers of apoptosis, common properties can be found but there are also differences espe-cially concerning the changes of the endomembrane system and the mitochondrial morphology. Ef-fects caused by staurosporine exposure can be prevented by the use of a caspase 3 inhibitor. Exposure to ultraviolet light lead to a mixtures of oncosis and apoptosis. Cell death due to clofibrate exposure could not be prevented by a caspase 3 inhibitor. Therefore, additional mechanisms of cell killing must exist. The bcl-2 family and the apoptosis inducing factor may be involved in killing cells if caspases are inhibited.
